Project description:The goal of this experiment was to examine the innate immune response to helminth infection in the lung. Hookworms (like many other helminths) use an obligate migration pathway through the lung. Their infection has been characterized in the gut in detail, but early immune responses in the lung have not been fully characterized. Experiment Overall Design: SCID mice were used to examine the innate immune response. SCID mice have no functional B or T cells but a fully functional innate immune system. SCID and WT mice were infected with 500 L3 stage infectious Nippostrongylus brasiliensis SC and their lungs were removed at days 2,3,4,8 and 12 post infection.

Project description:Purpose: Helminth infection and dietary intake can affect the intestinal microbiota, as well as the immune system. Methods: Here we analyzed the relationship between fecal microbiota and blood profiles of indigenous Malaysians, referred to locally as Orang Asli, in comparison to urban participants from the capital city of Malaysia, Kuala Lumpur. Results: We found that helminth infections had a larger effect on gut microbial composition than did dietary intake or blood profiles. Trichuris trichiura infection intensity also had the strongest association with blood transcriptional profiles. By characterizing paired longitudinal samples collected before and after deworming treatment, we determined that changes in serum zinc and iron levels among the Orang Asli were driven by changes in helminth infection status, independent of dietary metal intake. Serum zinc and iron levels were associated with changes in the abundance of several microbial taxa. Conclusions: There is considerable interplay between helminths, micronutrients and the microbiota on the regulation of immune responses in humans.

Project description:The intestinal epithelium is our first line of defense against infections of the gut and the plasticity in cellular differentiation of the intestinal epithelium is an important part of this response. Here we sequenced the small intestinal epithelium from mice infected with Nippostrongylus brasiliensis to determine how the intestinal epithelium adapts in the context of an infection. By comparing these data to small intestinal organoids treated with cytokines (see related accessions) we determine that the intestinal epithelial response to N. brasiliensis infection correspond to a type II infection driven by IL-13.

Project description:The discovery of naïve T cell transcriptional heterogeneity in specific-pathogen-free (SPF) mice represents a paradigm shift. While the ablation of Vista can alter the proportion of naïve CD4+ T cells with unique transcriptional signatures and the ‘readiness’ of the mice to break tolerance, the effect of non-heritable environmental exposures to commensals or infections remains entirely unknown. This is especially significant since individual variance in immune response, rather than being genetically inherited, is mostly attributed to environmental factors. Here we demonstrate that transcriptional heterogeneity characterizes even the germ-free (GF) mouse naïve CD4+ T cells. Naïve CD4+ T cell transcriptional subsets and their proportions were identical between GF, SPF and conventionalized GF mice, thus commensal-independent. Notwithstanding, Nippostrongylus brasiliensis (N.b.) infection altered the proportions of these transcriptional subsets, and the appearance of cells with a unique transcriptional signature. These naïve T cell-intrinsic changes accounted for the decreased immunization response of mice to an unrelated antigen. The compositional and functional changes in the naïve CD4+ T cell pool were dependent-variables of helminth infection, regressing with N.b. clearance. Therefore, naïve T cells are not uniformly uncommitted substrates for antigen-dependent activation but actively integrate environmental cues to permute the magnitude of the immune response.

Project description:Goal: To examine the effects of human resistin during helminth infection. Methodology: To examine the function of human resistin (hResistin), we utilized transgenic mice expressing the human resistin gene along with its entire regulatory region (hRetnTg+). Following infection with the helminth Nippostrongylus brasiliensis, whole lung RNA was sequenced in hRetnTg+ mice, control hRetnTg- and naïve mice. Conclusion: In hRetnTg+ mice, many genes involved in inflammation and the immune system, specifically toll-like receptor signaling and chemokines, are significantly upregulated, suggesting that human resistin promotes TLR signaling and inflammation during helminth infection. Project description:We were interested in investigating the heterogeneity of CD4+ Th2 cells during infection response. In order to do so we injected Nippostrongylus brasiliensis (Nb) in recipient mice as this is known to elicit a Type 2 response. Mice were subcutaneously injected with Nb larvae and single CD4+ cells were isolated from mediastinal lymph nodes, mesenteric lymph nodes, and lungs 5 days after infection.

Project description:Tissue lymphatics are crucial for fluid homeostasis and immunosurveillance. We asked whether the lymphatics are adapted to the organ in which they reside, such as along the gut. Duodenal lymphatic capillaries (lacteals) displayed the mot discontinuous tight junction composition versus lacteals at other sites, resulting in a dependence on duodenal lacteals for rapid dietary lipid uptake. Duodenal helminths abrogated these features. RNAseq of LECs and the mucosa along the intestine revealed that LEC transcriptomes matched the functional profile of the local mucosa. Helminth infection reverses key duodenal signatures like lipid metabolism and immune activation, instead triggering lymphangiogenic and mucosal antimicrobial responses. We identified a putative VEGFR2/3 signaling gradient that may explain differences in tight junction along the small intestine at homeostasis. Inflammation-induced lymphangiogenesis and decreased fat exposure likely underlie duodenal lymphatic impermeability upon helminth infection, while microbial depletion acted additively on lymphatic restructuring. Our study provides molecular insights into lymphatic function along the intestine and stages lymphatic permeability as subject to dynamic regulation by environmental queues.

Project description:Background: Parasitic helminths influence the composition of the gut microbiome. However, the microbiomes of individuals living in helminth-endemic regions are understudied. The Orang Asli, an indigenous population in Malaysia with high burdens of the helminth Trichuris trichiura, display microbiotas enriched in Clostridiales, an order of spore-forming obligate anaerobes with immunogenic properties. We previously isolated novel Clostridiales that were enriched in these individuals and found that a subset promoted the Trichuris life cycle. In this study, we aimed to further characterize the functional properties of these bacteria.Results: Clostridiales isolates were profiled for their ability to perform 57 enzymatic reactions and produce short chain fatty acids (SCFAs) and hydrogen sulfide, revealing that these bacteria were capable of a range of activities associated with metabolism and host response. Consistent with this finding, monocolonization of mice with individual isolates identified bacteria that were potent inducers of regulatory T cell (Treg) differentiation in the colon. Comparisons between variables revealed by these studies identified enzymatic properties correlated with Treg induction and Trichuris egg hatching.

Project description:Approximately one-third of the world is infected with parasitic helminths, and there are no available vaccines. Helminths induce robust type 2 immune responses and host CD4+ T cells are required for clearance, but how helminth-specific CD4+ T cells are generated and whether they adopt prototypical features of effector Th2 cells is entirely unknown. We generated a novel transgenic line of Strongyloides ratti to track and study helminth-specific T cells. This transgenic line, termed Hulk, stably expresses immunodominant T cell epitope 2W1S as a fusion protein with FLAG and green fluorescent protein (GFP). We consistently observe expansion of CD44hiCD11ahi 2W1S-specific CD4+ T cells in the lungs of Hulk-infected C57BL/6 mice. Surprisingly, these cells do not exhibit a Th2 or regulatory T cell (Treg) phenotype, and their expansion is minimal compared to expansion following 2W1S-peptide immunization. In order to more thoroughly interrogate the phenotype and function of 2W1S-specific CD4+ T cells during Hulk infection, we performed mRNA sequencing on 2W1S-specific CD4+ T cells from both Hulk-infected and 2W-Salmonella-infected mice. Interestingly, whereas 2W1S-specific CD4+ T cells that expand during 2W-Salmonella infection express canonical pro-inflammatory genes, 2W1S-specific CD4+ T cells from Hulk-infected mice preferentially upregulate genes associated with immune suppression and wound healing. Altogether, these data suggest that helminth antigen specific CD4+ T cells may comprise an unconventional subset that mediates immunosuppression and tissue repair following helminth infection and not necessarily type 2 cytokine responses.
