ABSTRACT: Gene expression was measured in isolated wheat embryos at physiological maturity. The gene expression measurements were done across a doubled haploid white winter wheat population between the variety Cayuga and the variety Caledonia (CxC). Data can be used for mapping gene expression QTL (eQTL). These data are from the second (Ketola 2005) of two field environments. Different target labeling was used so the environments are contained in two separate series.

Project description:Gene expression was measured in isolated wheat embryos at physiological maturity. The gene expression measurements were done across a doubled haploid white winter wheat population between the variety Cayuga and the variety Caledonia (CxC). Data can be used for mapping gene expression QTL (eQTL). These data are from the second (Ketola 2005) of two field environments. Different target labeling was used so the environments are contained in two separate series. The design was a two-channel dye-swap reference design with bulked total RNA from pre-anthesis wheat spikes serving as the reference. Normalization was done using the TIGR MIDAS suite using blockwise LOWESS smoothing and variance normalization. Between slide variance normalization was also done for all slides so direct comparisons could be made. Dye-swap was submitted unmerged to facilitate flexibility in downstream processing.

Winter wheat (Triticum aestivum L.), is an important crop plant all over the world. Long term cold exposure (vernalization) is necessary for winter wheat transition from vegetative growth to reproductive growth, yet the involvement of miRNAs in these stages remains unknown. Therefore, we performed next generation sequencing of small RNAs profiles in crown tissues at three-leaf stage, winter dormancy stage, spring greenup stage and jointing stage.

Project description:The effect of light during the development of freezing tolerance was studied in winter wheat (Triticum aestivum L. var. Mv Emese) and spring wheat variety Nadro. Ten-day-old plants were cold hardened at 5°C for 12 days either under normal (250 mmol m-2 s-1) or low light (20 mmol m-2 s-1) conditions. Samples of Emese (E) and Nadro (N) plants grown at 18°C under normal (NL) and low (LL) light fluences were compared to each other in a simple loop design and E-NL vs. E-LL; N-NL vs. N-LL; E-NL vs. NLL and E-LL vs. N-LL comparisons were made.

Dual RNA-sequencing of Fusarium Head Blight resistance in winter wheat

Project description:In this study, we used dual RNA-sequencing to profile FHB-resistant AC Emerson, FHB-moderately AC Morley, and FHB-susceptible CDC Falcon winter wheat cultivars prior to and in response to Fusarium graminearum at 7 days post inoculation. Differential expression analyses revealed distinct defense responses between resistant and susceptible wheat cultivars including increased mechanical defense through lignin biosynthesis and increased deoxynivalenol (DON) detoxification through UDP-glycosyltransferase activity in resistant cultivars. Further, differential expression analysis in F. graminearum challenging these distinct cultivars revealed changes genes involved in trichothecene mycotoxin biosynthesis.
