Project description:The endometrium plays a central role among the reproductive tissues in the context of early embryo-maternal communication and pregnancy. This study investigated transcriptome profiles of endometrium samples from Day 18 pregnant vs. non-pregnant heifers to get insight into the molecular mechanisms involved in conditioning the endometrium for embryo attachment and implantation. Using a combination of subtracted cDNA libraries and cDNA array hybridization 109 mRNAs with at least twofold higher abundance in endometrium of pregnant animals and 70 mRNAs with higher levels in the control group were identified. Among the mRNAs with higher abundance in pregnant animals at least 41 are already described as induced by interferons. In addition, transcript levels of many new candidate genes involved in regulation of transcription, cell adhesion, modulation of the maternal immune system, and endometrial remodeling were found as increased. The different expression level was confirmed with quantitative real-time PCR for nine genes. Localization of mRNA expression in the endometrium was shown by in situ hybridization for AGRN, LGALS3BP, LGALS9, USP18, PARP12, and BST2. A comparison with similar studies in humans and mice revealed species-specific and common molecular markers of uterine receptivity. Messenger RNA profiling of bovine endometrium during the estrous cycle using a custom-made cDNA array

Project description:The endometrium provides optimal conditions for the transport of sperm to the oviduct, to the site of fertilization, and later on for the reception of the embryo. To study these changes on the level of gene expression, a messenger RNA expression profiling of endometrium tissue samples collected from 19 cyclic heifers at five stages of the estrous cycle (days 0, 3.5, 12, 18, 20) was performed. RNA was extracted from these tissue samples and analyzed with a custom-made bovine oviduct and endometrium (BOE) cDNA array. The cDNAs present on the array were derived from several previously conducted differential gene expression studies of bovine endometrium between different stages of the estrous cycle, during early pregnancy, and from studies of bovine oviduct epithelial cells. In all of these studies cDNAs of differentially expressed genes were identified using a combination of subtracted cDNA libraries and cDNA array hybridization. 1,440 cDNA fragments are located on the array. Twenty radioactively labeled cDNA samples (n=4 for each cycle stage) were hybridized with BOE arrays. Raw data were normalized using the BioConductor package vsn. Transcriptome Analysis of Human Supragingival Dental Plaque Microbiome from 19 Twin Pairs with and without Dental Caries

Project description:The study aims to assess gene expression in plaque samples collected from twin pairs that are both concordant and discordant with respect to dental Caries diagnosis. File Naming Conventions are as follows: Patient ID : 4 digit identifier Diagnosis : Caries Negative(CN) or Caries Positive(CP) Type of Twin: Monozygotic(MZ)or Dizygotic(DZ) Pair to xxxx: 4 digit twin identifier maps to the Patient ID E.g: 2126_CP_MZ_PairTo_2125_fastqc - 2126 is a caries positive patient and pairs to monozygotic twin pair 2125. Plaque samples from twin pairs that are both concordant and discordant with respect to dental Caries diagnosis are enriched for bacterial messenger RNA to study the gene expression differences in the samples. RNA was extracted from RNAprotect (Qiagen, In c.) treated dental plaque scrapings from 38 patients. Amplified cDNA was created and rRNA sequence was removed by subtractive hybridization. Individual patient samples were run on a single lane of an Illumina Genome Analyzer.

Project description:Genome wide DNA methylation profiling with the HumanMethylation450 BeadChip (450k) of peripheral blood samples of 46 adult female monozygotic twin-pairs obtained at the same time point. Samples included 41 healthy females (not diagnosed with cancer to date) paired with their co-twin diagnosed with cancer within a 5 year window around the time of sampling as well as 5 extra pairs with a co-twin diagnosed with cancer within 11 to 5 years prior to blood sampling.

Differential Gene Expression in Uterine Endometrium during Implantation in Pigs

Project description:This study provide an opportunity to elucidate the genetic control of fetal implantation and improve our understanding of fetal implantation and gestation maintenance, thus make further improvement for litter size of pigs. Nine pregnant sows were slaughtered by electrical on day 13, day 18 and day 24 after insemination (the pregnant group, three sows every period). The non-pregnant sows were slaughtered on day 13 after inseminated (n=3).In pregnant sows, samples of the endometrium attachment sites and inter-sites were taken. Samples from the endometrium of the non-pregnant sows were taken from comparable locations.

Transcription profiling of porcine endometrium from chronological stages of psuedopregnancy and estrogen disrupted psuedopregnancy

Project description:Placentation of the conceptus to the surface epithelium is governed through a tightly regulated temporal and spatial window. Premature exogenous steriod exposure causes a shift in the maternal tissue's receptivity and prevents proper placentation. We used Affymetrix microarrays to detail the aberrant changes in uterine gene expression during this window, while removing all conceptus contribution to this dysfunction. Experiment Overall Design: Porcine endometrium was selected at chronological stages of psuedopregnancy and psuedopregnancy post endocrine disrupted early for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain homogeneous populations of normal endometrium at days 13 and 15 of the pseudo pregnant, and endometrium at days 13 and 15 of the endocrine disrupted pseudo pregnant in order to identify the temporal resolution of expression profiles. In brief, days 13 through 15 accounts for the window of attachment during pregnancy in the pig, so we created pseudo pregnant endometrium and obtained samples from days 13 (PIG13UN_TX)and 15 (PIG15UN_TX) of pseudo pregnancy. In the same, we created pseudo pregnant endometrium that was subjected to endocrine disruption of the pseudo pregnancy and collected samples on days 13 (PIG13_TX)and 15 (PIG15_TX) of pseudo pregnancy.
