Project description:The purpose of this experiment is to elucidate temporal activities and biological processes that can be inferred in response to a 200cGy challenging dose with or without a 10 cGy priming dose in embryonic human fibroblasts The experiment is designed around the human diploid embryonic lung fibroblast cell line WI38 (TP53 proficient), which was grown as a monolayer (2D) under physiological cell culture conditions (3% O2; 10% CO2). Cells are exposed to 2Gy (e.g., the challenge dose) of ionizing radiation (160 kV X-rays) with or without a priming dose of 10cGy (e.g., the adaptive dose) 4 hours prior to the challenge dose. Three biological replicates for each treatment group (e.g., with and without the priming dose) were collected 1, 2, 4 and 8 hours after the challenge dose. The time course was selected on the basis of our prior research on early response to ionizing radiation [1,2]. Purified total cellular RNA was extracted using an RNeasy Mini Kit (Qiagen) and quantified for Affymetrix microarray analysis using Human Gene 1.0 ST Array. Robust multi-array analysis (RMA) was performed to normalize data collected from the different samples. Samples were examined for quality control using the NUSE protocol. Project description:Radioadaptive response (RAR) in mammalian cells refers to the phenomenon where a low-dose ionizing irradiation alters the gene expression profiles, and protects the cells from the detrimental effects of a subsequent high dose exposure. We focused on mRNA microarray studies, and aimed to characterize the transcriptome for RAR in AG 1522 human skin fibroblasts and to examine the functional regulatory networks at the genetic level. AG 1522 cells were exposed at a specific time point to a challenging dose of 2 Gy in the RAR group, or a priming dose of 5 cGy in the low-dose group. We aimed to have a comprehensive investigation on the RAR induced in the AG 1522 human fibroblasts first exposed to 5 cGy (priming dose) and then followed by 2 Gy (challenge dose) of X-ray through comparisons to those cells which had only received a single 2 Gy dose.

Multi-omics profiling reveals ethylene signalling as a key pathway underlying both genetic and epigenetic responses to low-dose ionizing radiation in Arabidopsis

Project description:There is increasing interest in the effects of low-dose ionizing radiation (IR) on plants as might occur during spaceflight, or as a consequence of human activities, such as nuclear power generation, that may result in the release of radioactive materials into the environment. High doses of IR have long been used for the induction of mutations in plants with the goal of generating desirable traits for agribusiness. However, less is known about the responses of plants to acute low doses of IR exposure. Here, we take a multi-omics approach to characterize the response to low dose IR in A. thaliana. We adapt the Methyltransferase Accessibility Protocol for individual templates (MAPit) technique for use in plants allowing us to assay the epigenetic response to acute low-dose IR (10 cGy and 100 cGy) 72 hr after exposure, and, in parallel, use RNA sequencing to profile the transcription response at 1, 3, 24 and 72 hr after exposure. We observe that IR exposures as low as 10 cGy elicit robust genetic responses in Arabidopsis thaliana detectable as early as 1 hr after exposure. Examination of these responses revealed dose-dependent changes in gene expression, chromatin accessibility and DNA methylation that implicate the ethylene signalling pathway and response to abiotic stress as underlying the transcriptional and epigenetic changes associated with IR. These changes are observable up to 72 hr after exposure suggesting that they are maintained well after the initial acute exposure. Our findings indicate that A. thaliana executes a coordinated, multi-modal response to low-dose IR through induction and regulation of the ethylene response pathway.
