Project description:Dietary restriction (also known as caloric/calorie restriction; CR) extends the lifespan of species from all three eukaryotic kingdoms. The restriction of the diet interferes directly with the aging process by triggering a tightly controlled genetic program where specific sets of genes are either upregulated downreguled. We used microarray-technology to detail the global program of gene expression underlying the anti-aging effect of dietary restriction and identified distinct classes of up- and down-regulated genes. In order to apply dietary restriction in budding yeast we cultured cells on a reduced glucose medium (0.5% vs. 2.0%), which is known as moderate DR regimen. We then compared mRNA expression of yeast cells cultured under dietary restricted (0.5% glucose) and ad libitum (2.0% glucose) conditions.

Project description:Dietary restriction has shown benefits in physiological, metabolic, and molecular signatures associated with aging. In mice, periodic dietary restriction, that accompanies cyclical periods of reduced calories, mitigates aging phenotypes, yet the effects of this periodic restriction in higher order, genetically heterogenous mammals, have not been investigated. Reducing caloric intake in middle-aged Rhesus Macaques for four days followed by ad libitum eating for ten days, repeated for six cycles, led to significant changes in body composition, metabolome, and microbiome of the restricted monkeys compared to their ad libitum fed (age and sex matched) controls. These changes were also associated with stabilized blood parameters. The changes in metabolome, microbiome, and body compositions were additive, with consistent but increasingly pronounced changes in later cycles, suggesting sustaining benefits in non-human primates. These results suggest this type of cyclical dietary restriction, that is easy to adhere to, will also have translational benefits in humans while enhancing aging-associated parameters.

Project description:The SIRT1 deacetylase is one of the best-studied potential mediators of some of the anti-aging effects of calorie restriction (CR); but its role in CR-dependent lifespan extension has not been demonstrated. We previously found that mice lacking both copies of SIRT1 displayed a shorter median lifespan than wild type mice on an ad libitum diet. Here we report that median lifespan extension in CR heterozygote SIRT1+/- mice was identical (51%) to that observed in wild type mice but SIRT1+/- mice displayed a higher frequency of some certain pathologies. Although larger studies in different genetic backgrounds are necessary , these results provide strong initial evidence for the requirement of SIRT1 for the anti-aging effects of CR, but suggest that its high expression is not required for CR-induced lifespan extension. Non-growing but metabolically active cultures of S. cerevisiae are of major interest for application in industry and as model systems for aging in higher eukaryotes. Using retentostat cultivations, almost non-growing but metabolic active cultures can be obtained resulting from the severe caloric restriction, yet not starvation, yeast experiences. Rim15 plays an important role in several nutrient sensing pathways and is involved in activating stress response and glycogen accumulation upon nutrient shortage. To investigate the role of Rim15 in the extreme robustness and glycogen accumulation of anaerobic retentostat cultures, a rim15 deletion strain is compared with its parental strain under anaerobic calorie restriction on both physiology and transcriptome. Rim15 is described as essential for G0 entry and glycogen accumulation in yeast during diauxic shift and stationary phase. Anaerobic retentostat cultures display many stationary phase characteristics, including increased expression levels of Rim15 target genes, suggesting an important role for Rim15 under these conditions. Comparing a rim15 deletion strain and its parental strain revealed both on transcriptome level and physiology indeed a major role in the acquired robustness, glycogen accumulation, but also maintenance of viability and cell cycle arrest. The severe caloric restriction, but not starving, conditions applied together with a thorough physiological and transcriptome analysis of the cultures shows that Rim15 is essential in fine-tuning cell cycle progression with glucose availability under extreme growth-limiting conditions. To investigate the impact of rim15 deletion under severe calorie restricted conditions, transcriptome of a S. cerevisiae rim15 deletion mutant was monitored during anaerobic retentostat cultivation. Five time points were used, one in the starting point of the retentostat (T0 = chemostat) and four during the course of the retentostat (time points 2, 9, 16 and 20 days). Culture triplicates were performed for T0 in chemostat, while two independent cultures were run for retentostat.

Project description:Aims/hypothesis: Dietary restriction (DR) reduces adiposity and improves metabolism in patients with one or more symptoms of the metabolic syndrome. Nonetheless, it remains elusive whether the benefits of DR in humans are mediated by calorie or nutrient restriction. This study was conducted to identify whether isocaloric dietary protein restriction is sufficient to confer the beneficial effects of dietary restriction in patients with metabolic syndrome. Methods: We performed a prospective, randomized controlled dietary intervention under constant nutritional and medical supervision. A total of 21 individuals diagnosed with the metabolic syndrome was randomly assigned for caloric restriction (CR; n = 11, mean age 49 ± 8.5 years, female 63%; diet of 5,941 ± 686 KJ per day) or isocaloric dietary protein restriction (PR; n = 10, mean age 51.6 ± 8.9 years, female 50%; diet of 8,409 ± 2,360 KJ per day) and followed for 27 days. Results: Like CR, PR promoted weight loss (-6.6%, P= 0.0041) due to reduction in adiposity (-9.9%, P= 0.0007), associated with reductions in blood glucose (-52.7%, P= 0.0002), lipid levels (cholesterol, -35.4%, P= 0.0010; triglycerides, -39.5% P= 0.0022) and blood pressure (systolic, -37.7 P< 0.0001; diastolic, -73.2% P< 0.0001). PR resulted in enrichment of metabolic pathways related to the immune system such as B cell proliferation, lymphocyte proliferation and leukocyte proliferation in subcutaneous adipose tissue. Hence, a reduction in calorie intake or changes in the gut microbiome are not necessary to confer the metabolic benefits of DR. Instead, a reduction in protein intake with a mild increase in carbohydrate intake to maintain the isocaloric balance of the diet is sufficient to improve metabolic control. Conclusions/interpretation: Protein restriction is sufficient to confer almost the same clinical outcomes as calorie restriction without the need for a reduction in calorie intake. The isocaloric characteristic of the PR intervention makes this approach a more attractive and less drastic dietary strategy in clinical settings and has greater potential to be used as adjuvant therapy for people with the metabolic syndrome.
