ENU derived mouse line AEA001: A model for osteoporosis carrying a mutation in Ednra

ABSTRACT: Transcriptome analysis of white adipose tissue and bone (femur of the hind limbs) of the AEA001 mutant mouse. ENU generated mouse lines for osteoporosis. The mutation in the Ednra gene causes big ears and a flat short head. Due to a decreased body mass and a trend towards decreased glucose level white adipose tissue was analysed.

Next Generation Sequencing (NGS) for quantitative transcriptomic analysis of colorectal tumor cell lines and their ENU mutants

Project description:Purpose: To understand differences of gene expression profiles between colorectal cell lines and their ENU mutants. Methods: RNA profiles were generated by deep sequencing using Illumina Next-seq. Using an optimized data analysis workflow, we mapped about 30 million sequence reads per sample to the mouse genome (GRCm38 release-95). Conclusions: Our study represents the detailed differential transcriptomic analysis using ENU mutants of colorectal tumors, generated by RNA-seq technology. The optimized data analysis workflows reported here should provide a framework for comparative investigations of expression profiles. Our results show that NGS offers a comprehensive and more accurate quantitative and qualitative evaluation of mRNA content within a cell. We conclude that RNA-seq based transcriptome characterization would expedite genetic network analyses and permit the dissection of complex biologic functions.

Effect of ablation of Sfrp5 on gene expression in gonadal white adipose tissue

Project description:We identified secreted frizzled-related protein-5 (Sfrp5) as a transcript that is upregulated during adipocyte differentiation and that is increased in white adipose tissue (WAT) of obese mice, compared to lean mice. To investigate the function of sFRP5 in adipose tissue biology, we studied sFRP5Q27stop mice, in which ENU mutagenesis was used to create a premature stop codon at Gln27, thereby creating a likely null allele. Male wild-type or Sfrp5 KO (Q27Stop) mice were fed a high-fat diet from the ages of four weeks to twelve weeks. At twelve weeks of age, mice were euthanized. Total RNA was then isolated from gonadal WAT and RNA was analyzed by Affymetrix microarrays. Seven wild-type and eight Sfrp5 KO mice were used.
