Screening for potential markers of metastases between low versus highly metastatic derived cell lines of same parental origin

ABSTRACT: The objective of this study is to identify metastatic candidates via initial screening by gene expression profiling between subcloned cell lines of low (C5 and FG) and high (C5LM2 and L3.6) metastatic potential from their parental primary human pancreatic adenocarcinoma cell lines (Panc1 and COLO357). Agilent arrays were utilized for this experiment and there are 3 replicates for each cell line.

Project description:Two pancreatic cancer cell lines with different metastatic and growth potential were compared under hypoxic conditions and under normal atmospheric oxygen pressure. The FG cell lines shows very few metastases and slow growth in mouse xenograft models. L3.6pl, derived from FG by cycles re-implantation of metastatic cells obtained after orthotopic tumor growth in nude mice, shows high motility, aggressive growth and very high metastatic potential By comparison of the two cell lines under different oxygen concentration we tried to simulate in vivo conditions of tumors at different growth stages. Project description:The paired low-metastatic 95C and high-metastatic 95D cell lines were subcloned from a low differentiated human large cell lung carcinomacell line PLA-801. The cells were well authenticated and published by several research groups. The cells were kindly provided by Professor Ying-Lin Lu, Department of Pathobiology, Institute of Basic Medical Sciences, Academy of Military Medical Sciences, Beijing, China on Dec. 5M-oM-<M-^L2009. As described The 95C and 95D cells were cultured in RPMI 1640 (Invitrogen, USA) with 100 units/mL penicillin, 100 M-NM-<g/mL streptomycin and 10% calf bovine serum, and grown at 37M-BM-0 C in atmosphere with 5% CO2. miRNAs differential expression between 95C and 95D was measured using a miR human_01_H10.1_080277 miRNA array (LC Sciences Houston, USA). Paired subcloned human large cell lung carcinomacell line PLA-801 of low-metastatic 95C and high-metastatic 95D cell lines. Biological replicates:1. Five replicate per array.

Project description:Spontaneous cell fusion of MDA-MB-231 bone-metastatic subline Bm (i.e., SCP2) and lung metastatic subline Lm (i.e., LM2) gave rise to hybrid lines BLm-FACS or BLm-DRUG, as well as its single clones (#8, #12, #18). The hybrids acquired the metastasis tropisms from both parental cells. Expression profiles of the parental cells, the hybrids and several previously characterized MDA-MB-231 metastatic derivatives were compared. Hierarchical clustering showed the hybrids assimilated the organ-specific metastasis gene signatures from both parental cells. Experiment Overall Design: Twenty-six cell lines were analyzed, including the parental line MDA-MB-231; cell fusion partner lines Bm and Lm; self-fused lines BBm and LLm; hetero-fused lines BLm-FACS, BLM-DRUG and clones BLm-DRUG-8, -12 and -18; strongly bone-metastatic lines 1833, SCP14, SCP20, SCP25 and SCP46; strongly lung-metastatic lines 3481, 4142, 4173, 4175 and 4180; and weakly metastatic lines SCP3, SCP4, SCP6, SCP28, SCP32 and SCP43. Single sample for each line.
