ABSTRACT: Cumulus-oocyte complexes were isolated a seperate time-points to generate temporal complexes. Targets from two biological replicates at each time point (0h, 8h, 16h post-hCG treatment) were generated and the expression profiles were determined using Affymetrix GeneChip Mouse Genome 430 2.0 Arrays. Comparisons between the sample groups allow the identification of genes with temporal expression patterns. Keywords: time course

Project description:We sought to identify genes that are regulated by the ovulatory signals in mouse cumulus cells. We compared the transcriptomes between cumulus cells colected from mice before (hCG-0h) and after 6 h (hCG-6h) of hCG injection. 3 individual cumulus cell samples of hCG-0h and hCG-6h were collected and subjected to microarray study. Copmarison was done between hCG-0h and hCG-6h groups.

Expression data from progesterone receptor knockout versus heterozygous mouse ovaries: cumulus oocyte complexes

Project description:Ovulation requires sequential molecular events and structural remodeling in the ovarian follicle for the successful release of a mature oocyte capable of being fertilised. Critical to this process is progesterone receptor (PGR), a transcription factor highly yet transiently expressed in granulosa cells of preovulatory follicles. Progesterone receptor knockout (PRKO) mice are anovulatory, with a specific and complete defect in follicle rupture. Therefore, this model was used to examine the critical molecular and biochemical mechanisms necessary for successful ovulation. Although PGR is not expressed in the cumulus cells or oocyte of the preovulatory cumulus oocyte complex (COC), it is well known that the COC responds to the cascade of gene expression changes that occurs in preovulatory granulosa cells. We used microarrays to identify putative ‘ovulation’ genes in preovulatory COCs at a time when PGR expression is maximal in granulosa cells (eCG + 8h hCG) and the preovulatory COC and follicle are undergoing the final changes necessary for successful ovulation.

Expression data from cumulus cells from C57Bl/6 and Ptger2 (EP2) KO mice

Project description:To understand the role of prostaglandin (PG) receptor EP2 (Ptger2) signaling in ovulation and fertilization, we investigated time-dependent expression profiles in wild-type (WT) and Ptger2-/- cumuli before and after ovulation by using microarrays. We used Affymetrix U74A ver2 microarrays to investigate gonadotropin-induced gene expression profiles in WT and EP2KO cumulus cells before and after ovulation. Immature mice were primed with pregnant mare serum gonadotropin (PMSG) as an FSH-like stimulation. The cumulus-oocyte complexes (COCs) were isolated from mice 48 h after PMSG injection (H0) and from PMSG-primed mice exposed to human chorionic gonadotropin (hCG) as an LH-like stimulation for 3, 9 or 14 h (H3, H9 or H14, respectively).

Rhesus monkey cumulus cells revert to a mural granulosa cell state following an ovulatory stimulus

Project description:Follicular somatic cells (mural granulosa cells and cumulus cells) and the oocyte communicate through paracrine interactions and through direct gap junctions between oocyte and cumulus cells. Considering that mural and cumulus cells arise through a common developmental pathway and that their differentiation is essential to reproductive success, understanding how these cells differ is a key aspect to understanding their critical functions. Changes in global gene expression before and after an ovulatory stimulus were compared between cumulus and mural granulosa cells to test the hypothesis that mural and cumulus cells are highly differentiated at the time of an ovulatory stimulus and further differentiate during the periovulatory interval. The transcriptomes of the two cell types were markedly different (>1500 genes) before an ovulatory hCG bolus but converged after ovulation to become completely overlapping. The predominant transition was for the cumulus cells to become more like mural cells after hCG. This indicates that the differentiated phenotype of the cumulus cell is not stable and irreversibly established but may rather be an ongoing physiological response to the oocyte.

Multi-omics reveal the metabolic patterns in mouse cumulus cells during oocyte maturation

Project description:Bi-directional communication between cumulus cells and the surrounded oocytes is important for the development and functions of both compartments. However, the metabolic framework in cumulus cells has not been systematically described. In the present study, cumulus cells from cumulus-oocyte complexes (COCs) at three key time points were isolated, and the temporal metabolomic and proteomic profiling were performed. Integrated multi-omics analysis reveals the global metabolic patterns in cumulus cells during mouse oocyte maturation. In particular, we found the active hyaluronic acid metabolism, steroid hormone synthesis, and prostaglandin E2 (PGE2) production in cumulus cells. Meanwhile, accompanying the oocyte maturation, a progressive increase in nucleotide and amino acid metabolism were was detected in the surrounding cumulus cells. In addition, the comparison of metabolite changes between oocyte and cumulus cells implicates the metabolic coordination. In sum, the data serve as a valuable resource for probing follicular metabolism during follicular development, and provide the potential biomarkers for improving and predicting oocyte quality.
