Project description:We report that an intracellular NLR immune receptor from monocotyledonous barley is fully functional in dicotyledonous Arabidopsis thaliana. In contrast to common belief in plant NLR biology, this implies ~150 million years of evolutionary conservation of the underlying immune mechanism. This also suggests a conserved translocation mechanism for pathogen effectors in flowering plants. The deduced connectivity of the NLR to bifurcated signaling pathways likely confers increased robustness against pathogen interception, which could have contributed to the evolutionary preservation of the immune mechanism. RNA-seq experiments in Arabidopsis detected sustained expression of a single major MLA1-dependent gene cluster. 3 biological replicates per condition. The Arabidopsis plants without (pps) or with (B12) the MLA1-HA construct in a pen2 pad4 sag101 background were challenged with either the Bgh isolate K1 expressing the cognate AVRA1 effector for MLA1 or the Bgh isolate A6 expressing other AVRA effectors. Project description:Expression analysis was performed on total RNA from the Q21861 and SM89010 barley lines, and 75 derived doubled haploid progeny, with CI 16137 included as an internal Mla1 allele control. Samples were blocked by time-point and completely randomized within each block. For each sample, seven day old seedlings were inoculated with Blumeria graminis f. sp. hordei (Bgh) isolate 5874 (AVRa1, AVRa6, AVRa12), and first leaves were collected at 16 and 32 hours after inoculation (HAI). ****[PLEXdb(http://www.plexdb.org) has submitted this series at GEO on behalf of the original contributor, Roger Wise. Identification of avirulence genes in the barley powdery mildew fungus (Bgh) by RNA-sequencing and transcriptome-wide association analysis on a set of Bgh isolates

Project description:Disease resistance (R) genes encoding intracellular nucleotide-binding domain and Leucine-rich repeat proteins (NLRs) are key components of the plant innate immune system and typically detect the presence of isolate-specific avirulence (AVR) effectors from pathogens. NLRs define the fastest evolving gene family of flowering plants and are often arranged in gene clusters containing multiple paralogs, contributing to extensive copy number and allele-specific NLR variation within a host species. Barley mildew resistance gene locus A (MLA) represents one of only few R genes that have been subject to extreme functional diversification, resulting in allelic resistance specificities each recognizing a cognate but largely unidentified AVRA gene of the powdery mildew fungus, Blumeria graminis f sp hordei (Bgh). We performed RNA-sequencing of Bgh-infected barley leaves at two different time-points to obtain transcriptome data for 16 Bgh isolates, containing different AVRA genes. Subsequently, we analyzed the expression levels in the different isolates with a specific focus on previously described candidate secreted effectors (CSEPs) and additionally performed a transcriptome-wide association analysis including these 16 isolates and the previously published reference isolate DH14. These analyses identified AVRA1 and AVRA13, encoding two CSEPs that are recognized by MLA1 and MLA13 alleles, respectively. The avirulence function of both candidates could be verified by transient expression of the effector genes in barley leaves or protoplasts that was sufficient to trigger an MLA1 or MLA13 allele-specific cell death response. AVRA1 recognition by MLA1 is also retained in transgenic Arabidopsis lines.

A barley powdery mildew fungus non-autonomous retrotransposon encodes a peptide that supports penetration success on barley.

Project description:Pathogens overcome plant immunity by means of secreted effectors. Host effector targets often act in pathogen defense, but might also support fungal accommodation or nutrition. The barley ROP GTPase HvRACB is involved in accommodation of fungal haustoria of the powdery mildew fungus Blumeria graminis f.sp. hordei (Bgh) in barley epidermal cells. We found that HvRACB interacts with the ROP-interactive peptide 1 (ROPIP1) that is encoded on the active non-long terminal repeat retroelement Eg-R1 of Bgh. Overexpression of ROPIP1 in barley epidermal cells and host-induced post-transcriptional gene silencing (HIGS) of ROPIP1 suggested that ROPIP1 is involved in virulence of Bgh. Bimolecular fluorescence complementation and co-localization supported that ROPIP1 can interact with activated HvRACB in planta. We show that ROPIP1 is expressed by Bgh on barley and translocated into the cytoplasm of infected barley cells. ROPIP1 is recruited to microtubules upon co-expression of MICROTUBULE ASSOCIATED ROP GTPase ACTIVATING PROTEIN (HvMAGAP1) and can destabilize cortical microtubules. The data suggest that Bgh ROPIP targets HvRACB and manipulates host cell microtubule organization for facilitated host cell entry. This points to a possible neo-functionalization of retroelement-derived transcripts for the evolution of a pathogen virulence effector.

Project description:Membrane trafficking is vital to plant development and adaptation to the environment. It is suggested that post-Golgi vesicles and multivesicular bodies are essential for plant defence against directly penetrating fungal parasites at the cell wall. However, the actual plant proteins involved in membrane transport for defence are largely unidentified. We applied a candidate gene approach and single cell transient-induced gene silencing for the identification of membrane trafficking proteins of barley involved in the response to the fungal pathogen Blumeria graminis f.sp. hordei. This revealed potential components of vesicle tethering complexes [putative exocyst subunit HvEXO70F-like and subunits of the conserved oligomeric Golgi (COG) complex] and Golgi membrane trafficking (COPIγ coatomer and HvYPT1-like RAB GTPase) as essential for resistance to fungal penetration into the host cell.

Project description:Disease resistance is mediated by specific recognition of pathogen avriulence effectors (AVR) by nucleotide-binding leucine-rich repeat (NLR) receptors. The barley (Hordeum vulgare) mildew locus A (mla) resistance gene homolog 1 (RGH1) encoded NLRs (MLAs) confer isolate-specific resistance to the widespread mildew fungus Blumeria graminis forma specialis hordei (Bgh). In barley, MLA has been subject to extensive functional diversification, resulting in allelic resistance specificities, each recognizing a cognate Bgh AVRa. The by genetic association isolated AVRa1 and AVRa13 effectors belong to the candidate secreted effector protein (CSEP) gene family (Lu et al., 2016). To unravel the complex mechanisms underlying MLA functional diversification in barley and wheat, isolation of numerous Bgh AVRa genes and recognition of their gene products by MLA is necessary. Our here deployed higher resolution genetic association approach identified the Bgh avirulence gene candidate loci AVRa7, AVRa9, AVRa22 and AVRa10 by associating of transcript polymorphisms and AVRa phenotypes from a collection of 27 Bgh isolates. We collected 10 Bgh isolates from a local Bgh population in Cologne in addition to our previous collection of 17 Bgh isolates (Lu et al., 2016).
