There were 5 mice/time point for infections and 3 mice/time point for time-matched mocks. Lung samples were collected for virus load, transcriptional analysis and proteomics analysis. Weight loss and animal survival were also monitored.

Project description:Differential expression was determined in Calu-3 cells between mock infected and infection with one of 3 Influenza viruses (wild-type VN1203, VN1203 mutant PB1-F2del, VN1203 mutant PB2-627E) at different times post infection. Purpose: To obtain samples for transcriptional analysis in triplicate using the VN1203 pathogenicity mutants PB1-F2del and PB2-627E. Overview of Experiment: . Time Points: 0, 3, 7, 12, 18 and 24 hrs post infection. . There are two time points for wild type VN1203. . Done in triplicate. . Triplicates are defined as 3 different wells, plated at the same time using the same cell stock for all replicates. . Time-matched mocks were done in triplicate from the same cell stock as the rest of the samples. . Culture medium (the same as what the virus stock is in) was used for the mock infections. Calu-3 cells were infected with A/Vietnam/1203-CIP048_RG3/2004 (H5N1) (PB1-F2 deletion), A/Vietnam/1203-CIP048_RG3/2004 (H5N1) (PB2-627E mutant) or mock infected and samples were collected at 0, 3, 7, 12, 18 and 24 hpi. Calu-3 cells were infected with WT: A/Vietnam/1203/2004 (H5N1) and samples were collected at 7 and 24 hpi. There are 3 mock and 3 infected replicates for each time point. Expression profiles were determined.

Project description:A/Vietnam/1203-CIP048_RG4/2004 (H5N1) is an NS1trunc124 mutation in the wild type A/Vietman/1203/2004 (H5N1). The goal of this study was to determine the host response (C57BL/6 mouse model) to the NS1 truncation at both 10^3 PFU and 10^4 PFU. Groups of 20-week-old C57BL/6 mice were infected with the A/Vietnam/1203-CIP048_RG4/2004 (H5N1). This is an NS1trunc124 mutation in the wild type A/Vietnam/1203/2004 (H5N1) virus. Infections were done at 10^3 PFU or 10^4 PFU or time-matched mock infected. Time points were 1, 2, 4 and 7 d.p.i. There were 3-5 animals/dose/time point. Lung samples were collected for virus load, transcriptional and proteomics analysis. Weight loss and animal survival were also monitored.

Project description:The purpose is to obtain samples for mRNA, miRNA, proteomics, lipidomics, metabolomics, and histopathology analysis in mouse lung infected with WT A/Vietnam/1203/2004 (H5N1; 'VN1203'), VN12013-PB2-627E, VN1203-NS1trunc124, and WT A/California/04/2009 (H1N1; 'CA04'). Groups of 22-week-old C57BL/6 mice were infected with WT A/Vietnam/1203/2004 (H5N1; 'VN1203'), VN12013-PB2-627E, VN1203-NS1trunc124, and WT A/California/04/2009 (H1N1; 'CA04').Infections were done at 176 PFU/mouse for VN1203, 70 PFU/mouse for VN1203-PB2-627E, 1100 PFU/mouse for VN1203-NS1trunc124, and 10^4 PFU/mouse for CA04. Time points were 1, 2, 4, and 7 d.p.i. There were 8 mice for all infection groups and 5 mice for all mock groups. Lung samples were collected for virus load, transcriptional and proteomics analysis. Weight loss and animal survival were also monitored.
