Project description:Cumulus cells surrounding mature oocytes that developed to moruale/blastocyst stage on day 5 of IVF cycle were collected and used for gene expression profiling using Affymetrix Human Gene 1.0 ST Arrays in order to determine differences in gene expression between the modified natural and stimulated in vitro fertilization (IVF) procedures. Microarray analysis was made on 8 individual CC samples, derived from 5 patients that first underwent MNIVF, followed by a controlled ovarian hyperstimulation (COH) cycle. As the quality of the isolated RNA derived from 2 of the MNIVF CC samples was not good, these samples were not included in the analysis. Altogether, 3 CC samples from MNIVF and 5 CC samples from COH cycles were included in microarray analysis.

Project description:Currently, in vitro fertilization (IVF) is one of the most common approaches towards treating infertility. However, the success of this approach is still relatively low an the outcome of the IVF procedure is challenging to predict. At present, there is no highly successful marker that could reliably predict the pregnancy outcome in the process of assisted reproduction (AR). Currently, multiple embryo transfers are performed to warrant a successful pregnancy outcome, but this commonly results in multiple pregnancies, narrowing the feasibility of this method for a subset of couples affected by infertility. Recently IVF cycles with single-embryo transfers have become an option of interest in AR approaches. Such cycles, however, are significantly less successful and for this reason, there is an incentive to improve the pregnancy chance per IVF cycle by incorporating more criteria for selection of the embryo with the highest implantation potential. Gene expression in follicular cells, including granulosa cells (GC) and cumulus cells (CC) has previously been considered as a tool to predict the quality of the oocytes in several studies in addition to assessing the morphologic criteria of the embryos. Gene expression estimation in these cells, especially of the CC population, carries along several benefits as their harvesting is non-invasive, they are in direct contact with the oocyte and are usually discarded during the process of intracytoplasmatic sperm injection procedure (ICSI), making them any easily accessible surrogate tissue for gene expression analyses. Considering that several studies reporting various rates of success in prediction of AR cycle outcome with expression as a biomarker have been published, we attempted to perform the largest study of global gene expression alterations in finding novel biomarkers to outcome of in- vitro fertilization, which would encompass discovery of new biomarker genes and subsequent confirmational study of the selected biomarker set on an independent validation group. In this experiments 4 samples were hybridized twice, to allow for inspection of technical validity of microarray experiments.

Project description:The present study was undertaken to discover molecular markers in bovine cumulus cells predictive of oocyte competence and elucidate their functional significance. Differences in RNA transcript abundance in cumulus cells harvested from oocytes of adult versus prepubertal animals (model of poor oocyte quality) were identified by microarray analysis. Four genes of interest encoding for the lysosomal cysteine proteinases cathepsin B, S, K and Z and displaying greater transcript abundance in cumulus cells surrounding oocytes harvested from prepubertal animals were chosen for further investigation. Greater mRNA abundance for such genes in cumulus cells of prepubertal oocytes was confirmed by real time RT-PCR. Elevated transcript abundance for cathepsins B, S and Z was also observed in cumulus cells surrounding adult metaphase II oocytes that developed to the blastocyst stage at a low percentage following parthenogenetic activation, versus those that developed at a high percentage. Functional significance of cumulus cell cathepsin expression to oocyte competence was confirmed by treatment of cumulus oocyte complexes during in vitro oocyte maturation with a cell permeable cysteine proteinase (cathepsin) inhibitor. Inhibitor treatment decreased apoptotic nuclei in the cumulus layer and enhanced development of parthenogenetically activated and in vitro fertilized adult oocytes to the blastocyst stage. Stimulatory effects of inhibitor treatment during meiotic maturation on subsequent embryonic development were not observed when oocytes were matured in the absence of cumulus cells. Results support a functional role for cumulus cell cathepsins in compromised oocyte competence and suggest that cumulus cell cathepsin mRNA abundance may be predictive of oocyte quality. Cumulus cells were allocated into three groups according to the developmental potential of their enclosed oocyte: (1) not able to develop to the blastocyst stage (Bl–), (2) able to develop to the blastocyst stage but failing to establish a pregnancy (P–), or (3) able to develop to the blastocyst stage and to establish a clinical pregnancy (P+).

Project description:Follicle stimulating hormone (FSH) and epidermal growth factor (EGF) are currently used on cumulus-oocyte complexes to mimic the luteinizing hormone surge in vitro and induce oocyte maturation and cumulus expansion. We have previously shown that addition of exogenous recombinant growth differentiation factor 9 (GDF9) during oocyte in vitro maturation led to an improvement of oocyte quality, as shown by an increased blastocyst percentage and fetal survival. Our objective was to characterize the effect of FSH/EGF and GDF9 treatments on mouse cumulus cells expression profile by microarray analysis.

Project description:Besides the established selection criteria based on embryo morphology and blastomere number, new parameters for embryo viability are needed to improve the clinical outcome of in vitro fertilization (IVF) and more particular of elective single embryo transfer (eSET). The aim of the study was to analyse genome-wide whether the embryo viability was reflected by the expression of genes in the oocyte surrounding cumulus cells. Early cleavage (EC) was chosen as a parameter for embryo viability. Experiment Overall Design: Consenting patients visiting the IVF clinic underwent an IVF or ICSI treatment. Immediately following ultrasound-guided cumulus-oocyte-complex (COC) retrieval, a proportion of the cumulus cells surrounding the oocyte were removed. Gene expression in cumulus cells from eight oocytes resulting in an early cleavage embryo (EC-CC; n=8) and from eight oocytes resulting in a non-EC embryo (NEC-CC; n=8) derived from six patients were analysed using microarrays (n=16). To exclude a differential gene expression due to differences in patient characteristics, samples were paired. From four patients both an EC-CC and a NEC-CC sample were used. From two additional patients two EC-CC as well as two NEC-CC samples were used.

Project description:Differences between high- and low-potential COCs were examined by transcriptomic analysis of CC biopsies obtained from COCs of 2-6mm follicles from slaughterhouse ovaries before individual in vitro maturation, fertilization and culture until day 8 post-fertilization. Each COC was individually tracked and categorized based on his fate: embryo at blastocyst stage (CC-Blast) or embryo arrested at 2- to 8-cell stage (CC-2-8-cells). Average blastocyst rates were 27.7% for individual culture, and 31.2% for group control (not significantly different). Five cumulus biopsies per replicate were pooled for each fate. Three CC replicates underwent transcriptomic analysis using RNA microarray assay. Each cumulus cell sample is composed of 5 COC biopsies, 3 samples or biological replicates for each fate (CC-blast or CC-2-8-cells) were analysed by microarray. CC-2-8-cells condition was used as reference.

Project description:The granulosa cells in the mammalian ovarian follicle respond to gonadotropin signalling and are involved in the processes of folliculogenesis and oocyte maturation. Studies on gene expression and regulation in human granulosa cells are of interest due to their potential for estimating the oocyte viability and IVF success. The current study determined the mRNA profile by deep sequencing of the two intrafollicular somatic cell types: mural and cumulus granulosa cells isolated from women undergoing controlled ovarian stimulation and in vitro fertilization. Paired cumulus and mural granulosa samples were analysed from 3 women participating in IVF procedure. Differential gene expression study was performed. The identified gene expression profile was also used for predicting targets for miRNAs that were also identified from the same samples (GSE46489).
