Project description:To better understand the biological pathways by which UV inactivated SARS-CoV-induced pulmonary eosinophilia occurs, we examined global transcriptional changes in macrophages from the lungs of mouse. Female BALB/c mice were used at 21 weeks of age. Mice were subcutaneously immunized with UV inactivated SARS-CoV (UV-V) or UV-V and Toll like receptor (TLR) ligands at 6 and 1 weeks prior to mouse-adapted SARS-CoV (n=6 per group). Mice were intranasally challenged with 1E+6 TCID50 in 30M-NM-<L. MEM was challenged in six mice as control infection. Mice were sacrificed and collected lungs at 1 days after challenge, then CD11b positive cells were isolated from the lungs of these mice. These cells were used for the analysis of microarray.

Transcriptomes of SARS-CoV-2 naïve and recovered individuals vaccinated with inactivated vaccine

Project description:The urgent approval of the use of the inactivated COVID-19 vaccine is essential to reduce the threat and burden of the epidemic on global public health, however, our current understanding of the host immune response to inactivated vaccine remains limited. We performed transcriptomics analysis on 20 SARS-CoV-2 naïve individuals who received multiple doses of inactivated vaccine and five SARS-CoV-2 recovered individuals who received single dose of inactivated vaccine. These data help us understand the reaction mechanism of the host's molecular immune system to the inactivated vaccine, and provide a basis for the choice of vaccination strategy.

Detection SARS-CoV-2 infection in mice by ATR-FTIR

Project description:Vero-E6 cell supernatant, mice lavage and human saliva were evaulated after exposure to SARS-CoV-2 via ATR-FTIR spectoscopy. To provide further information about the physiological response, proteins contained in the mouse lavage were assessed via shotgun proteomics (data shown here). Mice were exposed to active SARS-CoV-2 or UV-inactivated SARS-CoV-2 for up to 4 days, lavage collected at day 0, day 2 and day 4 (D4). Lavage proteins were pooled for day 0 (due to low protein abundance) and compared to UV-I-SARS-CoV-2(n=4)and SARS-CoV-2(n=8)samples.

Mass-spectrometry phosphoproteomics of Calu-3 cell lines infected with SARS-CoV-2, in comparison to non-infected and UV mock-treated cell lines

Project description:To identify the proteome alterations in human epithelial cells upon SARS-CoV-2 infection, we performed an in vitro experiment, where we infected lung-adenocarcinoma cultured human airway epithelial cells (Calu-3) with an isolate of the ancestral variant and alpha variant of SARS-CoV-2 in two separate experiments. Samples were then collected 3 hours, 1 day, 3 days, and 7 days post infection. In parallel, for comparison, we collected samples on the same days from non-infected cells and cells treated with SARS-CoV-2 virus (of the corresponding variant) inactivated by ultraviolet radiation. All conditions were performed in biological triplicates and we performed phosphoproteomics analysis with high-pH fractionation and LC-MS/MS. Cell culture specifics Human lung adenocarcinoma Calu3 cells (ATCC, HTB-55) were grown in minimum essential medium (MEM) supplemented with 20% FBS, HEPES, L-glutamin, 100 U/ml penicillin, and 100 mg/ml streptomycin at 37°C and 5% CO2. The SARS-CoV-2 variants were propagated on Vero E6 cells and titrated via end point dilution assay. For UV-inactivation, the virus stock was incubated under UV-light for 3 x 1.5 min. Complete inactivation was confirmed via infection attempt and no live virus could be detected after the treatment. Cells were infected with active or UV-inactivated SARS-CoV-2 in complete MEM at a multiplicity of infection (MOI) of 1. After two  hours of incubation the virus solution was removed, the cells were washed, and fresh growth medium was added. Before the infection, the active and inactivated virus were treated with Trypsin at 1:100 dilution for one hour at 37°C.  Before sample collection at the indicated time points, the cells were washed thoroughly. Cells were lysed in lysis buffer containing 1mM DTT and boiled at 95°C for five minutes.

Project description:HAE cultures were infected with SARS-CoV, SARS-dORF6 or SARS-BatSRBD and were directly compared to A/CA/04/2009 H1N1 influenza-infected cultures. Cell samples were collected at various hours post-infection for analysis. Time Points = 0, 12, 24, 36, 48, 60, 72, 84 and 96 hrs post-infection for SARS-CoV, SARS-dORF6 and SARS-BatSRBD. Time Points = 0, 6, 12, 18, 24, 36 and 48 hrs post-infection for H1N1. Done in triplicate or quadruplicate for RNA Triplicates/quadruplicates are defined as 3/4 different wells, plated at the same time and using the same cell stock for all replicates. Time matched mocks done in triplicate from same cell stock as rest of samples. Culture medium (the same as what the virus stock is in) will be used for the mock infections. Infection was done at an MOI of 2.
