Project description:We observed that follicular dendritic cell line induced a new type of CD11b+ myeloid cells (FDMCs) when cultured with a lineage-negative c-kit+ population from mouse spleen cells. Thus, we used microarrays to characterize gene expression of FDMCs. FDMCs were induced on FDC line for RNA extraction and hybridization on Affymetrix microarrays. Microarray data of "FDC-induced myeloid cells (FDMCs)" sample were combined with a large collection of other cell- and tissue-specific gene expression data sets (total: 205 individual data sets). Raw data (.cel) files were normalized using Robust Multichip Analysis (RMA Express; http://rmaexpress.bmbolstad.com/), annotated using the latest libraries available from Affymetrix, and arranged according to cell-type grouping.

G-CSF mobilizes CD34+ regulatory monocytes that inhibit Graft-versus-Host Disease

Project description:Administration of G-CSF mobilizes a unique population of CD11b+Ly6C+CD34+mature monocytes that can inhibit GVHD in murine models of BMT via an iNOS-dependent mechanism. The transcriptional profiles of flow sorted lineage-CD11b+CD34+ cells from G-CSF treated mice were compared with conventional splenic Ly6C+ and Ly6C- monocytes, progenitor cells and cultured myeloid-derived suppressor cells. Further comparisons were made with lineage-CD11b+CD34+ cells from G-CSF treated mice that had been grown in culture or that were derived from iNOS ko mice. We used microarrays to detail the global programme of gene expression underlying diffrenetiation of each of these cell types Lin-CD11b+CD34+ populations were isolated directly from the spleens of G-CSF-treated C57BL/6 mice or iNOS ko mice. In untreated C57BL/6 mice, Lin-CD11b+CD115+Ly6C+ and Lin-CD11b+CD115+Ly6C- monocytes were isolated from the spleen and Lin-CD117+CD115+CD135-Ly6C+CD11b- common monocyte progenitors were isolated from the bone marrow. Myeloid-derived suppressor cells (Ly6C+CD11b+ cells derived from G-CSF, GM-CSF and IL-13 cultured C57BL/6 bone marrow) were also isolated and compared with the above populations. Lin-CD11b+CD34+ spleen cells derived from G-CSF-treated C57BL/6 mice were cultured for 3 days in Flt3 ligand and SCF and then compared to the original input population.

Lineage reprogramming of c-Kit+CD24+ natural killer cells into myeloid cells

Project description:Myeloid progenitor cells have generally been considered the exclusive source of myeloid cells under steady-state conditions. Here, we show that NK cells contributed to a myeloid cell lineage pool in naïve and tumor-bearing mice. By using fate tracing of NKp46+ cells, we found evidence that myeloid cell populations could be derived from NK-committed NKp46+ cells. Notably, among mature CD11b+CD27+ NK cells, c-Kit+CD24+ NK cells were capable of differentiating into a range of myeloid lineages in vitro and reprogrammed into neutrophils and monocytes in vivo. In addition to the potential for lineage conversion, c-Kit+CD24+ NK cells retained NK cell phenotypes and effector functions. Mechanistically, GATA-2 was essential for the conversion of the c-Kit+CD24+ NK cells into myeloid cells. Therefore, we discovered that GATA-2-mediated conversion of c-Kit+CD24+ NK cells contributes to myeloid cell development and identified a novel pathway for myeloid lineage commitment in physiological conditions.

Genome-wide expression study of the CD11b+ subsets of dermal myeloid cells and their migratory counterparts in the draining lymph node

Project description:Numerous CD11b+ myeloid cells are present within the dermis. They are very heterogeneous and can be divided in dermal DCs, tissue monocytes and tissue macrophages. At steady state, only CD11b+ DC migrate from the dermis to the skin draining lymph nodes whereas upon DNFB-induced inflammation, CD11b+ DC as well as dermal monocytes migrated to the lymph nodes. The objective of this study was to use gene expression profiling to rigorously identify the different subsets of dermal CD11b+ myeloid cells at steady state and upon inflammation and to characterize their functional potential.
