Project description:To elucidate roles that Nrf2 plays in regulating adaptive responses to the space stress, one month-long space travel and RNA-seq analysis of six Nrf2-KO mice and six wild-type mice were conducted . After 31-days-stay in the ISS, all the flight mice returned safely to Earth. We found that Nrf2 was indeed activated by the space stress in various tissues. The space stress and Nrf2-deficiency brought about changes in gene expression.

Project description:In situ synthesized oligo arrays, U74Av2, from Affymetrix were used to measure differential gene expression in RNA samples generated from the liver of Nrf2 knockout and wildtype mice at 5 month age. Total RNAs from two Nrf2 knockout or wildtype littermates were analyzed separately. There are two replicates (GSM 13431, 13435) for the female Nrf2 wildtype group, two replicates (GSM 13439, 13441) for the male Nrf2 wildtype group, two replicates (GSM 13436, 13437) for the female Nrf2 knockout group, and two replicates (GSM 13438, 13440) for the male Nrf2 knockout group. 5-Hydroxymethylcytosine profiling of aging hippocampi

Project description:We profiled 5-hydroxymethylcytosine patterns in hippocampi using hydroxymethylated DNA immunoprecipitation followed by Illumina deep sequencing (hMeDIP-Seq). We profiled the hippocampi of 4 young (3 month old) and 4 aged (18 month old) C57BL/6 mice. Additionally, we profiled the 5-hmC patterns of hippocampi that were stereotactically injected with lentiviral constructs that overexpressed Tet2 or luciferase.
