Project description:We used microarrays to compare the gene expression profiles of different H1N1 isolates (seasonal and pandemic) in lung epithelial cells in vitro. Well-differentiated primary human lung, bronchial epithelial cells (wd-NHBE) were infected with two pandemic and one seasaonl H1N1 influenza viruses at a multiplicity of infection of 3.0. The cells were then collected, in triplicate, at 36 hours after infection. This timepoint was selected as we saw significant differences in soluble cytokine and chemokine protein levels at this time point in the supernatant.

We showed that infection of macaques with two genetically similar but clinically distinct SOIV isolates from the early stage of the pandemic (A/Mexico/4108/2009 and A/Mexico/InDRE4487/2009) resulted in upper and lower respiratory tract infections and clinical disease ranging from mild to severe pneumonia. Disease associated with these SOIV isolates was clearly advanced over the mild infection caused by A/Kawasaki/UTK-4/2009, a current seasonal strain.

Importantly, A(H1N1)pdm09 virus was unable to develop resistance to MK2206. Thus, MK2206 is a potent anti-influenza A(H1N1)pdm09 agent.

Project description:Neutrophils are essential cells of host innate immunity. Although the role of neutrophils in defense against bacterial and fungal infections is well characterized, there is relative paucity of information about their role against viral infections. Influenza A virus (IAV) infection can be associated with secondary bacterial co-infection, and it has long been posited that the ability of IAV to alter normal neutrophil function predisposes individuals to secondary bacterial infections. To better understand this phenomenon, we evaluated the interaction of pandemic or seasonal H1N1 IAV with human neutrophils isolated from healthy subjects. These viruses were ingested by human neutrophils and elicited changes in neutrophil gene expression that are consistent with an interferon-mediated immune response. Viability of neutrophils following co-culture with either pandemic and seasonal H1N1 IAV was similar for up to 18 h of culture. Notably, neutrophil exposure to seasonal IAV (but not pandemic IAV) primed these leukocytes for enhanced functions, including production of reactive oxygen species and bactericidal activity. Taken together, our results are at variance with the universal idea that IAV impairs neutrophil function directly to predispose individuals to secondary bacterial infections. Rather, we suggest some strains of IAV prime neutrophils for enhanced bacterial clearance.

Project description:Pandemic H1N1 influenza A (H1N1pdm) elicits stronger pulmonary inflammation than previously circulating seasonal H1N1 influenza A (sH1N1), yet mechanisms of inflammatory activation in respiratory epithelial cells during H1N1pdm infection are unclear. We investigated host responses to H1N1pdm / sH1N1 infection and virus entry mechanisms in primary human bronchial epithelial cells in vitro. H1N1pdm infection rapidly initiated a robust inflammatory gene signature (3 h post-infection) not elicited by sH1N1 infection. Protein secretion inhibition had no effect on gene induction. Infection with membrane fusion deficient H1N1pdm failed to induce robust inflammatory gene expression which was rescued with restoration of fusion ability, suggesting H1N1pdm directly triggered the inflammatory signature downstream of membrane fusion. Investigation of intra-virion components revealed H1N1pdm viral RNA (vRNA) triggered a stronger inflammatory phenotype than sH1N1 vRNA. Thus, our study is first to report H1N1pdm induces greater inflammatory gene expression than sH1N1 in vitro due to direct virus-epithelial cell interaction. Total RNA obtained from Normal Human Bronchial Epithelial Cells (NHBEC) at 3 hours or 24 hours post-infection with either 0.9 MOI A/Mexico/4108/2009 (H1N1) or 0.9 MOI A/Brisbane/59/2007 (H1N1). Total RNA was also collected at 0 hours from uninfected NHBEC for each infection. Changes in gene expression relative to uninfected cells were then investigated.

Project description:Infection with pandemic A/H1N1 influenza virus induces high levels of circulating and lung pro-inflammatory cytokines and chemokines in experimental models and humans. These inflammatory mediators contribute to poor clinical outcomes of this infection. To compare the regulation of inflammatory responses in seasonal versus pandemic influenza A virus infections, we analyzed the expression of mRNA by the human gene st 1.0 array (affymetrix). We identifed a number of differentially expressed genes by infection of the A/H1N1 compared to the seasonal virus. Gene ontology analysis of these genes found that most of them are involved in several aspects of immunity. These studies suggest that factors inherent in the pandemic A/H1N1 viral strain may augment the production of inflammatory mediators by inhibiting SOCS- 1 expression and that infection with this viral strain also modifies the expression of several immunity related genes and pathways . The experimental design consist in 1 pool and its replicate containing 5 independent experiments from treated macrophages with Influenza A/H1N1 virus, and 1 more pool and its replicate containing 5 independent experiments from treated macrophages with seasonal influenza A virus. By this way, the total number of the samples analyzed was 10 in 4 microarrays.
