Gene expression profiling of sensitive skin

ABSTRACT: Sensitive skin is a hyperactive skin condtion, characterized by prickling, burning, itching, stinging, pain, or tingling sensation, which develops in response to various internal and external stimulants that can be tolerated by most people. The pathophysiology involved in the development of sensitive skin remains largely elusive. To characterize differential gene expression profiles in sensitive skin, we investigated the genome-wide patterns of gene expression from skin samples from subjects with sensitive or non-sensitive skin, following lactic acid challenge or normal saline application using cDNA microarrays.

The pathophysiology involved in the development of sensitive skin remains largely elusive. To characterize differential gene expression profiles in sensitive skin, we investigated the genome-wide patterns of gene expression from skin samples from subjects with sensitive or non-sensitive skin, following lactic acid challenge or normal saline application using cDNA microarrays. We recruited eighteen healthy volunteers with sensitive skin (n=9) or non-sensitive skin (n=9). From each subject, two skin samples were obtained following lactic acid or normal saline application.(18 samples/group) Based on the existence of sensitive skin and lactic acid challenge, samples were pooled into sets of three individuals each. Global transcriptome profiling was conducted using Affymetrix Human Genome U133 Plus 2.0 array.

We tested two different itch models-the AEW model (known commonly as the dry skin model) and the MC903 model. Performed peptide extraction from the dorsal horn (DH) and dorsal root ganglia regions of these two models and compared their levels with that of control mice.

Immune checkpoint blockade unleashes aberrant immune responses against the microbiota

Project description:Immune checkpoint inhibitors (ICIs) targeting PD-1 and CTLA-4 are essential components of the cancer therapeutic armamentarium. The remarkable responses seen with ICIs however, are also accompanied by immune-related adverse events (irAEs). These inflammatory side-effects impact virtually all organ systems, with the most common being the skin and gastrointestinal tract. Here, we establish a mouse model of commensal bacteria-driven skin irAEs and demonstrate that immune checkpoint blockade aberrantly unleashes commensal-specific T cell responses. Such responses caused widespread immune cell infiltration and inflammation throughout the skin, recapitulating the skin irAEs seen in patients treated with ICIs. This skin inflammation was dependent on production of the cytokine IL-17 by commensal-specific T cells, which in turn induced hyperactive responses from macrophages and myeloid cells. Importantly, these responses had long-term consequences, with commensal-specific T cells elicited in the context of ICIs being highly sensitive to re-activation by commensals months later. Together, our results establish a mouse model of skin irAEs and show that immune checkpoint blockade can potentiate aberrant immunity to skin commensals with long-lasting consequences.

RNA-seq of mouse tissue samples from animal model：Effects of MMP9 on gene expression.

Project description:In the present study, RNA-seq technique were performed To explore biological significance of MMP9 in the neuro-cutaneous signaling network of chronic itch. We used MMP-9-IN-1, a specific inhibitor of MMP9, or its vehicle to treat ACD model mice, and conducted high-throughput sequencing on ear tissue samples of mice. It was found that inhibition of MMP9 expression affected the expression of a variety of itching factors and skin inflammatory factors related to the skin pathology of chronic dermatitis. These changes may indicate that MMP9 is closely related to inflammation and pruritus. This finding might help us to better understand the immune-neuro-modulatory mechanism in itch circuits.

Metabolic Reprogramming and Reliance in Human Skin Wound Healing

Project description:Impaired skin wound healing is a significant global health issue, especially among the elderly. Wound healing is a well-orchestrated process involving the sequential phases of inflammation, proliferation, and tissue remodeling. Although wound healing is a highly dynamic and energy-requiring process, the role of metabolism remains largely unexplored. By combining transcriptomics and metabolomics of human skin biopsy samples, we mapped the core bioenergetic and metabolic changes in normal acute as well as chronic wounds in elderly subjects. We found upregulation of glycolysis, the tricarboxylic acid cycle, glutaminolysis, and β-oxidation in the later stages of acute wound healing and in chronic wounds. To ascertain the role of these metabolic pathways on wound healing, we targeted each pathway in a wound healing assay as well as in a human skin explant model using metabolic inhibitors and stimulants. Enhancement or inhibition of glycolysis and, to a lesser extent, glutaminolysis had a far greater impact on wound healing than similar manipulations of oxidative phosphorylation and fatty acid β-oxidation. These findings increase the understanding of wound metabolism and identify glycolysis and glutaminolysis as potential targets for therapeutic intervention.

Transcriptome analysis of the host immune responses to Sarcoptes scabiei

Project description:Scabies is a human skin disease due to the burrowing ectoparasite Sarcoptes scabiei resulting in intense itching and inflammation and manifesting as skin allergy. Little is known about the specific scabies molecules and genes involved in the host inflammatory and immunologic responses. We interrogated the transcriptome profiles of skin biopsies using next generation sequencing and a combined clustering and pathway mapping approach which will enable us in the identification of key signaling events in the host immune and inflammatory responses to S. scabiei infestation. With the comprehensive analysis of RNA-seq results, it has been found that the immune response of host inhibits S. scabiei immune pathway, proliferation and metabolism signaling pathways, out of which the signaling pathway seems to be the main pathway in innate immune response of S. scabiei. The analysis of immune pathway, hydrolase and allergen will contribute to the research of S. scabiei pathogenesis, diagnosis and development of vaccines.
