Project description:We report the results of chromatin immunoprecipitation following by high-thoughput tag sequencing (ChIP-Seq) using the GA II platform from Illumina for the human transcription factor STAT1 in HeLa S3 cells. The STAT1 ChIP was performed using HeLa S3 cells that are stimulated using gamma-interferon. We have also generated a seqenced input DNA dataset for gamma-interferon stimulated HeLa S3 cells. Project description:Cisplatin resistance is a problem in cancer treatment. Using DNA microarray, we detected differentially expressed genes in cisplatin-resistant cervix carcinoma HeLa cells compared to parental cells. Three cisplatin resistant cell lines were established by stepwise increasing cisplatin concentration. RNA from these resistant lines and its parental HeLa cells were labeled with Cy5 and Cy3. Equal amount of RNA from resistant cell line and HeLa were mixed and were hybridized to cDNA array. Signals were scanned and analyzed to find out the candidate genes involved in cisplatin resistant mechanism.

Project description:Transcriptional profiling of HeLa cells comparing control untreated HeLa cells with IFN-gamma-treated HeLa cells To infect host cells, many viruses use small cellular compartments, called endosomes, for entry, and the thiol/disulfide interchange in viral envelope glycoproteins (Envs) is crucial for infection. By screening cysteine-reacting chemicals, we found a compound, 4,4’-dithiopyridine, which is active at acidic pH, efficiently restricts retrovirus vector infection. We thus hypothesized that some products of endosome-localized, interferon-stimulated genes (ISGs) exhibit anti-viral activity by inhibiting thiol/disulfide interchange in viral Envs. Among the hundreds of ISGs, gamma-interferon (IFN)-inducible lysosomal thiolreductase (GILT) is the only molecule that resides in the endosomes/lysosomes and digests the S-S bonds of proteins under acidic conditions. We now report that GILT significantly inhibits the replication of retroviruses, including HIV-1 and MLV, by restricting both the early and late phases of their life cycles. Using the VSV-G pseudotyped HIV-1 model, we found that GILT digests the S-S bonds of viral Env proteins. GILT also inhibits HIV-1 viral release by digesting the S-S bond of CD63, an endosome-localized molecule reportedly involved in HIV-1 particle release. The effect of -IFN on HIV-1 is limited, although GILT induced by gamma-IFN is supposed to have anti-HIV-1 activity. We found that while gamma-IFN effectively inhibits MLV replication through GILT, the gamma-IFN signaling is remarkably inhibited by the HIV-1 Env protein, but not the MLV Env protein. These findings suggest that GILT functions as an anti-viral host factor induced by gamma-IFN, however, HIV-1 may have evolved to inhibit gamma-IFN signaling by the Env protein.

Project description:Epithelial cells provide an initial line of defense against damage and pathogens in barrier tissues such as the skin; however this balance is disrupted in obesity and metabolic disease. Skin gamma delta T cells recognize epithelial damage and release cytokines and growth factors that facilitate wound repair. To determine the impact of obesity and metabolic disease on skin gamma delta T cells, we isolated skin gamma delta T cells from 10-week old C57BLKS/J lean db/+ and obese db/db animals for further study. Due to a deficiency in the leptin receptor (db), homozygous db/db animals do not process satiety signals, continually eat and develop severe obesity and metabolic disease. Skin gamma delta T cells isolated from these animals were compared for changes in mRNA expression using microarray. We have determined that obesity and metabolic disease negatively impacts homeostasis and functionality of skin gamma delta T cells, rendering host defense mechanisms vulnerable to injury and infection. The goal of this experiment was to compare skin gamma delta T cells in a control mouse to skin gamma delta T cells isolated from an obese mouse to see what homeostatic changes occur in obesity and metabolic disease. gamma delta T cells were isolated from two 10-week old lean db/+ control and two 10-week old obese db/db animals for comparison. We wanted to determine which growth factors and signaling pathways were altered in skin gamma delta T cells residing in the obese environment.
