Brown Adipose Tissue (BAT) in Visceral Fat Depot

ABSTRACT: Case story. A patient with massive infiltration of the visceral adipose tissue depot by BAT in a patient with a catecholamine secreting paraganglioma. BAT tissue was identified by protein expression of UCP1 (western blotting and immunostaining) The goal of the study is to identify patterns of gene expression in BAT containing visceral fat compared to the patient's own subcutanous fat which did not express BAT. For comparison a pool of mRNA isolated from visceral fat from obese subjects was used.

Adipose depot differences in high fat fed mice

Project description:Purpose : To understand differences in microRNA (miRNA) signatures of different adipose tissue depots to gain mechanistic insight regarding their contribution to metabolic disorders in obesity. Method : We performed small RNA-sequencing of brown, subcutaneous and visceral adipose depots from high fat diet-fed mice (mice were fed a 45% kcal fat diets from 5-6 weeks of age for 11 weeks). The main animal study reference: Kalupahana et al., J. Nutr, 2010 Results:Using the Gunaratne Next Generation pipeline (published in Creighton et al. 2009) miRNA expression profiles were identified. Counts of each unique read were normalized to total usable reads, and had 40 counts added. We mapped about 13.8 million sequence reads per sample to the Mus musculus genome (build mm 10). Altogether 1251 miRNAs were identified in three adipose tissues and out of which 246 showed differential expression with 1.25 or more fold change and p value <0.05 in one or more of the pairwise comparisons of the 3 adipose depots. Hierarchical clustering, demonstrated that biological replicates behaved appropriately as they were clustered together. BAT exhibited a different gene/ miRNA expression pattern than the 2 other WATs (SAT and VAT). SAT and VAT showed a closer relationship with each other than with BAT Conclusion : High fat diet differentially regulate specific miRNAs expression in different adipose tissue depots

SARS-CoV-2 infects adipose tissue in a fat depot- and viral lineage-dependent manner

Project description:Visceral adiposity is a risk factor for severe COVID-19, and a link between adipose tissue infection and disease progression has been proposed. Here we demonstrate that SARS-CoV-2 infects human adipose tissue and undergoes productive infection in human primary adipose-derived stromal-vascular cells differentiated into adipocytes. However, the permissiveness to infection and the cellular response depends on the anatomical origin of the cells and the viral lineage. Cells of visceral adipose tissue origin express more ACE2 and are more permissive to SARS-CoV-2 infection than their subcutaneous counterpart. SARS-CoV-2 infection leads to inhibition of lipolysis in cells of subcutaneous origin, while in visceral fat cells, it results in higher expression of pro-inflammatory cytokines. Viral load and cellular response are attenuated when visceral adipose tissue cells are infected with the SARS-CoV-2 gamma variant. A similar degree of cell death occurs 4-days after SARS-CoV-2 infection, regardless of the cell origin or viral lineage. Hence, SARS-CoV-2 infects human adipose tissue cells, replicating and altering cell function and viability in a fat depot- and viral lineage-dependent fashion.

Lipid-Associated Macrophages Reshape BAT Cell Identity in Obesity [DBDB_Bulk]

Project description:Obesity and type 2 diabetes cause a loss in brown adipose tissue (BAT) activity in mice and human, but the molecular mechanisms that drive BAT cell remodeling remain largely. Using a multilayered approach, we comprehensively map a deep reorganization in the BAT cells. We uncovered a subset of macrophages as the lipid-associated macrophages (LAM), which were massively increased in genetic and dietary model of BAT expansion. LAM participate in this scenario by capturing extracellular vesicles carrying damaged lipids and mitochondria released from metabolically-stressed brown adipocytes. CD36 scavenger receptor drives LAM phenotype and through * in vitro* and *in vivo* models, we demonstrated that CD36-deficient LAM increased brown fat genes. LAM release Tgfb1 that reduces brown adipocytes identity through Aldh1a1 induction. This study provides the first description of cell dynamics in BAT of obese models identifying LAM as responder to tissue-level metabolic stress and key driver to loss of BAT cell identity.
