Expression data from footpad of AIRmax and AIRmin mice submitted to pristane arthritis induction

ABSTRACT: Mice selected for high and low acute inflammation were tested for pristane induced arthritis, showing to be susceptible and resistant, respectively. We used microarrays to detail the global programme of gene expression underlying arthritis progression during induction and identified distinct classes of differentially expressed genes during this process in the footpad of these mice.

Project description:Mice selected for high and low acute inflammation were tested for pristane induced arthritis, showing to be susceptible and resistant, respectively. We used microarrays to detail the global programme of gene expression underlying arthritis progression during induction and identified distinct classes of differentially expressed genes during this process in the footpad of these mice. AIRmax (high inflammation) and AIRmin (low inflammation) mouse footpads were collected for RNA extraction and hybridization on Affymetrix microarrays. We compared control and experimental footpads from male and female AIRmax and AIRmin mice on days 0 and 160, respectively. Please note that the AIRmax and AIRmin mice were obtained through bidirectional genetic selection, starting with a genetically heterogeneous founder population (F0) produced by intercrossing eight isogenic strains of mice of independent origins (A/J, DBA/2J, P/J, SWR/J, SJL/J, CBA/J, BALB/cJ and C57BL/6J).

The objective was to obtain a molecular understanding of the protective effects of phytol on pristane induced arthritis (PIA). The animals were sacrificed close to disease onset, i.e. ten days after administration.

Project description:Rhesus theta defensin-1 (RTD-1), a macrocyclic immunomodulatory host defense peptide from Old World monkeys, is therapeutic in pristane-induced arthritis (PIA) rat model. RNA-sequence (RNA-seq) analysis was used to interrogate the changes in gene expression in PIA rats diseased model. To, study the anti-arthritic mechanisms of systemic RTD-1, the differentially expressed genes between synovial tissues of rats with pristane induced arthritis (PIA) and naïve animals were identified and the effect of RTD-1 treatment for 1, 3 or 5 days was then evaluated. The results indicate a regulation of arthritogenic gene networks in a manner that correlates temporally with clinical resolution of rat PIA.

NK cell-derived GM-CSF is critical for enhancing pathological arthritis

Project description:GM-CSF is involved in immune complex (IC)-mediated arthritis. However, little is known about what is the cellular source of GM-CSF and how it is regulated during IC-mediated inflammation. Using novel GM-CSF reporter mice, we show that NK cells produce GM-CSF during an IC-mediated model of inflammatory arthritis. NK cells promoted STIA in a GM-CSF-dependent manner, as deletion of NK cells and selective removal of GM-CSF production by NK cells abrogated disease. Furthermore, we show that myeloid cell activation by GM-CSF is restrained by induction of JAK/STAT checkpoint inhibitor cytokine-inducible SH2-containing protein, CIS. Myeloid cells from CIS-deficient mice had exaggerated responses to GM-CSF, and these mice develop exacerbated STIA. Our data suggest that tissue NK cells may amplify joint inflammation in arthritis via GM-CSF production and thus represent a novel target in IC-mediated pathology. Endogenous CIS provides a key brake on signaling through the GM-CSF receptor and strategies that boost its function may provide an alternative anti-inflammatory approach.

Gene expression profile of preclinical arthritis and its modulation by antigen-induced tolerance

Project description:During the course of adjuvant arthritis, maximal changes in gene expression were observed at the incubation phase. A major group of genes affected was related to immune activity. Tolerance induction by mycobacterial heat-shock protein 65 (Bhsp65), the disease-related antigen, caused upregulation of a large number of genes. These included immune activity genes as well as cell proliferation-related genes. Functional differences of experimental arthritis splenic macrophages in DA and DAMut rats are revealed by quantitative proteomics

Project description:Rheumatoid arthritis (RA) is a devastating chronic inflammatory disorder affecting 0.5-1.0% of adults worldwide and is characterized by autoimmune reactivity and persistent active inflammation with concurrent tissue destruction. Animal models of RA provide an opportunity to understand disease pathogenesis and develop new therapeutic strategies . The DA inbred rat is commonly used in autoimmunity research due to its high susceptibility to several chronic inflammatory disorders such as arthritis and experimental autoimmune encephalomyelitis (EAE). The experimental arthritis induced by pristane, the arthritogenic component in mineral oil, in the DA rat can be classified with RA criteria, including a chronic relapsing disease course 5, 6. We have identified a mutation DA rat and the mutation plays a completely protective function in DA rats against EAE and pristane-induced arthritis (PIA). Macrophages are an essential source of pro-inflammatory cytokines in chronic inflammation and the progressive destruction of synovial joints. To elucidate the molecular mechanism by which the mutation regulates macrophage activation, we made quantitative mass spectrometry on the four groups of splenic macrophages: DA-control, DAMut-control, DA-PIA, DAMut-PIA.
