Project description:To characterize naproxen and NO-naproxen, their effects on gene expression in livers of treated rats was examined. We have previously worked with liver and could therefore identify a moderate to strong antioxidant response element signature. There were two goals to this study: 1) to determine whether naproxen and NO-naproxen yield similar gene expression profiles, which would imply that their effects are mediated by the parent NSAID, and 2) to determine whether NO-naproxen, due to the release of NO, might cause increases in expression of genes associated with the antioxidant response element (ARE). In this dataset, we include the expression data obtained from liver of untreated rats and rats treated with naproxen (400mg/kg diet) or NO-naproxen (550mg/kg diet) for 7 days. Total 12 samples were analyzed. We generated the following comparisons: NO-Naproxen vs. Control and Naproxen vs. Control. Genes with a p-value < 0.05 and a fold-change M-bM-^IM-%1.5 were selected.

Project description:The molecular mechanisms of exercise-induced cardiovascular protection are poorly understood. There is growing evidence that reactive oxygen species (ROS) are necessary for some of these adaptations and antioxidants may be used to investigate this effect. This study aimed to determine the effects of exercise and/or antioxidant supplementation on myocardial and vascular endothelium gene expression. Male Wistar rats were divided into four groups: i) endurance exercise (90min of treadmill running 4d/week, 14 weeks); ii) antioxidant-treated; iii) antioxidant and endurance exercise and iv) control. The supplemented animals received Vitamin E (1000 IU/kg diet) and ï¡-lipoic acid (1.6 g/kg diet) mixed with rat chow. cDNA microarray analysis was performed using purified endothelial RNA from myocardial and coronary artery endothelial cells and showed that the expression levels of 35, 40 and 40 genes were altered for groups i, ii, and iii respectively compared to control. Differentially expressed genes were analysed using the KEGG pathway database, hierarchical cluster and DAVID analysis. These analyses revealed that a gene involved in cardiovascular disease progression, Ras homolog gene family member A (RhoA) was down-regulated by exercise, upregulated by antioxidant supplementation and the combination of exercise and antioxidant blunted both effects. These findings were confirmed by real-time PCR. Project description:A rexinoid, targretin (TRG) and two retinoids, 9-cis retinoic acid (9cRA) and 4-hydroxyphenylretinamide (4HPR) were examined for their effects on gene expression in rat mammary gland, liver, and lung tissues. The chemopreventive effects of these agents have largely been attributed to their ability to interact with retinoic acid receptors (RAR) and/or retinoid X-receptors (RXR). TRG interacts with the RXR receptors. 9cRA interacts with both the RAR and RXR receptors, while 4HPR has a moderate affinity primarily for RAR gamma. Based on previously performed mammary chemoprevention studies, TRG (150mg/Kg diet), 9cRA (100 mg/Kg diet), and 4HPR (792 mg/Kg diet), were administrated to rats continually in their diet for 7 days. Tissue - and agent - specific expression differences were determined by comparing tissues from treated rats with those form rats administered a control diet. There were significantly more changes seen associated with TRG than 9cRA or 4HPR. Only a limited number of expression changes were found with 4HPR treatment. For each organ, TRG and 9cRA treated tissues clustered closely together, whereas 4HPR treated tissues clustered with the tissues from the control diet group. In contrast to 9cRA treatment, TRG treatment altered genes that involved fatty acid metabolism and modulation of various cytochromes P450 in the liver clearly demonstrating the very disparate nature of these two retinoids. These expression signatures could provide useful pharmacodynamic biomarkers for retinoidsâ treatment and chemoprevention. Project description:We investigated the novel molecular mechanisms of the antioxidant and anti-inflammatory properties of S-allylmercaptocysteine (SAMC) based on a transcriptomic study in a non-alcoholic fatty liver disease (NAFLD) rat model. NAFLD was induced in Sprague-Dawley rats by feeding with a high fat diet (HFD) for 12 weeks. 200mg/kg SAMC was fed by oral gavage for 4 weeks from 9-12 week. We found that SAMC co-administration attenuated HFD-induced liver injury, including the increased serum ALT, hepatic oxidative stress and inflammation. Transcriptomic analysis revealed that SAMC dramatically induced the XRE- and ARE-driven drug metabolising enzymes (DMEs) including Akr7a3, Akr1b8, and NQO1. The nuclear translocation of the upstream regulator of xenobiotics metabolism, AhR, and regulator of antioxidant responses, Nrf2, were significantly increased by SAMC treatment. Furthermore, SAMC counteracted the effects of HFD on NF-κB/IκB and NLRP3/6 pathways with decreasing protein levels of ASC, cleaved caspase-1, IL-18 and IL-1β. These results were further verified in another mice NASH model induced by an MCD diet with SAMC co-administration. Conclusion: we propose that SAMC triggers AhR/Nrf2-mediated antioxidant responses which may further suppress the NLRP3/6 inflammasome pathway and NF-κB activation, contributing to the improvement of NAFLD/NASH.

Project description:In our previous study, we reported that a diet rich in antioxidants such as coral calcium hydride (CCH) increased the endogenous antioxidant ability in the hippocampus of rats. We conducted this study to test the hypothesis that diet supplementation with CCH would change the gene expression in rats and to understand how CCH enhances antioxidant ability. We used a DNA array to compare the expression levels in the hippocampus of rats fed with CCH for 2 weeks with those of rats fed a normal diet. Immune response-related genes were down-regulated, while nuclear respiratory factor 2 and aldehyde dehydrogenase 3A were up-regulated. Our findings about the changes in the mRNA levels of these genes well explain the physiological finding of enhanced antioxidant ability in rat brain.

Project description:Selenium (Se) is an essential cofactor of the antioxidant enzyme glutathione peroxidase beside other functions. The evaluation of optimal selenium supplementation in chicken feed and the subsequent effects on animal health and performance requires comprehensive knowledge of the overall metabolic effects of selenium. Therefore the gene expression was measured in the control group with a standard diet and in the group with a Se supplemented diet (0.5mg Se/kg diet) to determine significantly altered gene expression. The selenium was supplemented in the form of selenized yeast (Se-yeast), which mainly consists of organic Se in the form of L-selenomethionine and L-selenocysteine. The control group received a diet, which contained 70μg of Se / kg diet and the Se-yeast group 620μg of Se / kg diet (analyzed). The one-day old broiler chicks were separated into two groups and received the control or the Se-supplemented diet ad libitum for 35 days. After slaughter the gene expression was determined in the liver of four control and five samples from the Se-yeast group. One sample from the control group did not correspond to the quality requirements and was excluded from the analysis.

Project description:The standardized extract of Centella asiatica (ECa233) contains >80% triterpenoids with a ratio of madecassoside to asiaticoside of 1.5(±0.5):1. Hepatoprotective effect of ECa233 has not yet been reported. The aim of this study was to examine the effect of ECa233 on liver metabolome of rotenone-treated rats to better understand the underlying mechanisms of this bioactive substance in liver tissue. Rats were randomly divided into three groups (with six rats/group): sham negative control, rotenone positive control, and the ECa233 test group. Rats in the ECa233 group received 10 mg/kg ECa233 orally for 20 days, followed by 2.5 mg/kg intraperitoneal rotenone injection to induce toxicity before being sacrificed. The liver samples were collected for antioxidant assays and untargeted metabolomic analysis using a GC-MS. Antioxidant tests revealed that ECa233 inhibited lipid peroxidation and increased catalase activities in liver tissue. Orthogonal partial least squares-discriminant analysis clearly showed complete separation between control and rotenone groups, whereas the ECa233 group was located between these two groups and tended to reverse the effects of rotenone treatment. Two candidate markers were identified, pipecolinic acid and Unknown #21. Findings from this study suggested that supplementation of ECa233 protected rat liver against rotenone toxicity. However, further investigations are needed to elucidate the mechanisms involved.
