Project description:This study tests whether hepatic steatosis, independent of inflammation, alters the hepatocyte protein secretory profile, and examines whether changes in the secretory products contribute to the development of metabolic dysfunction in other cell types. Mouse hepatocytes were isolated by flow cytometry and cultured in serum-free conditions. Conditioned media was used for LC/MS analysis to compare hepatocytes from chow fed animals to high-fat diet animals with liver steatosis.

Effect of hepatocyte-specific PPARgamma knock out in the regulation of hepatic gene expression in mice fed a low fat diet or and high fat, cholesterol and fructose diet that induces NASH

Project description:In this study we sought to determine how hepatocyte-specific PPARgamma expression regulates hepatic gene expression in normal (healthy) mice fed a LFCF diet, or mice that were fed a HFCF diet for 24 weeks to induce non-alcoholic steatohepatitis (NASH). Specifically, we used chow-fed adult (10 week-old) PPARgamma floxed male mice and injected them with AAV8-TBG-Cre to knockout PPARgamma in hepatocytes (KO). A subset of PPARgamma floxed mice were injected with AAV8-TBG-Null to generate controls (C). Two weeks after AAV injections, half of the mice in each group were fed a low fat, cholesterol and fructose (LFCF) diet, or a high fat, cholesterol and fructose (HFCF) diet for 24 weeks. Livers were collected at the end of the study to assess hepatic gene expression with RNA-seq (performed by Novogen, Inc.)

Systems analysis of hepatic insulin resistance identifies RGS4 as a key mediator of metabolic adaptation

Project description:We profiled the transcriptomes of chow diet-fed and high-fat diet-fed mice across insulin stimulation time points. We also profiled the effects of metformin treatment on mouse liver transcritomes. Our temporal analyses of chow-diet and 16 week high-fat diet transcriptomics following insulin stimulation uncovered several regulator of G-protein signaling genes, particularly Rgs4, that are specifically regulated transcriptionally in HFD livers following insulin stimulation. We validated these findings with additional assays of RGS4 mRNA and protein levels and characterized this molecule's role in regulating hepatic insulin responses in mice.

Liver-specific matrin-3 knockout alters the hepatic transcriptome of chow diet-fed mice [RNA-seq]

Project description:Matrin-3 is an RNA-binding protein involved in the pathogenesis of human diseases. Here we examined the hepatic transcriptome of chow-diet fed mice. Bulk RNA-seq and bioinformatics analysis identified 646 and 145 differentially expressed genes (Padj < 0.05), respectively in the livers of female and male matrin-3 LKO mice. Enrichr analysis of these DEGs revealed several common Hallmark terms including cholesterol homeostasis, fatty acid metabolism, xenobiotic metabolism, and epithelial mesenchymal transition between female and male mice. Our data demonstrated that liver-specific matrin-3 deficiency re-programs the hepatic transcriptome in the liver of chow diet-fed mice.

Nordihydroguaiaretic Acid Improves Metabolic Dysregulation and Aberrant Hepatic Lipid Metabolism in Mice by Both PPAR-alpha-Dependent and -Independent Pathways

Project description:To test whether NDGA attenuate dyslipidemia and hepatic steatosis by enhancing fatty acid oxidation through activation of PPAR-α. Using wild type (WT, C57BL/6) fed with chow diet as control, WT mice were either fed with high-fat diet or high-fat diet with NDGA (2.5g/kg food); ob/ob mice were fed with either chow or chow with NDGA (2.5 g/kg food), and maintained on the respective diets for 16 weeks. The expression of lipid metabolism related genes in the liver of these mice were analyzed using Phalanx GPL6845 platform (Mouse OneArray V1). Together with other biochemical/physiological data, our results suggest that the beneficial actions of NDGA on dyslipidemia and hepatic steatosis in ob/ob mice are exerted primarily through enhanced fatty acid oxidation and energy utilization via the activation of PPAR- α receptor activity.

Hepatic transcriptome profiling of liver-specific Xbp1 knockout mice fed a high fat diet

Project description:Xbp1 is an important regulator of unfolded protein response and lipid metabolism. Its dyregulation has been associcated in human NASH. Feeding a high fat diet with fructose/sucrose to mice causes progressive, fibrosing steatohepatitis. This study is to use RNA-Seq to identify differentially expressed genes in hepatic Xbp1 deficient mice livers fed with a high fat diet compared to controls. Hepatic Xbp1 deficient mice or flox controls were fed either regular chow or a high fat diet (n=4). Samples from each cohort were pooled into two replicates.
