Project description:A high-resolution time series study of transcriptome dynamics following antimiR--mediated inhibition of miR-9 in a Hodgkin lymphoma cell-line revealed both general and miR-9 specific aspects of the miRNA--mediated post--transcriptional dynamic response.MiR-9 inhibition induced a multiphasic gene response, with an initial direct response at approximately 4 hours and multiple later responses which showed transcription factor enrichments indicative of indirect causally downstream responses, and an overall shift of gene product function from predominantly mRNA processing at early time points to translation at later time points. The full time course used was: 2,4,6,8,10,12,14,16,18,20,22,26,32,40,56,80,112 hours post-miR-9 inhibition. Microarrays were Affymetrix HuGene-10-st-v1. Each LNA-antimiR treated cell sample was matched with an LNA scramble control at each time point. This overall time schedule was divided into two independent biological replicates: one covering early time points (2 hours to 20 hours), and another covering later time points (22 hours to 112 hours).

Project description:Expression of the miR-34 family (miR-34a, -34b, -34c) is elevated in settings of heart disease, and inhibition with antimiR-34a/antimiR-34 has emerged as a promising therapeutic strategy. Under chronic cardiac disease settings, targeting the entire miR-34 family is more effective than targeting miR-34a alone. The identification of transcription factor (TF)-miRNA regulatory networks has added complexity to understanding the therapeutic potential miRNA-based therapies. Here, we sought to determine whether antimiR-34 targets secondary miRNAs via TFs which could contribute to antimiR-34-mediated protection. Using miRNA-Seq we identified differentially regulated miRNAs in hearts from mice with cardiac pathology due to transverse aortic constriction (TAC), and these miRNAs were also regulated by antimiR-34. Two clusters of stress-responsive miRNAs were classified as “pathological” and “cardioprotective”. Using ChIPBase we identified 45 TF binding sites on the promoters of “pathological” and “cardioprotective” miRNAs, and 5 represented direct targets of miR-34, with the capacity to regulate other miRNAs. The expression of two “pathological” miRNAs (let-7e and miR-31) was independently experimentally validated in hearts from antimiR-34 treated TAC mice, and may explain why targeting the entire miR-34 family is more effective than targeting miR-34a alone. AntimiR-34 regulates the expression of other miRNAs and this has significant implications for drug development.

Project description:Background MicroRNAs are potent regulators of biology and disease. The miR-15 family has been shown to regulate cardiomyocyte proliferation and antimiR-based inhibition induces a cardioprotective effect after myocardial infarction in mice. However, systemic delivery of antimiRs leads to accumulation in kidneys and liver, with relatively poor cardiac exposure. pH-responsive injectable hydrogels serve as a sustained-release drug delivery depot and could potentially be used to improve cardiac efficacy of antimiR therapeutics. Objective Examine whether hydrogel can improve local delivery of antimiR-195 in ischemic hearts to increase cardiac efficacy and limit off-target effects. Methods Study the effect of intramyocardial injections of hydrogel-formulated antimiR-195 under both baseline conditions and after ischemic injury. Results Intracardiac injections of UPy-PEG induced a transient inflammatory response that was no longer present 7 days post-injection. In vitro experiments showed that antimiR-195 was released from the gel, and induced microRNA inhibition leading to downstream cardiomyocyte proliferation. In vivo, intramyocardial delivery of antimiR-195 in UPy-PEG enhanced cardiac target de-repression compared to PBS-dissolved antimiR-195, despite a low cardiac retention. After ischemic injury, this translated into a greater therapeutic effect by increasing both target de-repression and cardiomyocyte proliferation. Conclusions UPy-PEG can be used as a cardiac delivery vehicle of antimiRs and intramyocardial injection of UPy-PEG formulated antimiR-195 is sufficient to improve cardiac efficacy of antimiR-195. Follow up experiments in large animals will enable us to assess the true added value of using UPy-PEG to increase cardiac exposure of antimiR therapies.

Fluorescence in situ hybridization demonstrated uptake of the LNA-antimiR in mouse liver cells, which was accompanied by markedly reduced hybridization signals for mature miR-122 in treated mice. Functional antagonism of miR-122 was inferred from a low cholesterol phenotype and de-repression within 24 h of 199 liver mRNAs showing significant enrichment for miR-122 seed matches in their 3' UTRs. Expression profiling extended to 3 weeks after the last LNA-antimiR dose revealed that most of the changes in liver gene expression were normalized to saline control levels coinciding with normalized miR-122 and plasma cholesterol levels. Combined, these data suggest that miRNA antagonists comprised of LNA are valuable tools for identifying miRNA targets in vivo and for studying the biological role of miRNAs and miRNA-associated gene-regulatory networks in a physiological context. Project description:Rapid and reliable methods for conducting biological dosimetry are a necessity in the event of a large-scale nuclear event. Timely and accurate methods are thus critical in order to correctly evaluate and identify radiation dose exposure and triage appropriately. Alterations in microRNAs (miRNAs), small non-coding RNAs of 19-22 nucleotides, have been reported in cells/tumors subjected to radiation exposure, implying that miRNAs play an important role in cellular stress response to radiation. To examine the possibility of using microRNA as stable blood based biomarkers for radiation response, we have used microRNA microarray analysis. Differential microRNA expression pattern was evaluated (>1.5 fold and p value <0.05) at 6, 24, 48 h and 7day time points in whole blood from mice exposed to 2, 4, 8, 12 and 15Gy irradiation. More differentially expressed microRNAs (up and down) were seen after higher dose of radiation and later time points. Among the significantly upregulated microRNAs, miR-193b-3p and mir-92a-3p were consistently upregulated for all doses at earlier time points. We observed significant downregulation of several members of the miR-17 family (miR-17-5p, miR-20a-5p, miR-93-5p and miR-106b-5p) in response to radiation from 2 Gy onwards at 24 h, 48 h and at higher doses for the 7 d time-point. Whereas miR-34a, which regulates G1/S cell cycle checkpoint activation in response to radiation induced DNA damage, showed consistent upregulation for both low and high doses at later time points up to one week after exposure. Significant down regulation of miR-150-5p, miR-101, miR-140, miR-29b, miR-193b and miR-30 family of microRNAs were seen for low and high doses of radiation for all time points up to 7day. Low dose radiation (2Gy) exposure also resulted in down regulation of a group of microRNAs after one week; whereas these microRNAs showed upregulation after high dose exposure (8, 12 and 15Gy) up to one week. This dose and time dependent differential microRNA expression pattern might be useful in evaluating radiation response during and after therapeutic radiation treatment in patients. Ultimately, a combined approach of identifying biomarkers by assessing functional miRNA, target mRNAs and the resulting proteins to assess radiation exposure after mass-casualty incidents could provide a valuable tool in developing and implementing effective and timely medical countermeasures.
