Project description:Brains are sexually dimorphic in adult zebrafish. We dissected brains from young and old, adult zebrafish, from both males and females. Brains are not pooled but analyzed as indivual samples. Four groups of wild-type zebrafish (AB strain) were used for this study: young (7.5-8.5 months old) male and female and old (31-36 months old) male and female. There were three animals per group for a total of 12 animals in the study.

Project description:Background: Changes in gene regulation are thought to be crucial for the adaptation of organisms to their environment. Transcriptome analyses can be used to identify candidate genes for ecological adaptation, but can be complicated by variation in gene expression between tissues, sexes, or individuals. Here we use high-throughput RNA sequencing of a single Drosophila melanogaster tissue to detect brain-specific differences in gene expression between the sexes and between two populations, one from the ancestral species range in sub-Saharan Africa and one from the recently colonized species range in Europe. Results: Relatively few genes (<100) displayed sexually dimorphic expression in the brain, but there was an enrichment of sex-biased genes, especially male-biased genes, on the X chromosome. Over 340 genes differed in brain expression between flies from the African and European populations, with the between-population divergence being highly correlated between males and females. The differentially expressed genes include those involved in stress response, olfaction, and detoxification. Expression differences were associated with transposable element insertions at two genes implicated in insecticide resistance (Cyp6g1 and CHKov1). Conclusions: Analysis of the brain transcriptome revealed many genes differing in expression between populations that were not detected in previous studies using whole flies. There was little evidence for sex-specific regulatory adaptation in the brain, as most expression differences between populations were observed in both males and females. The enrichment of genes with sexually dimorphic expression on the X chromosome is consistent with dosage compensation mechanisms affecting sex-biased expression in somatic tissues. mRNA profiles of Drosophila melanogaster brains from adult males and females from a European and an African population (2 biological replicates each)

The objective is to identify genes that show sexually dimorphic expression in the brain of this species. For this experiment, Asian seabass were collected from the Marine Aquaculture Center of the Agri-Food & Veterinary Authority of Singapore. There were no treatments carried out in this experiment. Four brains from adult male seabass (5 years old) with M3-type testis and four brains from adult female seabass (5 years old) with F3-type ovaries were used in this experiment. (Gonads were examined by histology and classified according to sexual maturation status as described by Guiguen and colleagues (Guiguen et al. Environmental Biology of Fishes, 1994)). Project description:Purpose: The goal of this study was to compare gene expression patterns in the male and female human cortex Methods: We performed RiboZero Gold (rRNA depleted) 50bp PE RNA-seq in a set of control samples of both sexes to identify sexually dimorphic gene expression patterns. Results: Within these samples, we corroborated findings from a discovery set of RNA-seq data from adult human cortex tissue from the BrainSpan consortium which demonstrated male-biased expression of astrocyte marker genes and a gene co-expression module found to be up-regulated in the adult autistic cortex. Conclusions: These findings suggest that sex-differential risk for autism spectrum disorder is not the result of sex-differential regulation of ASD risk genes, but of naturally occurring sexually dimorphic processes that modulate the impact of risk variants for autism spectrum disorder. 13 cerebral cortex samples from 10 individuals (7 samples from 5 males, 6 samples from 5 females). Project description:Stalk-eyed flies (family Diopsidae) are a model system for studying sexual selection due to the elongated and sexually dimorphic eye-stalks found in some species. These flies are of additional interest because their X chromosome is derived largely from an autosomal arm in other flies. To investigate how sex-biased expression arose on the novel X we compared gene expression between males and females using oligonucleotide microarrays and RNA from developing eyestalk tissue in the sexually monomorphic diopsid, Teleopsis quinqueguttata. We use probe sequence divergence to evaluate cross-species ascertainment bias and chromosome assignment to determine if sex linkage influence expression. Microarray analysis revealed sex-biased expression for only 1.9% of 3,748 genes expressed in eye-antennal imaginal discs. Analysis of probe sequences between species indicates that the lack of sex-biased expression is not due to ascertainment bias. These findings indicate that the the majority of sex-biased gene expression observed in developing heads of the dimorphic species, T. dalmanni, is causally related to development of dimorphic head shape. Project description:There is increasing appreciation for sexually dimorphic effects, but the molecular mechanisms underlying these effects are only partially understood. In the present study, we explored transcriptomics and epigenetic differences in the small intestine and colon of prepubescent male and female mice. In addition, the microbiota composition of the colonic luminal content has been examined. At postnatal day 14, male and female C57BL/6 mice were sacrificed and the small intestine, colon and content of luminal colon were isolated. Gene expression of both segments of the intestine was analysed by microarray analysis. DNA methylation of the promoter regions of selected sexually dimorphic genes was examined by pyrosequencing. Composition of the microbiota was explored by deep sequencing. Sexually dimorphic genes were observed in both segments of the intestine of 2-week-old mouse pups, with a stronger effect in the small intestine. Amongst the total of 349 genes displaying a sexually dimorphic effect in the small intestine and/or colon, several candidates exhibited a previously established function in the intestine (i.e. Nts, Nucb2, Alox5ap and Retnlγ). In addition, differential expression of genes linked to intestinal bowel disease (i.e. Ccr3, Ccl11 and Tnfr) and colorectal cancer development (i.e. Wt1 and Mmp25) was observed between males and females. Amongst the genes displaying significant sexually dimorphic expression, nine genes were histone-modifying enzymes, suggesting that epigenetic mechanisms might be a potential underlying regulatory mechanism. However, our results reveal no significant changes in DNA methylation of analysed CpGs within the selected differentially expressed genes. With respect to the bacterial community composition in the colon, a dominant effect of litter origin was found but no significant sex effect was detected. However, a sex effect on the dominance of specific taxa was observed. This study reveals molecular dissimilarities between males and females in the small intestine and colon of prepubescent mice, which might underlie differences in physiological functioning and in disease predisposition in the two sexes. Project description:Purpose: This study assessed putative sex differences in miRNA expression in the bed nucleus of the stria terminalis (BNST)—a sexually dimorphic brain region implicated in anxiety—of adult male and female rats that had been exposed throughout adolescence to social isolation (SI) stress. Methods: Male and female Sprague-Dawley rats underwent SI during adolescence or remained grouped housed (GH) till adulthood. Small RNA sequencing was performed on tissue extracted from the anterodorsal BNST. Results: SI compared to GH induced more anxiogenic-like effects in females compared to males and sex-dependent changes in miRNA expression in the BNST. Conclusions: The current study shows that housing conditions (either SI or GH) during adolescence regulates miRNA expression in a sex-specific manner in the sexually dimorphic area of the adBNST
