To identify genes rapidly responding to elevated ATAF1 expression estradiol induction experiments were performed on ATAF1-IOE seedlings after 1 h, 2 h and 5 h of ATAF1 induction. In addition, expression profiles were obtained from mature leaves after 5 h of ATAF1 induction.

Genome wide expression analysis of EST-induced MYB112 (MYB112-IOE) using Affymetrix ATH1 array

Project description:Global transcriptome patterns were determined in MYB112-IOE seedlings induced for 3 h and 5 h with b-estradiol in order to identify the genes early regulated by MYB112 transcription factor. We used microarrays to identify genes differentially expressed in EST-inducible MYB112 over-expression line #1, 3 h and 5 h after 10µM EST induction, compared to mock treated plants. Two independent biological replications were performed for 5 h timepoint and one replicate for 3 h timepoint.

Project description:We designed an experiment to identify BAK1 interactors considering that it could be a coreceptor for the GOLVEN6 (GLV6) signaling peptide involved in lateral root organogenesis in Arabidopsis thaliana. A BAK1pro:BAK1-GFP/bak1-4 line crossed to an estradiol-inducible GLV6 overexpression line (BAK1pro:BAK1-GFP/bak1-4xiGLV6) was treated with mock or estradiol followed by affinity purification mass spectrometry (AP-MS) experiments using anti-GFP beads in microsomal-enriched fractions. Statistical analysis of estradiol-treated vs. mock seedlings did not reveal significant differences. Nevertheless, we remarked that regardless of the treatment, several LRR-RLKs co-eluted with BAK1-GFP, comprising known, as well as, unknown interactors. We confirmed interaction of BAK1 with these LRR-RLKs by BIFC and identified the PXC3 receptor as a BAK1 constitutive interactor, which was further verified by co-IP.

Transcription profiling of Arabidopsis pER22-AGL24 seedlings treated by estradiol vs. controls

Project description:To understand the transcriptional program controlled by AGL24, we took advantage of a functional estradiol-inducible AGL24 expression system in combination with microarray analysis to identify AGL24 target genes. Experiment Overall Design: We firstly compared the transcriptomes in pER22-AGL24 seedlings induced by estradiol- relative to mock-treatment, which revealed the differentially expressed genes corresponding to the elevated AGL24 expression induced by estradiol. As a chemical, estradiol itself could trigger a series of elevated or repressed genes, which might represent a large portion of false positive targets of AGL24. To rule out the genes expressed exclusively in response to estradiol instead of induced AGL24 activity, we also compared the estradiol-induced transcriptomes in pER22-AGL24 relative to those in estradiol-induced wild-type seedlings because only the former contained the induced AGL24 expression. The genes showing different expression in these two comparisons only responded to induced AGL24 activity rather than estradiol- or mock-treatment. Experiment Overall Design: Two replicates of microarray experiments were performed with biologically independent pools of samples. For pER22-AGL24 seedlings treated by estradiol- relative to mock-treatment, gene expression in mock-treated seedlings was used as the baseline, while for estradiol-treated pER22-AGL24 and wild-type seedlings, gene expression in estradiol-treated wild-type seedlings was used as the baseline.
