Metabolomic Profiling of Human Pluripotent Stem Cell Differentiation into Lung Progenitors

Project description:Metabolism is vital to cellular function and tissue homeostasis during human lung development. In utero, embryonic pluripotent stem cells undergo endodermal differentiation towards a lung progenitor cell fate that can be mimicked in vitro using induced human pluripotent stem cells (hiPSCs) to study genetic mutations. To identify differences between wild type and surfactant protein B (SFTPB)-deficient cell lines during endoderm specification towards lung, we used an untargeted metabolomics approach to evaluate the developmental changes in metabolites. We found that the metabolites most enriched during the differentiation from pluripotent stem cell to lung progenitor cell, regardless of cell line, were sphingomyelins and phosphatidylcholines, two important lipid classes in fetal lung development. The SFTPB mutation had no metabolic impact on early endodermal lung development. The identified metabolite signatures during lung progenitor cell differentiation may be utilized as biomarkers for normal embryonic lung development.

The quantitative proteomes of human-induced pluripotent stem cells and embryonic stem cells

Project description:Assessing relevant molecular differences between human-induced pluripotent stem cells (hiPSCs) and human embryonic stem cells (hESCs) is important, given that such differences may impact their potential therapeutic use. Controversy surrounds recent gene expression studies comparing hiPSCs and hESCs. Here, we present an in-depth quantitative mass spectrometry-based analysis of hESCs, two different hiPSCs and their precursor fibroblast cell lines. Our comparisons confirmed the high similarity of hESCs and hiPSCS at the proteome level as 97.8% of the proteins were found unchanged. Nevertheless, a small group of 58 proteins, mainly related to metabolism, antigen processing and cell adhesion, was found significantly differentially expressed between hiPSCs and hESCs. A comparison of the regulated proteins with previously published transcriptomic studies showed a low overlap, highlighting the emerging notion that differences between both pluripotent cell lines rather reflect experimental conditions than a recurrent molecular signature. circRNome of human pluripotent stem cells

Project description:Induction of pluripotency in somatic cells needs the full activation of the pluripotency gene regulatory network (PGRN). The core transcription factors of the PGRN establish a crosstalk with specific micro RNA (miRNA) families to sustain the pluripotent program and govern cell fate decisions. Very recently, circular RNA (circRNA) have been proposed as novel players in the regulation of this molecular circuitry. Herein, we successfully generated human induced pluripotent stem cells (hiPSC) by zero-footprint reprogramming of cord blood mesenchymal stem cells. The hiPSC were extensively characterized for stemness and tri-lineage differentiation potential compared to human embryonic stem cells and parental unreprogrammed cells to assess complete acquisition of the pluripotent identity. High-throughput array-based molecular analyses of messenger RNA (mRNA) (631 genes), miRNA (754 miRNA) and, for the first time, circRNA (13,617 circRNA) were performed to address the role of circRNA in the PGRN. As a result, a circRNA-guided map of miRNA and mRNA associated to naïve and primed pluripotent identity is provided.
