Rd10 transcriptome analysis

ABSTRACT: Retinitis pigmentosa (RP) is a photoreceptor disease that affects approximately 100,000 people in the United States. There are currently very limited treatment options and the prognosis for most patients is progressive vision loss. Unfortunately, the understanding of the molecular underpinnings of RP initiation and progression is still poorly understood. However, the development of animal models of RP, coupled with high-throughput sequencing, has provided an opportunity to study the underlying cellular and molecular changes of this disease. Using RNA-Seq, we present the first retinal transcriptome analysis of the rd10 murine model of retinal degeneration.

Project description:In inherited retinal disorders such as retinitis pigmentosa (RP), rod photoreceptor-specific mutations cause primary rod degeneration that is followed by secondary cone death and loss of high-acuity vision. Mechanistic studies of retinal degeneration are challenging because of retinal heterogeneity. Moreover, the detection of early cone responses to rod death is especially difficult due to the paucity of cones in the retina. To resolve heterogeneity in the degenerating retina and investigate events in both types of photoreceptors during primary rod degeneration, we utilized droplet-based single-cell RNA sequencing in an RP mouse model, rd10.

Project description:Retinal degeneration is the leading cause of irreversible blindness. Retinitis pigmentosa (RP) is a genetically heterogenous group of diseases. In the United States, approximately one in 4000 individuals is affected. RP begins with the loss of night vision due to the loss of rod photoreceptor cells. The disease progresses slowly with the loss of peripheral vision, and eventually leads to complete debilitating and irreversible blindness. The first mutation associated with human RP was identified in the gene encoding rhodopsin, the G-protein coupled receptor of rod photoreceptor cells. Mutations within the rhodopsin gene account for significant portion of RP cases. Specifically, mutations of the proline at residue 347 in rhodopsin have been linked to human RP.

Mesenchymal Stem Cell-Derived Extracellular Vesicles Protect Retina in a Mouse Model of Retinitis Pigmentosa by Anti-inflammation Through miR-146a-Nr4a3 Axis

Project description:Retinitis pigmentosa (RP) is a rod-cone degenerative disease that induces irreversible vision loss. Stem cell-derived extracellular vehicles (EVs) have been reported to prevent retinal neurons from apoptosis and inflammation, but the molecular mechanisms remained unclear. Here we probed the ability of mesenchymal stem cell-derived EVs (MSC-EVs) to protect the retinas of RP model mice rd10 and explored the underlying mechanisms. Treatment with MSC-EVs increased the survival of photoreceptors and preserved their structure and visual function. Mechanistically, MSC-EVs suppressed the activation of microglial, Müller glial, macrophages and the NF-κB pathway. MSC-EVs upregulated anti-inflammatory cytokines and downregulated pro-inflammatory cytokines. MSC-EVs application in vitro decreased the number of TUNEL-positive photoreceptor cell line 661W co-cultured with LPS-stimulated glial cell BV2, with similar impact on the cytokine expression as in vivo study. MiR-146a, one of the major miRNAs in MSC-EVs was upregulated in co-cultured cells after MSC-EVs treatment. Upregulation of miR-146a decreased the expression of the transcription factor Nr4a3, and its inhibition promoted Nr4a3 expression in both cells. Nr4a3 was further identified as the target gene of miR-146a. Therefore, MSC-EVs delays retinal degeneration in rd10 mice mainly by its anti-inflammatory effect via the miR-146a-Nr4a3 axis, indicating a strong potential of applying MSC-EVs to treat neurodegenerative diseases.

Retinal proteome profiling of inherited retinal degeneration across three different mouse models suggests common drug targets in retinitis pigmentosa

Project description:Inherited retinal degenerations (IRDs) are a leading cause of blindness among the young population in the developed world. Approximately half of IRDs initially manifest as gradual loss of night vision and visual fields, characteristic of retinitis pigmentosa (RP). Due to challenges in genetic testing, and the large heterogeneity of mutations underlying RP, targeted gene therapies are an impractical largescale solution in the foreseeable future. For this reason, identifying common key pathophysiological pathways in IRDs that could be used as targets for mutation-agnostic and disease-modifying therapies (DMTs) is warranted. In this study, we investigated retinal proteome of three distinct IRD mouse models, comparing to sex- and age-matched wild-type mice. Specifically, we used the Pde6βRd10 (rd10) and RhoP23H/WT (P23H) mouse models of autosomal recessive and autosomal dominant RP, respectively, as well as the Rpe65-/- mouse model of Leber´s congenital amaurosis type 2 (LCA2). The mice were housed at two distinct institutions and analyzed using LC-MS in three separate facilities/instruments following data-dependent and data-independent acquisition modes. This cross-institutional and multi-methodological approach signifies the reliability and reproducibility of the results. The largescale retinal proteome profiling, coupled with in vivo electroretinography recordings, provided us with a reliable basis for comparing the disease phenotypes and severity.
