Project description:The roles of histone demethylase KDM7 in gene expression were analyzed by gene expression profiling experiments with mouse neuroblastoma cell line Neuro2A. Keywords: mouse neuroblastoma, Neuro2A, gene expression profiling, microarray, Affimetrix M430 2.0 chip In order to examine the effect of KDM7 in gene expression, we generated stable KDM7 knockdown cell lines in mouse neuroblastoma cell line Neuro2A. Total RNAs were extracted from 5 cell lines (parental cells: Neuro2A, empty vector: Neuro2A transfected with empty vector, EGFP KD: Neuro2A transfected with vector for EGFP knock down, KDM7 KD1: Neuro2A transfected with vector 1 for KDM7 knock down, and KDM7 KD2: Neuro2A transfected with vector 2 for KDM7 knock down) and analyzed for gene expression profiles using Affymetrix platform.

Project description:To clarify the effect of SHP in LXRs-mediated signaling pathway, we performed global gene expression analysis of SHP siRNA transfected- or control siRNA transfected- astrocytes after IFN-γ and LXRs agonist. Microarray analysis revealed that expression of several genes encoding inflammatory mediators were reversed in SHP siRNA transfected-astrocytes, when compared with control siRNA transfected-astrocytes.

Project description:The molecular mechanism responsible for cell fate after mitotic slippage is unclear. We investigate the postmitotic effects of different mitotic aberrations, misaligned chromosomes produced by CENP-E siRNA (siCENP-E), and monopolar spindles resulting from Eg5 siRNA (siEg5). To determine which signaling pathways contribute to the postmitotic effect of siCENP-E in the presence of siBubR1 (siCENP-E+siBubR1) compared with siEg5+siBubR1, we performed a comprehensive gene expression analysis by microarray comparisons. HeLa cells were treated with siNS (non-silencing) +siBubR1, siCENP-E+siBubR1, or siEg5+siBubR1. The siNS cells were used as a control. Each siRNA treatment was duplicated. Three days after the siRNA treatments, the siRNA-transfected HeLa cells were collected for RNA extraction and hybridization on Affymetrix microarrays.
