Project description:The plant sterol pathway exhibits a major biosynthetic difference as compared with that of metazoans. The committed sterol precursor is the pentacyclic cycloartenol (9β,19-cyclolanost-24-en-3β-ol) and not lanosterol (lanosta-8,24-dien-3β-ol), as it was shown in the late sixties. However, plant genome mining over the last years revealed the general presence of lanosterol synthases encoding sequences (LAS1) in the oxidosqualene cyclase repertoire, in addition to cycloartenol synthases (CAS1) and to non-steroidal triterpene synthases that contribute to the metabolic diversity of C30H50O compounds on earth. Furthermore, plant LAS1 proteins have been unambiguously identified by peptidic signatures and by their capacity to complement the yeast lanosterol synthase deficiency. A dual pathway for the synthesis of sterols through lanosterol and cycloartenol was reported in the model Arabidopsis thaliana, though the contribution of a lanosterol pathway to the production of 24-alkyl-Δ(5)-sterols was quite marginal (Ohyama et al. (2009) PNAS 106, 725). To investigate further the physiological relevance of CAS1 and LAS1 genes in plants, we have silenced their expression in Nicotiana benthamiana. We used virus induced gene silencing (VIGS) based on gene specific sequences from a Nicotiana tabacum CAS1 or derived from the solgenomics initiative (http://solgenomics.net/) to challenge the respective roles of CAS1 and LAS1. In this report, we show a CAS1-specific functional sterol pathway in engineered yeast, and a strict dependence on CAS1 of tobacco sterol biosynthesis.

Project description:To obtain an interpretation from the view of transcriptome on distinct metabolite accumulation between ecologically different regions in China, next-generation sequencing technology was performed on E-L 31, 35，36 and 38 stages of Muscat Blanc a Petits Grains grape berries from Changli (CL, eastern) and Gaotai (GT, western). Transcriptome analysis revealed that some key genes involved in terpene synthesis were markedly up-regulated in the CL region. Particularly in the MEP pathway, the expression of VvHDR (1-hydroxy-2-methyl-2-butenyl 4-diphosphate reductase) paralleled with the accumulation of terpenes, which can promote the flow of isopentenyl diphosphate (IPP) into the terpene synthetic pathway. The glycosidically bound monoterpenes accumulated differentially along with maturation in both regions, which is synchronous with the expression of a monoterpene glucosyltransferase gene (VvGT14). Other genes were also found to be related to the differential accumulation of terpenes and monoterpene glycosides in the grapes between regions. Transcription factors that could regulate terpene synthesis were predicted through gene co-expression network analysis. Additionally, the genes involved in abscisic acid (ABA) and ethylene signal responses were expressed at high levels earlier in GT grapes than in CL grapes.

Project description:Diterpene biosynthesis commonly originates with the methylerythritol phosphate (MEP) pathway in chloroplasts, leading to the C20 substrate, geranylgeranyl pyrophosphate (GGPP). The previous work demonstrated that over-expression of genes responsible for the first and last steps in the MEP pathway in combination with GERANYLGERANYL PYROPHOSPHATE SYNTHASE (GGPPS) and CASBENE SYNTHASE (CAS) is optimal for increasing flux through to casbene in Nicotiana benthamiana. When the gene responsible for the last step in the MEP pathway, 4-HYDROXY-3-METHYLBUT-2-ENYL DIPHOSPHATE REDUCTASE (HDR), is removed from this combination, casbene is still produced but at lower amounts. Here, we report the unexpected finding that this reduced gene combination also results in the production of 16-hydroxy-casbene (16-OH-casbene), consistent with the presence of 16-hydroxy-geranylgeranyl phosphate (16-OH-GGPP) in the same material. Indirect evidence suggests the latter is formed as a result of elevated levels of 4-hydroxy-3-methyl-but-2-enyl pyrophosphate (HMBPP) caused by a bottleneck at the HDR step responsible for conversion of HMBPP to dimethylallyl pyrophosphate (DMAPP). Over-expression of a GERANYLLINALOOL SYNTHASE from Nicotiana attenuata (NaGLS) produces 16-hydroxy-geranyllinalool (16-OH-geranyllinalool) when transiently expressed with the same reduced combination of MEP pathway genes in N. benthamiana. This work highlights the importance of pathway flux control in metabolic pathway engineering and the possibility of increasing terpene diversity through synthetic biology.

Additionally, the genes involved in abscisic acid (ABA) and ethylene signal responses were expressed at high levels earlier in GT grapes than in CL grapes. cDNA libraries generated from four developmental stages (E-L 31, 35, 36 and 38) of Muscat Blanc a Petits Grains grape berries in two consecutive years from Changli (CL) and Gaotai (GT) in China were sequenced using Illumina HiSeq™ 2000.Only one RNA-seq library was constructed for each of the E-L31 and E-L36 stage samples because of the small amount of high quality RNA acquired, whereas two libraries were constructed for each of the E-L35 and E-L38 stage samples. As a result, a total of 24 libraries were obtained for the two regions within two years.

Project description:The sesquiterpene β-caryophyllene is an ubiquitous component in many plants that has commercially been used as an aroma in cosmetics and perfumes. Recent studies have shown its potential use as a therapeutic agent and biofuel. Currently, β-caryophyllene is isolated from large amounts of plant material. Molecular farming based on the Nicotiana benthamiana transient expression system may be used for a more sustainable production of β-caryophyllene. In this study, a full-length cDNA of a new duplicated β-caryophyllene synthase from Artemisia annua (AaCPS1) was isolated and functionally characterized. In order to produce β-caryophyllene in vitro, the AaCPS1 was cloned into a plant viral-based vector pEAQ-HT. Subsequently, the plasmid was transferred into the Agrobacterium and agroinfiltrated into N. benthamiana leaves. The AaCPS1 expression was analyzed by quantitative PCR at different time points after agroinfiltration. The highest level of transcripts was observed at 9 days post infiltration (dpi). The AaCPS1 protein was extracted from the leaves at 9 dpi and purified by cobalt-nitrilotriacetate (Co-NTA) affinity chromatography using histidine tag with a yield of 89 mg kg-1 fresh weight of leaves. The protein expression of AaCPS1 was also confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and western blot analyses. AaCPS1 protein uses farnesyl diphosphate (FPP) as a substrate to produce β-caryophyllene. Product identification and determination of the activity of purified AaCPS1 were done by gas chromatography-mass spectrometry (GC-MS). GC-MS results revealed that the AaCPS1 produced maximum 26.5 ± 1 mg of β-caryophyllene per kilogram fresh weight of leaves after assaying with FPP for 6 h. Using AaCPS1 as a proof of concept, we demonstrate that N. benthamiana can be considered as an expression system for production of plant proteins that catalyze the formation of valuable chemicals for industrial applications.

Project description:Plant triterpenoids play a critical role in plant resistance against Phytophthora infestans de Bary, the causal pathogen of potato and tomato late blight. However, different triterpenoids could have contrasting functions on plant resistance against P. infestans. In this study, we targeted the key biosynthetic gene of all plant triterpenoids, SQUALENE SYNTHASE (SQS), to examine the function of this gene in plant-P. infestans interactions. A post-inoculation, time-course gene expression analysis revealed that SQS expression was induced in Nicotiana benthamiana but was transiently suppressed in Solanum lycopersicum. Consistent with the host-specific changes in SQS expression, concentrations of major triterpenoid compounds were only induced in S. lycopersicum. A stable overexpression of SQS in N. benthamiana reduced plant resistance against P. infestans and induced the hyperaccumulation of stigmasterol. A comparative transcriptomics analysis of the transgenic lines showed that diverse plant physiological processes were influenced by SQS overexpression, suggesting that phytosterol content regulation may not be the sole mechanism through which SQS promotes plant susceptibility towards P. infestans. This study provides experimental evidence for the host-specific transcriptional regulation and function of SQS in plant interactions with P. infestans, offering a novel perspective in examining the quantitative disease resistance against late blight.
