RNA-seq analysis provided insight into how inactivation of AprE, GidA and a PIN-domain protein influences global gene expression mRNA profiles of wild type and mutants were generated by deep sequencing and mapped to SCBI genome by CLC Gemonic Workstation. The transcriptome profiles of the mutants were compared to the wild type.

Stationary phase stimulon and CspC regulon

Project description:The cold shock proteins belong to a family of RNA binding proteins presenting a highly conserved domain, called cold shock domain (CSD). They are involved in various cellular processes, including adaptation to low temperature, nutritional stress, cell growth and stationary phase. Here we investigate the role of CspC in C. crescentus stationary phase and the molecular mechanisms underlying gene regulation by this protein. A global transcriptional profiling experiment comparing cspC and the wild type strain both at exponential and stationary phases was carried out. The results showed that the absence of cspC affected the transcription of 20 genes at exponential phase and 65 genes at stationary phase. Genes encoding enzymes of the glyoxylate cycle were severely downregulated in the mutant at stationary phase. The stationary phase-induced RNA binding protein CspC has an important role in gene expression at this phase. It is required for the expression of the essential gene sciP, the ECF sigma factor sigU, as well as of the genes for the glyoxylate cycle enzymes and for oxidative stress response.

D6 PROTEIN KINASE plasma membrane polarity is dependent on a repeated CXX(X)P motif and serine phosphorylation

Project description:D6 PROTEIN KINASE (D6PK) is a polarly localized plasma membrane-associated kinase from Arabidopsis thaliana that activates polarly distributed PIN-FORMED (PIN) auxin transporters. D6PK traffics rapidly to and from the plasma membrane, independently from its PIN targets. D6PK plasma membrane association is dependent on the middle domain, an insertion between kinase subdomains VII and VIII. How D6PK polar plasma membrane targeting is established and maintained remains to be understood. Here, we show that cysteines of repeated CXX(X)P motifs in the middle domain are S-acylated and required for D6PK membrane association. While D6PK S-acylation is not detectably regulated during intracellular trafficking, the phosphorylation of adjacent serine residues, in dependence of the upstream regulatory 3-PHOSPHOINOSITIDE-DEPENDENT PROTEIN KINASES1, regulates D6PK residence time at the plasma membrane, lateral diffusion, and trafficking. We describe a mechanism for the regulation of D6PK plasma membrane interaction and polarity, which may also explain the polarity regulation of related kinases.
