UV Stress Regulates Differential Associations of Brd4 and P-TEFb with Chromatin and Modulates Transcription by Brd4 Acting as either Activator or Repressor

ABSTRACT: Purpose: The goal of this study is to investigate the role of Brd4 in UV-regulated genome-wide gene expression. Methods: RNA samples from control and Brd4-depleted cells at 0, 0.5 and 2 hr after UV treatment were analyzed using RNA-sequencing (RNA-seq). Results: Signals higher or lower than >1-fold by log2 were regarded as significant and analyzed further. And only either unregulated or downregulated genes in Brd4-depleted cells compared with control cells at the same time point (0, 0.5 or 2 hr) after UV treatment were analyzed. Indeed, a transcriptome analysis indicated that UV-upregulated genes are positively affected by the Brd4 level.
