Project description:Understanding toxicity pathways of engineered nanomaterials (ENM) has recently been brought forward as a key step in 21st century ENM risk assessment. Molecular mechanisms linked to phenotypic end points is a step towards the development of toxicity tests based on key events, which may allow for grouping of ENM according to their mechanisms of action. This study identified molecular mechanisms underlying mitochondrial dysfunction in human bronchial epithelial BEAS 2B cells following exposure to one of the most studied multi-walled carbon nanotubes (MWCNTs; Mitsui-7). Asbestos was used as a positive control and a non-carcinogenic glass wool material was included as a negative fibre control. Decreased mitochondrial membrane potential (MMP↓) was observed for MWCNTs at a biologically relevant dose (0.25 μg/cm2) and for asbestos at 2 μg/cm2, but not for glass wool. Extensive temporal transcriptomic and microRNA expression analyses identified a 330-gene signature related to MWCNT- and asbestos-induced MMP↓. Fourty-nine of the MMP↓-associated genes showed highly similar expression patterns over time (six time points) and the majority was found to be regulated by two transcription factors strongly involved in mitochondrial homeostasis, APP and NRF1. In addition, four miRNAs were associated with MMP↓ and one of them, miR-1275, was found to negatively correlate with a large part of the MMP↓-associated genes. Cellular processes such as gluconeogenesis, glucose metabolism, mitochondrial LC-fatty acid β-oxidation and spindle microtubule function were enriched among the MMP↓-associated genes and miRNAs. These results are expected to be useful in the identification of key events in ENM-related toxicity pathways for the development of molecular screening techniques. BEAS 2B cells were exposed to 0.25 and 2 μg/cm2 of MWCNT, asbestos and glass wool (MMVF10) for 1, 4, 6, 12, 24 and 48 hours. Three independent experiments were performed to obtain a total of 3 replicates for each experimental condition. Non-exposed cells for each time point were used as controls. Three samples failed to give adequate quality RNA and a total of 123 samples were hybridized to Agilent miRNA arrays.

Project description:Nanomaterials are a relatively new class of materials that acquire novel properties based on their reduced size. While these materials have widespread use in consumer products and industrial applications, the potential health risks associated with exposure to them remain to be fully characterized. Carbon nanotubes are among the most widely used nanomaterials and have high potential for human exposure by inhalation. These nanomaterials are known to penetrate the cell membrane and interact with intracellular molecules, resulting in a multitude of documented effects, including oxidative stress, genotoxicity, impaired metabolism, and apoptosis. While the capacity for carbon nanotubes to damage nuclear DNA has been established, the effect of exposure on mitochondrial DNA (mtDNA) is relatively unexplored. In this study, we investigated the potential of multi-walled carbon nanotubes (MWCNTs) to impair mitochondrial gene expression and function in human bronchial epithelial cells (BECs). Primary BECs were exposed to sub-cytotoxic doses (up to 3 μg/ml) of MWCNTs for 5 d and assessed for changes in expression of all mitochondrial protein-coding genes, heteroplasmies, and insertion/deletion mutations (indels). Exposed cells were also measured for cytotoxicity, metabolic function, mitochondrial abundance, and mitophagy. We found that MWCNTs upregulated mitochondrial gene expression, while significantly decreasing oxygen consumption rate and mitochondrial abundance. Confocal microscopy revealed induction of mitophagy by 2 hours of exposure. Mitochondrial DNA heteroplasmy and insertion/deletion mutations were not significantly affected by any treatment. We conclude that carbon nanotubes cause mitochondrial dysfunction that leads to mitophagy in exposed BECs via a mechanism unrelated to its reported genotoxicity.

Project description:Carbon nanotubes could accumulate in organism and have a negative impact on the structure and function of the ecosystem when they were discharged into environment. Furthermore, it will affect the migration and fate of pollutants in the water body. The study is mainly to explore the adsorption behavior and mechanism of beta-blocker on multi-walled carbon nanotubes (MWCNTs). Propranolol (PRO) was selected as the representative of beta-blocker. The effects of different environmental factors such as pH, ionic strength and humic acid (HA) on the adsorption process were investigated. The adsorption results were characterized by Zeta potential. At the same time, the effects of different types of drugs on the adsorption process were explored and the possible adsorption mechanisms were analyzed. The experimental results showed that the adsorption behavior was significantly different under different pH conditions. ?-? EDA interaction, hydrophobic interaction and hydrogen bonding were speculated to be the main adsorption mechanisms for PRO adsorption on MWCNTs.

Project description:Long carbon nanotubes (CNTs) resemble asbestos fibers due to their high length to diameter ratio and they thus have genotoxic effects. Another parameter that might explain their genotoxic effects is contamination with heavy metal ions. On the other hand, short (1-2 µm) CNTs do not resemble asbestos fibers, and, once purified from contaminations, they might be suitable for medical applications. To identify the role of fiber thickness and surface properties on genotoxicity, well-characterized short pristine and carboxylated single-walled (SCNTs) and multi-walled (MCNTs) CNTs of different diameters were studied for cytotoxicity, the cell's response to oxidative stress (immunoreactivity against hemoxygenase 1 and glutathione levels), and in a hypoxanthine guanine phosphoribosyltransferase (HPRT) assay using V79 chinese hamster fibroblasts and human lung adenocarcinoma A549 cells. DNA repair was demonstrated by measuring immunoreactivity against activated histone H2AX protein. The number of micronuclei as well as the number of multinucleated cells was determined. CNTs acted more cytotoxic in V79 than in A549 cells. Plain and carboxylated thin (<8 nm) SCNTs and MCNTs showed greater cytotoxic potential and carboxylated CNTs showed indication for generating oxidative stress. Multi-walled CNTs did not cause HPRT mutation, micronucleus formation, DNA damage, interference with cell division, and oxidative stress. Carboxylated, but not plain, SCNTs showed indication for in vitro DNA damage according to increase of H2AX-immunoreactive cells and HPRT mutation. Although short CNTs presented a low in vitro genotoxicity, functionalization of short SCNTs can render these particles genotoxic.

Project description:There have been a number of theoretical and experimental studies on tensile properties of carbon nanotubes (CNT), reporting the Young's modulus of the individual CNT up to 1 TPa. Although CNT shows the promise to be used as reinforcement in a high modulus/strength composite material, it exhibits quite disappointing in terms of modulus or strength. Along with recent advance in CNT growth technique, we will be able to directly measure tensile properties of millimeter-long MWCNTs. This study firstly tackles the direct measurement of the tensile properties of millimeter-long MWCNTs that can be used as reinforcement in a composite system. A carefully designed tensile testing technique for the MWCNTs is developed, which allows us to obtain more accurate and reliable measured values. The average tensile strength and Young's modulus of the CNTs investigated in this study are measured to be 0.85?GPa and 34.65?GPa, respectively. Also, this work statistically investigates the effect of the CNT dimensions including length, diameter and volume on the tensile properties. To the best of our knowledge, this is the very first report on the tensile properties of macroscopically long and continuous CNTs.

Project description:The studies focusing on magnesium oxychloride cement (MOC) composites have recently become fairly widespread because of MOC's excellent mechanical properties and environmental sustainability. Numerous fillers, admixtures and nano-dopants were studied in order to improve the overall performance of MOC-based derivatives. Some of them exhibited specific flaws, such as a tendency to aggregate, increase in porosity, aeration of the composite matrix, depreciation in water resistance and mechanical strength, etc. In this manuscript, MOC-based composites doped by multi-walled carbon nanotubes (MWCNTs) are designed and tested. In order to modify the final properties of composites, diatomite was admixed as partial substitution of MgO, which was used in the composition of the researched material in excess, i.e., the majority of MgO constituted part of MOC and the rest served as fine filler. The composites were subjected to the broad experimental campaign that covered SEM (scanning electron microscopy), EDS (energy dispersive spectroscopy), HR-TEM (high-resolution transmission electron microscopy), XRD (X-ray diffraction), OM (optical microscopy) and STA-MS (simultaneous thermal analysis with mass spectroscopy). For 28 days hardened samples, macrostructural and microstructural parameters, mechanical properties, hygric and thermal characteristics were experimentally assessed. The incorporation of MWCNTs and diatomite resulted in the significant enhancement of composites' compactness, mechanical strength and stiffness and reduction in water absorption and rate of water imbibition. The thermal properties of the enriched MOC composites yielded interesting values and provided information for future modification of thermal performance of MOC composites with respect to their specific use in practice, e.g., in passive moderation of indoor climate. The combination of MWCNTs and diatomite represents a valuable modification of the MOC matrix and can be further exploited in the design and development of advanced building materials and components.
