Project description:Sow milk yield and quality is crucial for the survival and growth of piglets. Mammary epithelial cell proliferation, differentiation, milk component production and galactopoietic hormone and cytokine synthesis result from the quantities of differentially expressed genes. This study used the Affymetrix Porcine GeneChip to acquire a comprehensive picture of transcriptional adaptations in the mammary glands of lactating sows. Mammary tissue samples were collected from six sows at -17, 1 and 17 d relative to parturition. Mammary tissues from two sows were used to extract RNA and were subsequently pooled in equal volumes. Nine pooled samples were hybridized to porcine Affymetrix GeneChips.

Project description:Background: While the mechanisms underlying the lactation-induced adaptations of intermediary metabolism and immune response have been extensively studied in rodents and dairy cows, little is known in this regard in sows. Therefore, the present study aimed to explore the lactation-induced changes in hepatic gene expression in sows during lactation. Results: Using a porcine whole-genome microarray a total of 632 differentially expressed genes in the liver of lactating compared to non-lactating sows (each n = 10) could be identified. Enrichment analysis revealed that the differentially expressed genes were mainly involved in fatty acid metabolism, pyruvate metabolism, glutathione metabolism, glycine, serine and threonine metabolism, citrate cycle, glycerophospholipid metabolism, PPAR signaling, nitrogen metabolism, and focal adhesion. The most striking observation with respect to intermediary metabolism was that genes involved in fatty acid catabolism, the catabolism of gluconeogenic amino acids, the citrate cycle and the respiratory chain were up-regulated in the liver of sows during lactation. With respect to immune response, it could be demonstrated that genes encoding acute phase proteins and genes involved in tissue repair were up-regulated and genes encoding focal adhesion molecules were down-regulated in the liver of sows during lactation. Conclusion: The results from this study indicate that energy-generating pathways and pathways involved in the delivery of gluconeogenic substrates are induced in the liver of sows during lactation. The alterations of expression of genes encoding proteins involved in immune response suggest that lactation in sows may cause an adaptive immune response which possibly counteracts hepatic inflammation. For microarray analyses, four RNA pools each, for the non-lactating group (n = 4) and the lactating group (n = 4), were prepared from eight individual sows per group. To each RNA pool, RNA from two different sows per group contributed identical amounts of RNA.

Project description:Epigenetic changes such as DNA methylations regulate gene expression patterns in response to environmental cues including infections. Microbial infections induced DNA methylation may play a potential role in modulating host-immune response. In the present study, we sought to determine DNA methylation changes induced by the mastitis causing Escherichia coli (E coli) in porcine mammary epithelial cells (PMEC). Two time points (3hr and 24 hr) were selected based on the specific transcriptomic changes as early and late phase immune responses. Genome-wide methylation information was obtained to identify significant differential methylation patterns in E coli infected PMEC.

Project description:Mastitis is a multietiological complex disease, which is defined as inflammation of parenchyma of mammary glands. Bacterial infection is the predominant cause of mastitis, though fungal, viral and mycoplasma infections also have been reported. Based on severity of the disease, mastitis can be classified into- subclinical, clinical and chronic forms. Bacterial pathogens from the fresh cow milk were isolated and classified by standard microbiological tests and multiplex PCR. Epidemiological studies have shown that Escherichia coli is the 2nd largest mastitic pathogen after Staphylococcus aureus in India. Based on Enterobacterial Repetitive Intergenic Consensus (ERIC) -PCR profile and presence of virulence genes, a field isolate of E. coli was used for intramammary inoculation in lactating mice. Histopathological examination of H&E stained sections showed severe infiltration of polymorphonuclear neutrophils (PMNs), mononuclear inflammatory cells in the alveolar lumen and also in interstitial space and necrosis of alveolar epithelial cells after 24h. Western blot and immunohistochemical analysis of the mice mammary tissues have shown significant hyperacetylation at histone H3Lys14 residue of both mammary epithelial cells and migrated inflammatory cells. Quantitative real-time PCR and genome-wide gene expression profile in the E. coli infected mice mammary tissue revealed differential expression of genes related to inflammation, immunity, antimicrobial peptide expression, acute phase response and oxidative stress response. Expression levels of milk proteins were also suppressed. ChIP assay from the paraffinized tissues have shown selective enrichment of acetylated histone H3K14 and H4K8 at the promoters of over expressed genes. These data suggests that E. coli infection in the mice mammary tissue leads to histone hyperacetylation at the immune gene promoters which is pre-requisite for expression of inflammatory genes to mount drastic immune response. Abdominal mammary glands of 7 Day post partum lactating female Swiss albino mice was challenged with a pure culture of E. coli isolated from cow milk. Mammary tissues were harvested after 24h of infection. Total RNA was isolated by Qiagen RNeasy mini columns and used for global gene expression and RT-PCR analysis. For RT-PCR, cDNAs corresponding to total mRNAs were synthesized using Oligo-dT primer. Global gene expression analysis were done by single colour labelling of total RNA and hybridization to Agilent 8x60k mouse Microarray.

Project description:Mastitis is a common disease that hinders the development of dairy industry and animal husbandry. It leads to the abuse of antibiotics, the emergence of super drug-resistant bacteria, and poses a great threat to human food health and safety. Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) are the most common pathogens of mastitis in dairy cows and usually cause subclinical or clinical mastitis. CircRNAs and N6-methyladenosine (m6A) play important roles in immunological diseases. However, the mechanisms by which m6A modifies circRNA in bovine mammary epithelial cells remain poorly understood. The aim of our study was to investigate m6A-modified circRNAs in bovine mammary epithelial cells (MAC-T cells) injured by S. aureus and E. coli. The profile of m6A-modified circRNA showed a total of 1599 m6A peaks within 1035 circRNAs in the control group, 35 peaks within 32 circRNAs in the S. aureus group, and 1016 peaks within 728 circRNAs in the E. coli group. Compared with the control group, 67 peaks within 63 circRNAs were significantly different in the S. aureus group, and 192 peaks within 137 circRNAs were significantly different in the E. coli group. Furthermore, we found the source genes of these differentially m6A-modified circRNAs in the S. aureus and E. coli groups with similar functions according to GO and KEGG analyses, which were mainly associated with cells injury, such as inflammation, apoptosis, and autophagy. CircRNA-miRNA-mRNA interaction networks predicted the potential circRNA regulation mechanism in S. aureus- and E. coli-induced cell injury. We found that the mRNAs in the networks, such as BCL2, MIF and TNFAIP8L2, greatly participated in the MAPK, WNT, and inflammation pathways. This is the first report on m6A-modified circRNA regulation of cells under S. aureus and E. coli treatment, and sheds new light on potential mechanisms and targets from the perspective of epigenetic modification in mastitis and other inflammatory diseases.
