Anaimals were alowed to recover for 6 hours before brains were removed for RNA extraction and hybridization on Affymetrix microarrays to obtain expression profiles from which complex networks could be generated

General anaesthesia leads to phosphoproteome changes in proteins involved in cytoskeleton remodelling and synaptic processes in the supraoptic nucleus of the hypothalamus

Project description:Despite the widespread use of general anaesthetics, the mechanisms mediating their effects are still not understood. Although suppressed in most parts of the brain, neuronal activity, as measured by FOS activation, is increased in the hypothalamic supraoptic nucleus (SON) by numerous general anaesthetics, and evidence points to this brain region being involved in the induction of general anaesthesia and natural sleep. Posttranslational modifications of proteins, including changes in phosphorylation, enable fast modulation of protein function which could be underlying the rapid effects of general anaesthesia. In order to identify potential phosphorylation events in the brain mediating general anaesthesia effects, we have explored the phosphoproteome responses in the rat SON, and compared these to cingulate cortex (CC) which displays no FOS activation is response to general anaesthetics. We found many changes in the phosphoproteomes of both the CC and SON in response to 15 minutes of isoflurane exposure. Pathway analysis indicated that proteins undergoing phosphorylation adaptations are involved in cytoskeleton remodelling and synaptic signalling events. Importantly, changes in protein phosphorylation appeared to be brain region-specific suggesting that differential phosphorylation adaptations might underlie the different neuronal activity responses to general anaesthesia between the CC and SON. In summary, these data suggest that rapid posttranslational modifications in proteins involved in cytoskeleton remodelling and synaptic signalling events might mediate the central mechanisms mediating general anaesthesia.

Targeting mitochondrial metabolism for detection and treatment of doxorubicin-induced heart failure

Project description:We conducted RNAseq using mRNA extracted from rat hearts to elucidate the effect of doxorubicin chemotherapy on the heart. Rats were 250g at the start of the treatment protocol. Tissues were collected post five weekly intravenous injections of either sterile saline or 3mg/kg/week doxorubicin. Hearts were excised under deep isoflurane anaesthesia and rapidly frozen with liquid-nitrogen cooled Wallenberger tongs. Total mRNA was extracted with a Qiagen RNeasy fibrous tissue mini kit. Gene set enrichement analysis (GSEA) shows that unlike previously reported, oxidative stress is not involved in the cardiac pathology of our model (reduced cardiac function and atrophy). However, gene sets responsible for mRNA processing, ribosomes and protein synthesis and processing are decreased in doxorubicin-treated rat hearts compared to saline control hearts.

Transcriptional effects of a 6h treatment with 2% isoflurane in the anterior cingulate cortex of the 24h old neonatal piglet brain

Project description:Exposure of the developing brain to general anesthesia during early infancy may adversely affect its neural and cognitive development. In a range of animal species early developmental exposure to general anaesthesia without surgery, induces an increase in brain cell death, reductions in neuro and synapto-genesis, a disruption to the expression of cognitively salient genes and deficits in cognitive function that persist throughout life. We examined the effects of the anesthetic isoflurane (2% concentration for 6h) without surgery on gene expression in the anterior cingulate cortex of the neonatal male piglet brain. The developing piglet brain demonstrates remarkable neurodevelopmental similarities to that seen in the human infant, which has led to extensive use as models of perinatal neural injury. Isoflurane induced significant disruption to the expression of 79 gene transcripts, of these 26 are important for the control of transcription and 23 are important for the mediation of neural plasticity, memory formation and recall. Overall this data may suggest novel additional mechanisms by which isoflurane may induce adverse neural and cognitive development by disrupting the expression of genes mediating activity dependent development of neural circuits, the predictive adaptive responses of the brain, memory formation and recall.

Project description:Environmental stressors such as repeated social defeat may trigger powerful activation of sub-conscious parts of the brain. We examine the consequences of such stress in male rats on the pituitary gland. We prepared 20 Sprague Dawley rats. 10 of them were exposed to stress induced by the resident-intruder paradigm while the other 10 were controls. After dislocation of the neck under isoflurane anaesthesia, the pituitary gland was harvested. Total RNA was isolated from the tissues and used in mRNA sequencing.

We prepared 10 Sprague Dawley rats. 5 of them were exposed to stress induced by the resident-intruder paradigm while the other 5 were controls.After dislocation of the neck under isoflurane anaesthesia, the pituitary gland was harvested. To depict an initial epigenetic pituitary stress response, total DNA was isolated from the tissues and used for bisulfite DNA sequencing. This data presented here is a subset of the samples involved in E-MTAB-11285 RNA-seq project.
