Project description:In the complex lifecycle of schistosomes, four developmental stages are closely associated with their definitive hosts: cercaria (infective stage), schistosomula and adult worm (parasitic stages), egg (pathogenic- and pathophoresis-stage). We have examined the gene expression profiles of Schistosoma japonicum in the four developmental stages. Genes with different expression patterns were identified and the information obtained will help indentify new anti-schistosomal intervention targets in the future.

Project description:The body’s defense against schistosome infection can take many forms. For example, upon developing acute schistosomiasis, patients often have fever coinciding with larval maturation, migration and early oviposition. As the infection becomes established, the parasite comes under oxidative stress generated by the host immune system. The most common treatment for schistosomiasis is the anti-helmenthic drug praziquantel. Its effectiveness, however, is limited due to its inability to kill schistosomes 2 - 4 weeks post-infection. Clearly there is a need for new anti-schistosomal drugs. We hypothesize that gene products expressed as part of a protective response against heat and/or oxidative stress are potential therapeutic targets for future drug development. Using a 12,166 element oligonucleotide microarray to characterize Schistosoma mansoni genes induced by heat and oxidative stress we found that 1,878 Schistosoma mansoni elements were significantly induced by heat stress. These included previously reported heat-shock genes expressing homologs of HSP40, HSP70 and HSP86. One thousand and one elements were induced by oxidative stress including those expressing homologs of superoxide dismutase, glutathione peroxidase and aldehyde dehydrogenase. Seventy-two elements were common to both stressors that could potentially be exploited in the development of novel anti-schistosomal therapeutics. Adult female Schistosomes produce eggs of which approximately 40% fail to pass into faeces or urine (species-dependent) but are dispersed by the blood stream into different organs where they provoke severe inflammation. This parasitic infection is known as schistosomiasis and considered by the WHO as the second most socioeconomically devastating parasitic disease, next only to malaria, with hundreds of millions infected worldwide. As the eggs represent the causative pathogenic agents, the understanding of egg forming processes, and therefore of the schistosomal reproductive biology in general, is of fundamental interest. Additionally the differences between immature (pre-mating) and mature (post-mating) adult worms are of significant interest; adults only reach maturity upon mating. Trasncriptomic sequencing of gonad-specific cellular material will help to unravel signal transduction cascades involved in e.g. gametogenesis and/or vitellogenesis. This project aims to characterize the transcriptome profiles of immature and mature adult ovaries and adult testes.

As a result there is an urgent need to develop a new generation of anti-schistosomal drugs, a task that will be made easier by understanding the mechanism of action of PZQ. As yet, neither the molecule to which PZQ binds nor the means by which it kills mature schistosomes is known. The overarching aim of this study was to understand the molecular basis of PZQ sensitivity in S. mansoni. We believe that juvenile worms survive PZQ treatment in vivo due to the induction of, as yet, unidentified protective molecular pathways. To address this hypothesis juvenile and adult PR1 S. mansoni were treated in vitro with sub-lethal concentrations of PZQ. mRNA was extracted from replicate samples, cRNA prepagreen and labeled with cyanine dyes for analysis using a 44K S. mansoni microarray. The data was then analyzed using Genespring. Our findings suggest that a number of genes associated with drug transport, iron homeostasis and apoptosis are induced in juvenile but not adult schistosomes and that this allows the juvenile worms to protect themselves against the lethal effects of PZQ long enough for the drug to be metabolized by the human host.

All experiments were performed in biological triplicates. The 0 µg/mL PZQ exposures were used as controls to the appropriate groups and a universal reference was used in all experiments that was comprised of RNA derived from 100 mixed sex, unexposed 42 DPE schistosomes spiked with 0.5 % (v/v) RNA isolated from 42 DPE mixed sex worms exposed to 10 µg/mL PZQ, 0.5 % (v/v) RNA isolated from 28 DPE worms exposed to 10 µg/mL PZQ and 0.5 % (v/v) RNA isolated from 28 DPE worms unexposed to PZQ. Project description:Schistosomiasis is one of the most socioeconomically harmful neglected tropical diseases in the world. It occurs following infection from parasites of the Schistosoma genus, such as Schistosoma mansoni, which must transition within a molluscan and mammalian host to survive. Previous chemical analyses of schistosome-molluscan interactions indicate that schistosomes orientate towards potential hosts partially through chemosensation, displaying a preference for naïve (uninfected) hosts. Recent advances in proteomic techniques enable sophisticated comparative analyses between infected and naïve snail host proteins. This study aimed to compare the snail-conditioned water (SCW) released by F1 resistant, infected and naïve Biomphalaria glabrata to identify potential attractants and deterrents.

Project description:Schistosomiasis is a chronic parasitic disease that affects more that 230 million people worldwide. In the Americas, it caused by the blood flook Schistosoma mansoni. This parasite has a complex life cycle with two intermediary hosts. When infecting mammals, schistosomes cause an intense immune response to the presence of the different stages of the parasite within the host. After maturation of the worms and commencing of the oviposition, some eggs are transported via circulation to the liver, where they elicit an exaggerated inflammatory response in the tissue. This could lead to the formation of granulomas and liver fibrosis and along with this, the developent of hepatosplenomegaly and in the worst scenario could lead to the dead of the patient. In this project, we aim to make a shotgun evaluation of the proteome from splenocytes in a mice model, in order to identify novel molecular signatures associated with the immune respose to the presence of the parasite in the hosts.

Project description:Background: Background: In bovines, the development and growth of adipose involves many physiological processes and plays an extremely important role in the quality of beef, however, the regulatory mechanisms underlying the differences in meat quality are largely unknown. Therefore, a buffalo adipose transcriptome analysis was performed to compare gene expression profiles between different growth stages. Results: Finally, a total of 21,693 mRNAs were obtained, of which 2506 were significantly differentially expressed mRNAs in the two groups, 9494 lncRNAs, a total of 281 lncRNAs and 2506 mRNAs showed significant differential expression patterns and 14990 between the two buffaloes CircRNA, 252 circRNAs were significantly differentially expressed between young and adult buffalo.

Project description:Schistosomiasis (Bilharziasis) is caused by the helminth Schistosoma. The acute stage of the infection extends from the penetration of the parasite through the host skin until it reaches its final location as adult female and male in copula, at the blood vessels around the gut or bladder. The female starts then depositing fertile eggs. The chronic stage of schistosomiasis starts with laying the eggs which promote a strong immunological Th2 response. It is unclear, so far, how this Th2 response gradually declines even though the parasitic worms live for years and continue to produce eggs. Here we demonstrate that schistosomes downregulate the differentiation toward the Th2 lineage by modulating the Th2 transcriptional program. Moreover, we found that adult schistosomes secrete micro-vesicles, possessing exosomal characteristics, and containing miRNAs, which have been found in Th2 cells that exposed indirectly to schistosomes. Importantly, we demonstrate that, the schistosoma miR-10 molecule targets MAP3K7, which consequently down modulates NF-kB. Since NF-kB is a critical factor necessary for Th2 differentiation and function, our results can at least partially explain, the decrease of the Th2 response over time of infection This exosomal worm-host immune cell interaction, can be further utilized for the development of novel diagnostic and therapeutic approaches to schistosomiasis.
