ABSTRACT: To investigate effects of intake of mulberry leaf extracts on hypercholesterolemia, we performed gene expression profiling on rat liver by microarray analysis. Microarray analysis revealed that mulberry leaf extracts up-regulated the gene expression involved in suppression of cholesterol synthesis and stimulation of innate-adaptive Immunity.

Project description:To investigate effects of intake of mulberry leaf extracts on hypercholesterolemia, we performed gene expression profiling on rat liver by microarray analysis. Microarray analysis revealed that mulberry leaf extracts up-regulated the gene expression involved in suppression of cholesterol synthesis and stimulation of innate-adaptive Immunity. Mice were fed a high-cholesterol diet without/with orally administration of mulberry leaf extracts for 4 weeks. Livers were taken for RNA extraction and hybridization on Agilent microarrays.

Identification and Comparative Analysis of Drought Responsive microRNA Targets in Mulberry (Morus alba L.) by Degradome Sequencing

Project description:MicroRNAs (miRNAs) are endogenous non-coding small RNAs containing 21-24 nucleotides, and play an important function in the course of stand up to adversity. Identification of miRNA target mRNAs is essential for their functional analysis. In order to anatomize miRNA guided gene regulation under drought stress, we performed a transcriptome-wide experimental method using high throughput degradome sequencing to directly detect drought stress responsive miRNA targets in mulberry. In this study, drought library (DL) and contrast library (CL) which captured the cleaved mRNA were constructed from mulberry for sequencing by Illumina sequencer, and 409, 373 target genes for 30 conserved miRNA families and 990, 950 target genes for 199, 195 novel miRNAs were identified in CL and DL, respectively. Among those conserved miRNA families in DL, mno-miR156, mno-miR172 and mno-miR396 refer to the highest number of targets, indicating that mno-miR156, mno-miR172 and mno-miR396 families and their target genes might play a key important function in corresponding to drought stress in mulberry. By construction and analysis of miRNA-target network, we found that the DL independent networks consist of 838 miRNA-mRNA pairs (63.34%). Besides, the expression patterns of 11 target genes and 12 correspondent miRNAs were compared and analyzed by qRT-PCR. In addition, 5 of 6 miRNA targets were further verified by RNA ligase-mediated 5’ rapid amplification of cDNA ends (RLM-5’ RACE). Gene ontology (GO) annotations and Kyoto Encyclopaedia of Genes and Genomes (KEGG) pathway analysis showed that these target transcripts were implicated in a broad range of biological processes and various metabolic pathways. The characterization of target genes and the associated miRNAs in response to drought exposure provides a framework for understanding the molecular mechanism of resistance to drought in plants.

RNA-seq-based transcriptome profiling of hawk tea extract (HTE)-fed hypercholesterolemic rat livers

Project description:The high-fat diet (HFD)-feeding significantly stimulated hypercholesterolemia in male SD rats. Simultaneous feeding of hawk tea extracts (HTE) reversed the hypercholesterolemia in rats by lowering the serum total cholesterol and low-density lipoprotein cholesterol levels. To understand the molecular mechanism underlying the cholesterol-lowering effects of HTE, we performed the RNA-seq analysis. Consequently, the change in mRNA levels of genes invovled in metabolism of lipid and lipoprotein, primary bile acid synthesis, and ABC transporters itriggered by HFD-feeding were reversed by the co-administration of HTE (200 mg/kg/day for 11 weeks).

White mulberry (Morus alba) foliage methanolic extract can alleviate Aeromonas hydrophila infection in African catfish (Clarias gariepinus).

Project description:Two experiments were simultaneously conducted with Morus alba (white mulberry) foliage extract (MFE) as a growth promoter and treatment of Aeromonas hydrophila infection in separate 60 and 30 days trail (Experiments 1 and 2, resp.) in African catfish (Clarias gariepinus). In Experiment 1, four diets, control and control supplemented with 2, 5, or 7 g MFE/kg dry matter (DM) of diet, were used. In Experiment 2, fish were intraperitoneally infected with Aeromonas hydrophila and fed the same diets as experiment 1 plus additional two diets with or without antibiotic. Results of experiment 1 showed that growth was unaffected by dietary levels of MFE. Treatments with the inclusion of MFE at the levels of 5 and 7 g/Kg DM had no mortality. Red blood cells (RBC), albumin, and total protein were all higher for the treatments fed MFE (5 and 7 g/Kg DM). Results of experiment 2 showed RBC, hemoglobin, hematocrit, globulin, albumin, and total protein improved with the increase in MFE in the infected fish. The dietary MFE at the level of 7 g/kg DM reduced mortality rate. In conclusion, MFE at the level of 7 g/kg DM could be a valuable dietary supplement to cure the infected fish.

Prokinetic Activity of Mulberry Fruit, <i>Morus alba</i> L.

Project description:The fruit of Morus alba L. (MAF) has been consumed as a food worldwide. MAF has also been widely used in traditional medicine for thousands of years in East Asia, and its diverse bioactivities have been reported in numerous publications. However, no prokinetic activity has been reported for MAF or its components. In the present study, therefore, we investigated the effects of MAF on gastrointestinal motor function by measuring the intestinal transit rate (ITR) of Evans blue in mice in vivo. The ITR values accelerated by MAF were significantly higher than those accelerated by cisapride or metoclopramide, suggesting that MAF has potential as a new prokinetic agent to replace cisapride and metoclopramide. We also investigated the effects of MAF on myogenic and neurogenic contractions in human intestinal smooth muscles by measuring spontaneous contractions of smooth muscle strips, smooth muscle contractions induced by neural stimulation, and migrating motor complexes from intestinal segments in the human ileum and sigmoid colon in situ. MAF increased both myogenic and neurogenic contractions to enhance ileal and colonic motility in the human intestine. Taken together, these results indicate that MAF enhanced intestinal motility by increasing both myogenic and neurogenic contractions, thereby accelerating the ITR.

Drought stress was imposed at whole plant leve; usig gravimetric approach. Leaf tissue was harvested 14 days post drought stress imposition and used for transcriptome analaysis. Two levels of drought stress (100% and 40% soil field capacity) was maintained by controlled irrigation. The leaf tissue was collected, total RNA was isolated converted to cDNA and used for analysis
