Project description:Lavender oil (LO) is commonly used oil in aromatherapy, with well-defined volatile components linalool and linalyl acetate, in non-traditional medicine. To understand and demonstrate the potential positive effects of LO on the body, we have established an animal model for investigating the orally administered LO effects genome-wide in the rat tissues. In this data submission, we investigate the effect of LO ingestion in the blood. These results are the first such inventory of genes that are affected by lavender essential oil (LO) in the blood of an animal model, forming the basis for further functional analysis and investigation. Briefly, rats were administered LO at 5 mg/kg (usual therapeutic dose in humans) followed by screening of differentially expressed genes in the tissues, using a 4×44-K whole-genome rat chip (Agilent microarray platform; Agilent Technologies, Palo Alto, CA, USA) in conjunction with a dye-swap approach.

Reverse transcription-polymerase chain reaction (RT-PCR) validation of highly up- and down-expressed genes revealed the regulation of Papd4, Lrp1b, Alb, Cyr61, Cyp2c, and Cxcl1 genes, by LO, as examples in these tissues. Using bioinformatics functionally categorization of differentially expressed genes was done by their Gene Ontology (GO) revealing their diverse functions and potential roles in LO-mediated effects in rat. Present results are a first such inventory of genes affected by the essential oil of Lavender (LO) in an animal model. Project description:Lavender oil (LO) is a commonly used essential oil in aromatherapy as non-traditional medicine. With an aim to demonstrate LO effects on the body, we have recently established an animal model investigating the influence of orally administered LO in rat tissues, genome-wide. In this brief, we investigate the effect of LO ingestion in the blood of rat. Rats were administered LO at usual therapeutic dose (5 mg/kg) in humans, and following collection of the venous blood from the heart and extraction of total RNA, the differentially expressed genes were screened using a 4 × 44-K whole-genome rat chip (Agilent microarray platform; Agilent Technologies, Palo Alto, CA, USA) in conjunction with a two-color dye-swap approach. A total of 834 differentially expressed genes in the blood were identified: 362 up-regulated and 472 down-regulated. These genes were functionally categorized using bioinformatics tools. The gene expression inventory of rat blood transcriptome under LO, a first report, has been deposited into the Gene Expression Omnibus (GEO): GSE67499. The data will be a valuable resource in examining the effects of natural products, and which could also serve as a human model for further functional analysis and investigation.

Project description:Terpene volatiles play an important role in the interactions among specialized pathogens and fruits. Citrus Black Spot (CBS), caused by the fungus Phyllosticta citricarpa, is associated with losses in different citrus-growing areas worldwide. The pathogen may infect the fruit for 20-24 weeks after petal fall but the typical hard spot symptoms appear when the fruit become almost mature, as consequence of fungal colonization and induction of cell lyses around essential oil cavities. D-limonene represents about 95% of total oil gland content in mature orange fruit. We have used here d-limonene synthase downregulated orange fruit generated via an antisense (AS) approach to investigate whether d-limonene decrease content in peel oil glands may affect fruit interaction with P. citricarpa compared to that of empty-vector (EV) controls. AS fruit showed enhanced resistance to the fungus compared to EV ones. Because of d-limonene decreased content, an over-accumulation of linalool and other monoterpene alcohols was found in AS compared to EV fruit. A global gene expression analysis at 2h and 8d after inoculation with P. citricarpa revealed activation of defence responses in AS fruit, via the upregulation of different PR genes, likely due to the enhanced constitutive accumulation of linalool and other alcohols. When assayed in vitro and in vivo, the main altered terpene volatile compounds including linalool at concentrations emulating those present in AS fruit, showed strong antifungal activity. We show here that terpene engineering in fruit peels could be a promising method for developing new strategies to obtain resistance to fruit diseases.

The Forl strain was cultured on PDA supplied with 0.8 mL/L linalool for 6 days at 25°C. The fungal strain on PDA supplied with only 0.1% Tween80 was cultured as the control. However, it was not natural to think that full-length cDNA libraries was included almost rice genes, because the transcription of 3K CDS-ANE were confirmed by other transcriptome resources in TIGR OSA1. So, for detection of gene expression from FL-locus and TIGR-locus, we designed about 82K 60mer oligo probes based on TIGR Pseudomolecules release 2. However, TIGR-locus was revised according to revised Pseudomolecules and other additional transcriptome resources.
