ABSTRACT: The goal of this study was to compare RNA Seq profiles of Asian longhorned beetle larval guts feeding on two different substrates to identify key genes involved in woody tissue degradation, nutrient scavenging, and detoxification of host plant defensive compounds.

Firefly genomes illuminate parallel origins of bioluminescence in beetles

Project description:Fireflies and their fascinating luminous courtships have inspired centuries of scientific study. Today firefly luciferase is widely used in biotechnology, but the evolutionary origin of their bioluminescence remains unclear. To shed light on this long-standing question, we sequenced the genomes of two firefly species that diverged over 100 million-years-ago: the North American Photinus pyralis and Japanese Aquatica lateralis. To compare bioluminescent origins, we also sequenced the genome of a related click-beetle, the Caribbean Ignelater luminosus, with bioluminescent biochemistry near-identical to fireflies, but anatomically unique light organs, suggesting the intriguing but contentious hypothesis of parallel gains of bioluminescence. Our analyses support two independent gains of bioluminescence between fireflies and click-beetles, and provide new insights into the genes, chemical defenses, and symbionts that evolved alongside their luminous lifestyle.

Transcriptional Profiling of Midguts Prepared from Trypanosoma/T. congolense-Positive Glossina palpalis palpalis Collected from Two Distinct Cameroonian Foci: Coordinated Signatures of the Midguts' Remodeling As T. congolense-Supportive Niches.

Project description:Our previous transcriptomic analysis of Glossina palpalis gambiensis experimentally infected or not with Trypanosoma brucei gambiense aimed to detect differentially expressed genes (DEGs) associated with infection. Specifically, we selected candidate genes governing tsetse fly vector competence that could be used in the context of an anti-vector strategy, to control human and/or animal trypanosomiasis. The present study aimed to verify whether gene expression in field tsetse flies (G. p. palpalis) is modified in response to natural infection by trypanosomes (T. congolense), as reported when insectary-raised flies (G. p. gambiensis) are experimentally infected with T. b. gambiense. This was achieved using the RNA-seq approach, which identified 524 DEGs in infected vs. non-infected tsetse flies, including 285 downregulated genes and 239 upregulated genes (identified using DESeq2). Several of these genes were highly differentially expressed, with log2 fold change values in the vicinity of either +40 or -40. Downregulated genes were primarily involved in transcription/translation processes, whereas encoded upregulated genes governed amino acid and nucleotide biosynthesis pathways. The BioCyc metabolic pathways associated with infection also revealed that downregulated genes were mainly involved in fly immunity processes. Importantly, our study demonstrates that data on the molecular cross-talk between the host and the parasite (as well as the always present fly microbiome) recorded from an experimental biological model has a counterpart in field flies, which in turn validates the use of experimental host/parasite couples.

Project description:Huanglongbing, or citrus greening disease, has devastated the citrus industry. It is associated with the gram negative bacterium Candidatus Liberibacter asiaticus (CLas) that can be transmitted by Diaphorina citri, the Asian citrus psyllid. For transmission to occur, CLas must cross the gut of the ACP to circulate through the insect body. The insect gut is the first site of widespread interactions between the CLas and the ACP and forms a barrier to transmission. To investigate the effect of CLas exposure on this dynamic interface, we performed RNAseq and mass spectrometry-based proteomics to analyze the transcriptome and proteome respectively of dissected ACP guts. We found changes in iron metabolism, insecticide resistance, immune system, and apoptosis. We identified 83 long non-coding RNAs that are responsive to CLas, two of which have no homology to other organisms in NCBI. We also determined that Wolbachia, a symbiont of the ACP, undergoes protein regulation when CLas is present. Fluorescent in situ hybridization (FISH) confirmed that Wolbachia and CLas can inhabit the same ACP gut cell, but do not co-localize. These data provide a snapshot of the ACP gut under normal and CLas-exposed conditions, and provide tools to better understand the insect vector of the citrus greening pathosystem.

Transcriptomic assessment of interactions between doublesex and its target genes across sexes and tissues in horned beetles

Project description:Purpose: Sexual dimorphisms fuel significant intraspecific variation and evolutionary diversification. Yet the developmental-genetic mechanisms underlying sex-specific development remain poorly understood. We focused on the conserved sex-determination gene doublesex (dsx) and the mechanisms by which it mediates sex-specific development in a species of horned beetle by combining systemic dsx knockdown, high-throughput sequencing of diverse tissues, and a genome-wide analysis of Dsx binding sites. We find that Dsx regulates sex-biased expression predominantly in males, that Dsx's target repertoires are highly sex- and tissue-specific, and that Dsx can exercise its regulatory role via two distinct mechanisms: as a sex-specific modulator by regulating strictly sex-specific targets, or as a switch by regulating the same genes in males and females in opposite directions. More generally, our results suggest Dsx can rapidly acquire new target gene repertoires to accommodate evolutionarily novel traits, evidenced by the large and unique repertoire identified in head horns, a recent morphological innovation.
