Effect of Smyd1 conditional knockout on gene expression in skeletal muscle

ABSTRACT: Transcriptome analysis by RNA-seq of tibialis anterior muscle from control and Smyd1 myocyte-specific conditional knockout mice at 6 weeks of age. Smyd1 is a methyltransferase specifically expressed in striated muscle and CD8+ T cells. Smyd1 deficiency resulted in centronuclear myopathy primarily affecting fast-twitch muscle fibers. These results provide insight into how loss of Smyd1 altered transcriptional programs resulting in centronuclear myopathy.

Drosophila temperature sensitive col4a1 mutant (DTS-3) and control (OreR) comparison at permissive and at restrictive temperature

Project description:We have recently reported an allelic series of dominant, conditional, temperature-sensitive (DTS) mutations in the type IV collagen gene col4a1 in Drosophila and the onset of severe myopathy by massive degradation of striated muscle fibers, degeneration of epithelial and circular smooth muscle cells of the gut. In order to get a closer look at the molecular causes of the observed phenomena we have executed microarray experiments with the aforementioned mutant. Two growth temperatures for both DTS-3 and OreR flies with dye-swap technical control replicates.

Mitochondrial dynamics define muscle fiber type by modulating cellular metabolism pathways

Project description:Skeletal muscle is highly developed after birth, consisting of glycolytic fast- and oxidative slow-twitch fibers; however, the mechanisms of fiber type-specific differentiation are poorly understood. Here, we found an unexpected role for mitochondrial fission in the differentiation of fast-twitch oxidative fibers. Depletion of the mitochondrial fission factor dynamin-related protein 1 (Drp1) in mouse skeletal muscle and cultured myotubes resulted in the specific reduction of fast-twitch muscle fibers independently of respiratory function. Altered mitochondrial fission caused the activation of the Akt/mammalian target of rapamycin (mTOR) pathway via the mitochondrial accumulation of mTOR complex 2 (mTORC2), and rapamycin administration rescued the reduction of fast-twitch fibers in vivo and in vitro. Under Akt/mTOR activation, the mitochondria-related cytokine growth differentiation factor-15 was upregulated, which repressed fast-twitch fiber differentiation. Our findings reveal a novel role for mitochondrial dynamics in the activation of mTORC2 on mitochondria, resulting in the differentiation of muscle fibers.

5'RNA-seq analysis of soleus, tibialis anterior (TA), diaphragm and left ventricle myocardial tissue from adult wild-type mice.

Project description:We applied a 5'RNA-seq methodology to assess gene and differential isoform expression in striated muscle tissues extracted from adult wild-type mice. 5'RNA-seq analysis of transcriptomes from mouse soleus, tibialis anterior (TA), diaphragm and left ventricle myocardial tissues. Three biological replicates per tissue were pooled into a single sequencing run. 5'RNA-seq methodology consists of enhanced sequencing of 5' ends and computational assessment of changes at start-sites of genes.

Transcription profiling of mouse Hexose-6-phosphate dehydrogenase knockout muscle at 4 weeks

Project description:Hexose-6-phosphate dehydrogenase (H6PD)is the initial component of a pentose phosphate pathway inside the endoplasmic reticulum (ER) that generates NADPH for ER enzymes. In liver, H6PD is required for the 11-oxoreductase activity of 11ss-hydroxysteroid dehydrogenase type 1 (11ss-HSD1), which converts inactive 11-oxo glucocorticoids to their active 11-hydroxyl counterparts; consequently, H6PD null mice are relatively insensitive to glucocorticoids, exhibiting fasting hypoglycemia, increased insulin sensitivity despite elevated circulating levels of corticosterone, and increased basal and insulin-stimulated glucose uptake in muscles normally enriched in Type II (fast) fibers which have increased glycogen content. They also display a progressive vacuolar myopathy evident after 4 weeks of age. We carried out microarray analysis on TA and soleus muscles from 4 week old WT and KO mice to determine an expression profile predicting myopathy. Experiment Overall Design: 4 week old mice are weaned and do not display overt histological evidence of myopathy. Soleus and tibialis anterior are used as comparison groups as they have distinct fibre type content and differing metabolic properties. 3 biological replicates are used for each genotpye and sample type.
