ABSTRACT: Male patients (n=6, mean age 62 years) with NYHA III-IV and an left ventricular ejection fraction of <35% despite pharmacological therapy received 35 hours of enhanced external counterpulsation (EECP) over a period of 7 weeks. Before and after treatment, lateral vastus muscle biopsies were obtained and skeletal muscle gene expression was evaluated using the Affymetrix HuGene 1.0 platform.

Caloric restriction rejuvenates myofiber growth in heart failure

Project description:Heart failure with preserved ejection fraction (HFpEF) is characterised by a distinct but poorly understood in skeletal muscle pathology. In a rat model ZSF1, we identify impaired myonuclear accretion as a mechanism for ablated myofiber growth in HFpEF following resistance exercise intervention. Muscle vascular or mitochondrial dysfunction, or impaired protein synthesis signalling, do not appear as primary limiting mechanisms, and cardiac therapies do not attenuate skeletal muscle pathology. However, we identify that acute caloric restriction rejuvenates myofiber growth in HFpEF during resistance exercise intervention.

Gene expression profiling reveals potential prognostic biomarkers associated with the progression of heart failure

Project description:Heart failure (HF) is the most common cause of morbidity and mortality in the developed countries, especially considering the present demographic tendencies in those populations. We identified biologically relevant transcripts that are significantly altered in the early phase of myocardial infarction (MI) and are associated with the development of post-myocardial infarction HF. We collected peripheral blood samples from patients (n=111) with ST-segment elevation myocardial infarction (STEMI) at four time points (admission, discharge, 1 month after MI, and 6 months after MI). Control group comprised patients (n=46) with a stable coronary artery disease and without a history of myocardial infarction. Affymetrix HuGene 1.0 ST arrays were used to analyze mRNA levels in periperal blood mononuclear cells (PBMCs) isolated from the study and control groups. Samples from the first three time points were compared with the samples from the same patients collected 6 months after MI (stable phase) and with the control group. Additionaly, based on plasma NT-proBNP level and left ventricular ejection fraction parameters the STEMI patients were divided into HF and non-HF groups.We attempted to identify transcripts whose differential expression on the 1st day of myocardial infarction predicted which patients would develop symptoms of HF during the 6 months of follow-up. For this purpose, we compared the microarray results for samples collected on admission for the HF group versus the non-HF group.

Project description:Females with heart failure and reduced ejection fraction (HFrEF) have greater physical limitations and a lower quality of life compared to males, however, the role of non-cardiac mechanisms remains poorly resolved. We hypothesized that differences in skeletal muscle pathology between males and females with HFrEF may explain clinical heterogeneity. In this study, we performed an unbiased RNA sequencing of 5 male controls, 5 female controls, 6 male HFrEF patients, and 5 female HFrEF patients.

Divergent immunometabolic changes in adipose tissue and skeletal muscle with ageing in healthy humans

Project description:RNA-sequencing from 12 young (20-35 years old) and 12 older (60-85 years old) participants' skeletal muscle and adipose tissue. Biopsies were taken at rest following an overnight fast, and following alcohol/caffeine restriction for 24 hours, and no strenuous physical activity for 48 hours. Skeletal muscle was taken by the Bergstrom technique under local anaesthesia (Lidocaine hydrochloride) from the dominant leg Vastus Lateralis, and adipose tissue was taken by needle aspiration 5cm lateral to the umbilicus.

Project description:10 male subjects performed ~45 min one-legged cycling and 4 x 7 maximal concentric-eccentric knee extensions for each leg 15 min later. Thus, one limb performed aerobic and resistance exercise (AE+RE), while the opposing leg did resistance exercise only (RE). Biopsies were obtained from m. vastus lateralis of each leg 3 h after the resistance exercise bout. Gene expression analysis was carried out on the Affymetrix HuGene-2.1-st platform.
