Expression data from Huh7 cells after 1,2,3,4,6-penta-O-galloyl-beta-D-glucopyranoside (PGG) treatment

ABSTRACT: PGG is a natural product exhibits anti-cancer effects on different type of cancers including liver cancer cell lines. In present study we identified a set of genes affected by PGG treatment in Huh7 cells. Some of them, for example, control cell cycle and cell proliferation. We used microarrays to delineate the mechnism of action of PGG on GNMT expression in HCC. We identified distinct classes of up-regulated and down-regulated genes after PGG treatment in Huh7 cells.

Project description:To delineate the transcriptomic changes induced by AD80 relative to sorafenib, we performed RNA-sequencing (RNA-seq) on HUH7 and THLE5B cells treated for 24 hours with either DMSO, sorafenib, or AD80 near respective growth inhibitory GI50 values. HUH7 cells were plated in 6 well plates, treated for 24 hrs with 0.1% DMSO, 5 µM sorafenib, and 50 nM AD80. Cells were collected in “RNAlater”, a RNA storage buffer, and RNA was collected same day using the Qiagen RNeasy mini kit.

Expression Profiling in Selenite-induced Proliferation of HuH7 cells

Project description:Experimental studies confirmed that seleium induced a global change of gene expression in HuH7 cells. Several novally induced secretory growth factors may contribute to the autocrine mode of growth elicited by selenium treatment. Keywords: Treatment HuH7 were cultured in 0.1 Î¼M sodium selenite-containing medium for 15 passages (SELT) or in serum-free medium for 30 h (SF30) or in the regular complete medium containing 10 % fetal calf serum (HUH7) for 30 h. The total RNA were reverse transcried and hybridized to Human 8K-1 (Egenomix Technology Corp., Taiwan). The hybridization time and temperature was 16 h and 62â??, respectively. SELT and HUH7 samples were labeled with Cy5 while SF30 sample was labeled with Cy3. Local background correction and interchip global normalization were accomplished by GenePix Pro 5.0 (Molecular Devices, Sunnyvale, CA ). Genes with differential up- or down-regulation by 1.5 fold with respect to the SF30 cells were obtained using GeneSpring software 7.0 (Silicon Genetics, Agilent Technologies).
