Project description:Xbp1 is a major transcription factor in the unfolded protein response. To uncover its function in DCs we generated a conditional KO for Xbp1 in dendritic cells. We here compare the expression of mRNAs in two different splenic DC subpopulations, CD8a and CD11b DCs in both WT and KO mice. Reference: Inositol-requiring enzyme 1-alpha regulates CD8a dendritic cell function via regulated mRNA decay. Project description:Dendritic cells (DCs) are a complex group of cells which play a critical role in vertebrate immunity. Spleen or lymph node resident DCs are subdivided into conventional DC (cDC) subsets (CD11b and CD8alpha in mouse; BDCA1 and BDCA3 in man) and plasmacytoid DCs (pDCs). It is currently unclear if these various DC populations belong to a unique hematopoietic lineage and if the subsets identified in the mouse and human systems are evolutionary homologs. To bring novel insights into these questions, we sought conserved genetic signatures for these DCs through the analysis of a compendium of genome-wide expression profiles of mouse or human leukocytes. We show through clustering analysis that all spleen resident DC subsets form a distinct branch within the leukocyte family tree, and reveal a transcriptomal signature evolutionary conserved in all these DC subsets. Moreover we identify a large gene expression program shared between mouse and human plasmacytoid DCs, and smaller conserved profiles shared between mouse and human cDC subsets. Finally, we use compendium analysis to re-evaluate the classification of interferon-producing killer DCs (IKDCs) and lin-CD16+HLA-DR+ cells, which have both been claimed to be DCs, and show that these cells are more closely linked to NK or myeloid cells, respectively. Our study thus provides a unique resource for future investigation of the evolutionarily conserved molecular pathways governing the ontogeny and functions of leukocyte subsets, especially DCs. Project description:Innate sensing of viruses by dendritic cells (DCs) is critical for the initiation of anti-viral adaptive immune responses. Virus, however, have evolved to suppress immune activation in infected cells. We now analyze the susceptibility of different populations of dendritic cells to viral infections. We find that circulating human CD1c+ DCs support infection by HIV and influenza virus. Viral infection of CD1c+ DCs is essential for virus-specific CD8+ T cell activation and cytosolic sensing of the virus. In contrast, circulating human CD141+ DCs and pDCs constitutively limit viral fusion. The small GTPase RAB15 mediates this differential viral resistance in DC subsets through selective expression in CD141+ DCs and pDCs. Therefore, dendritic cell sub-populations evolved constitutive resistance mechanisms to mitigate viral infection during induction of antiviral immune response.

Project description:Diet plays an important role in lifestyle disorders and our study from Methionine Choline deficient diet induced non-alcoholic fatty liver disease led to specific decrease in pDC fraction from murine spleens. Delineating the role for individual components identified correlation of L-methionine supplementation with representation of pDC subtype. Considering implication of L-methionine in maintaining methylation status, we investigated the DNA methylation profiles among DC subtypes. Whole genome bisulfite sequencing analysis identified that pDCs display distinctly methylated regions in comparison to cDC subsets whereas comparable methylation in cDC1 and cDC2 served as a control. Further analysis of the pathways associated with DMRs, we reported that inhibition of MAPK pathway guides DC development towards pDC subtype. We demonstrate that supplementation with S-adenosylmethionine but not Homocysteine reversed the defect in pDC development induced by methionine deficiency. Overall, our study identifies important role for methionine in pDC biology.

Our results demonstrate that pDCs and cDC2s induce a remarkably similar transcriptomic profile in CD8_ T cells, which. is tailored for increased effector function, survival and sensitivity. towards secondary signals. Nonetheless, differences between pDC- and cDC2-induced T cell gene expression signatures encompass genes with a role in proliferation, cytolytic capacity and differentiation. Combining both DC subsets results in a T cell. transcriptomic program that. is very. similar to. the program induced by. pDCs alone.
