Cell cultures in passage numbers 5, 7, 9, 13, 14, 21, and 28 of lot#4F1560 were subjected to the microarray analysis.

Transcription profiling of human mesenchymal stem cells (six batches)

Project description:Human mesenchymal stem cells (hMSCs), which are multipotent cells to differentiate into several cell types, are expected to be a useful tool for cellular therapy. In some clinical settings, hMSCs have immuno-suppressive effects for GVHD (Graft-versus-host disease) and are expanded in vitro before application. To find biomarkers that indicate the culture stage of hMSCs, we performed microarray analysis for hMSCs derived from bone marrow, using Affymetrix GeneChip Human Genome U133 Plus 2.0 (54,613 probe sets). Experiment Overall Design: We profiled the gene expression in 6 different batches of hMSCs. The cells were cultured and subjected to total RNA extraction. We performed microarray analysis using 6 batches provided by 6 different individuals in early stage (passage #4 or 5), middle stage (#7 or 9) or senescing stage (#22, 24, or 28).

A mesenchymal stromal cell gene signature for donor age

Project description:Human aging is associated with loss of function and regenerative capacity. Human bone marrow derived mesenchymal stromal cells (hMSCs) are involved in tissue regeneration, evidenced by their capacity to differentiate into several lineages and therefore are considered the gold standard for cell-based regeneration therapy. Tissue maintenance and regeneration is dependent on stem cells and declines with age and aging is thought to influence therapeutic efficacy, therefore, more insight in the process of aging of hMSCs is of high interest. We, therefore, hypothesized that hMSCs might reflect signs of aging. In order to find markers for donor age, early passage hMSCs were isolated from bone marrow of 61 donors, with ages varying from 17-84, and clinical parameters, in vitro characteristics and microarray analysis were assessed. Although clinical parameters and in vitro performance did not yield reliable markers for aging since large donor variations were present, genome-wide microarray analysis resulted in a considerable list of genes correlating with human age. By comparing the transcriptional profile of aging in human with the one from rat, we discovered follistatin as a common marker for aging in both species. The gene signature presented here could be a useful tool for drug testing to rejuvenate hMSCs or for the selection of more potent, hMSCs for cell-based therapy. Identity, proliferation capacity, genomic stability and novel senescence markers of mesenchymal stem cells isolated from low volume of human bone marrow (III)

Project description:Human bone marrow mesenchymal stem cells (hBM-MSCs) hold promise for treating diseases for which currently no therapeutic options exist. High quantities of MSCs are needed, requiring extensive cell expansion in long-term culture. However, the fundamental biological properties of MSCs can be altered by culture conditions. In this study, hBM-MSCs were isolated from residual human bone marrow (hBM) material and expanded to clinically relevant numbers at passage 3-4. The cells had normal morphology, proliferation rate, immunophenotype and karyotype. Despite the chromosomal stability, after additional three passages (P6-P7) hBM-MSCs entered senescence state, confirmed by SA-β-galactosidase staining and paralleling the slower proliferation, altered morphology and imunophenotype. qRT-PCR array profiling revealed 33 out of 420 miRNAs significantly (p <0.05) differentially (≥2 fold) expressed in the late passage cells compared to the early passage cells.

Expression data from rat lung-resident mesenchymal stem cells and bone marrow-derived mesenchymal stem cells

Project description:Rat lung-resident mesenchymal stem cells (LR-MSCs) were isolated via bronchoalveolar lavage from fibroblast growth factor-10 (FGF-10) pretreated lungs. We characterized the similarity and diversity between LR-MSCs and bone marrow-derived mesenchymal stem cells (BM-MSCs) by transcriptional profiling of these two types of cells. In this dataset, we include the expression data obtained from LR-MSCs and BM-MSCs cultured under similar conditions at passage 5. These data are used to obtain genes that are differentially expressed by these two types of cells. 6 Total samples were analyzed. Both LR-MSCs and BM-MSCs are in three replicates.
