Project description:We hypothesized that miRNAs in the bone maroow mesenchymal stem cells (BM-MSC)-derived exosomes contributed to the phenotype change of breast cancer cells through exosome transfer. We analyzed the miRNA expression signature in BM-MSC-derived exosomes. We compared the miRNA expression levels in exosomes between BM-MSCs and adult fibroblasts (as a control). In this study, miRNA expression including in bone-marrow mesenchymal cell (BM-MSC)-derived exosomes was examined, and compared with that of exosomes derived from adult fibroblast cells or the BM-MSC cells. In addition, miRNA expression of BM-MSC exosomes was also compared with that of breast cancer cells with or without cancer stem cell marker.

Project description:C2C12 mouse myoblasts were transduced with either PAX3-FOXO1 expression vector (P3F-C2C12), a system that is commonly used to study the more aggressive alveolar rhabdomyosarcoma subtype, or empty vector (Ctrl-C2C12). Exosomes were isolated from both cell lines by differential centrifugation, and exosomal markers were characterized by western blot. Then, the Affymetrix GeneChip miRNA 3.0 array was used to identify the miRNA content of the extracted exosomes where the differentially deregulated miRNA (either enriched or depleted) in P3F-C2C12 exosomes were determined relative to Ctrl-C2C12 exosomes. Results showed that PAX3-FOXO1 fusion gene alters the content of exosomes and that the enriched miR-486-5p is a downstream effector of PAX3-FOXO1 in mediating the oncogenic effects of exosome-mediated paracrine signaling in this setting.

Project description:Identify miRNAs enriched or overexpressed in melanoma-derived exosomes compared to melanoma cells. We analyzed the 2,578 human miRNAs located in the array according to two ways. The first one identified the differentially expressed miRNAs between melanoma-derived exosomes and their parent cells. In this way, we found 198 miRNAs up-regulated in melanoma cell lines compared to their exosomes and 206 miRNAs up-regulated in exosomes compared to their donor cells. The second way of analysis identifies the most expressed miRNAs in the melanoma-derived exosomes without assumption about their expression in parent cells. We defined two criteria; the first one is a RMA (Robust Multi-Array Average, a good alternative to gene expression value) above 5, corresponding to the mean expression level of the array and a SD ≤ 0.2 between all samples to discover miRNAs expressed uniformly between melanoma-derived exosomes M113 and M117. We identified 44 miRNAs under these criteria.

Project description:Myeloid-derived suppressor cells (MDSCs) are immature myeloid cells that accumulate in the tumor microenvironment of most cancer patients. There MDSCs suppress both adaptive and innate immune responses, hindering immunotherapies. Moreover, many cancers are accompanied by inflammation, a processes that further intensifies MDSC suppressive activity, causing aggressive tumor progression and metastasis. MDSCs collected from tumor-bearing mice profusely release nano-scale membrane-bound extracellular vesicles, called exosomes, which carry biologically active proteins between cells and contribute directly to the immune suppressive functions of MDSC. Many studies on other cell types have shown that exosomes may also carry microRNAs (miRNAs) and messenger RNAs (mRNAs) which can also be transferred to surrounding and distant cells. However, to the best of our knowledge, the miRNA and mRNA cargo of MDSC-derived exosomes has not yet been interrogated. This study aims to identify and quantify the cargo of MDSC and their immunosuppressive exosomes to gather knowledge that can offer insights on the mechanisms by which MDSCs contribute to immune suppression, focusing on the role of exosomes as intercellular communication mediators in the tumor microenvironment. In order to achieve our objective a well-established mouse model based on a conventional mammary carcinoma (4T1 cells) and heightened inflammation (4T1 transduced to express the cytokine interleukin-1b) was used. We provide evidence that MDSC-derived exosomes carry proteins, mRNAs and miRNAs. Relative quantitation demonstrated quantitative differences between the exosome cargo and the cargo of their parental cells, supporting the hypothesis that selective loading into the exosomes is possible. Additionally, quantitative and functional analyses of the exosome cargo generated under conventional and heightened inflammation conditions are consistent with clinical observations that inflammation is linked to cancer development.

Pre-miRNAs, along with Dicer, AGO2, and TRBP, are present in exosomes of cancer cells. CD43 mediates the accumulation of Dicer specifically in cancer exosomes. Cancer exosomes mediate an efficient and rapid silencing of mRNAs to reprogram the target cell transcriptome. Exosomes derived from cells and sera of patients with breast cancer instigate non-tumorigenic epithelial cells to form tumors in a Dicer-dependent manner. These findings offer opportunities for the development of exosomes based biomarkers and therapies. Exosomes from cancer cells and non-tumorigenic cells were isolated using established ultracentrifugation methods. The global miRNA content of cancer exosomes and normosomes was investigated. Profiling of cells themselves was also used as a control. Exosomes with Dicer down regulation (MCF10AshDicer and MDA-MB-231shDicer exosomes), as well as MDA-MB-231 exosomes that contain a Dicer antibody inside were used to study the function Dicer protein in the microRNA biogenesis in exosomes.

Project description:Human saphenous vein perciytes (hSVPs) were proved increase angiogenesis in mouse myocardial infarction model by producing and releasing miR-132, which has proangiogenic, prosurvival, and antifibrotic activity. To investigate whether their exosomes have a role in their proangiogenic activity, we examined exosomes secreted by these cells. Nanoparticle tracking analysis (NTA) results indicated the presence of exosome size particles in the preparations. Experiments performed with hypoxic (1% O2) human umbilical vein endothelial cells (HUVECs) confirmed that the pericyte exosomes had proangiogenic and anti-apoptotic features. To understand the miRNA cargo of these exosomes that is important in their unique properties, we performed a miRNA array for profiling of 752 miRNAs.

Project description:Kashin-Beck disease (KBD) is an endemic, chronic degenerative joint disease in China. Exosomes miRNAs, as signaling molecules in intercellular communication, can transfer specific biological martials into target cell to regulate the their function and might participate in the pathogenesis of KBD. We isolated serum and chondrocytes-derived exosomes, miRNA sequencing revealed exosomes miRNA profiles and differentially expressed miRNAs (DE-miRNAs) were identified. The target genes were predicted of known and novel DE-miRNAs with TargetScan 5.0 and miRanda 3.3a database. Single-cell RNA sequencing (scRNA-seq) was performed to identify chondrocyte clusters and their gene signatures in KBD. And we performed comparative analysis between the serum and chondrocytes-derived exosomes DE-miRNA target genes and differentially expressed genes of each cell clusters.A total of 20 DE-miRNAs were identified in serum-derived exosomes. In the miRNA expression of chondrocytes-derived exosomes, 53 DE-miRNAs were identified. 16063 predicted targets were identified as the target genes in the serum-derived exosomes, 57316 predicted targets were identified as the target genes in the chondrocytes-derived exosomes. Seven clusters were labelled by cell type according to the expression of previously described markers. 315 common genes were found among serum/chondrocytes-derived exosomes DE-miRNA target genes and DEGs identified by scRNA-seq analysis. We firstly integratly analyzed the serum and chondrocytes exosomes miRNA with single-cell RNA sequencing (scRNA-seq) data of KBD chondrocyte, the results showed that DE-miRNAs in exosomes might play a potential role in regulating genes expression in different KBD chondrocytes clusters by exosomes mediating cell-cell communications functions, which could improve the new diagnosis and treatment methods for KBD.
