Project description:Purpose: The goals of this study are to compare the effects of vehicle, PTH, and YKL-05-093 on gene expression by NGS-derived RNA-sequencing Methods: Ocy454 cells were grown at 37C for 14 days . Cells were then treated with vehicle, PTH (1 nM), or YKL-05-093 (0.5 uM) for 4 hours. Total RNA was isolated using Qiagen Rneasy columns, and submitted for NGS library generation and sequencing. All experiments performed in duplicate.

Rapid genomic changes induced by PTH, SIK-inhibition, FGF23, and 1,25(OH)2D3 in the mouse kidney

Project description:In vivo ChIP-seq in mouse kidney: we examine the genomic recruitment of VDR, RXR, pCREB, CBP, CRTC2 (TORC2), SRC1, SRC3, BRD4, RNApolII, H3K9ac, H3K27ac, and H3K36me3 after intraperitoneal injection of 10 mg/kg body weight (bw) 1,25(OH)2D3 (in propylene glycol), 230 mg/kg bw PTH (1–84) (in phosphate buffered saline (PBS)), 50 mg/kg bw FGF23 (in PBS + 0.1% BSA), 30 mg/kg YKL-05-099 (PBS + 25mM HCl), 40 mg/kg SK-124 (15% HPBCD (hydroxypropyl β-cyclodextrin)), or vehicle (EtOH, PBS, or HPBCD). YKL-05-099 and SK-124 are salt-inducible kinase inhibitors that liberate CRTC2 from sequestration in the cytoplasm and allow CRTC2 translocation to the nucleus. Our findings show that these TFs and coactivators contribute to transcription of genes in the kidney involved in vitamin D metabolism, demonstrates SIK-inhibition as a key modulator of vitamin D metabolism, and provides molecular insight into the coordinated mechanistic actions of PTH, FGF23, and 1,25(OH)2D3 in the mouse kidney that regulate mineral homeostasis.

Stimulation of fracture mineralization by salt-inducible kinase inhibitors

Project description:Over 6.8 million fractures are reported annually in the US, accounting for 20% of all musculoskeletal injuries. Approximately 10% of fractures will experience delayed- or non-union. Parathyroid hormone (PTH) analogs and anabolic osteoporosis therapeutics can stimulate fracture repair in animal models and patients with chronic nonunion. Previous studies have demonstrated that small molecule salt inducible kinase (SIK) inhibitors mimic PTH action in vitro and in vivo. Therefore, we hypothesize that YKL-05-099, a small molecule SIK inhibitor, will accelerate fracture callus osteogenesis.

Expression data from placenta harvested from WT and Pth-null fetuses treated 90 minutes prior with saline or PTH (1-84)

Project description:Parathyroid hormone (PTH) plays an essential role in regulating calcium and bone homeostasis in the adult, but whether PTH is required at all for regulating fetal-placental mineral homeostasis is uncertain. To address this we treated Pth-null mice in utero with 1 nmol PTH (1-84) or saline and examined placental calcium transfer 90 minutes later. It was found that placental calcium transfer increased in Pth-null fetuses treated with PTH as compared to Pth-null fetuses treated with saline. Subsequently, to determine the effect of PTH treatment on placental gene expression, in a separate experiment, 90 minutes after the fetal injections the placentas were removed for subsequent RNA extraction and microarray analysis.

PTH and PTHrP treatment of primary adipocytes

Project description:Parathyroid hormone (PTH) and PTH-related protein (PTHrP) are involved in cachexia associated with chronic kidney disease and cancer respectively. Tumor-derived PTHrP triggers adipose tissue browning and thereby leads to wasting of fat tissue in tumor-bearing mice. Similarly, elevated in 5/6 nephrectomized mice, PTH stimulates adipose tissue browning and wasting. Mice lacking the PTH/PTHrP receptor in their fat tissue are resistant to wasting of both adipose tissue and skeletal muscle. Therefore, the PTH/PTHrP signaling in adipocytes should activate various pathways that contribute to hypermetabolism and muscle wasting.

Kindlin-2 modulates PTH1R to regulate bone mass and PTH actions in bone

Project description:To investigation the role of PTH and Kindlin-2 in bone development, we performed single-cell RNA-sequencing. From Con-veh, Con-PTH, cKO-veh, cKO-PTH, we profiled more than 20k single cells, including multi-potent mesenchymal stromal cells (MSC), osteoprogenitors, osteoblasts, chondrocytes, fibroblasts, endothelial cells, smooth muscle cells, skeletal muscle cells, pericytes, and schwann cells. We found proportion of part of these cells were significant altered by PTH or Kindlin-2 loss, especially for MSC, osteoblast, chondrocyte, and fibroblast. Transcriptomic analysis revealed gene expression was dramatically regulated by PTH or Kindlin-2 loss.
