Project description:Several mechanisms have been proposed to account for Sdh-mutation-induced tumorigenesis, the most accepted of which is based on the constitutive expression of the hypoxia-inducible factor 1alpha (Hif1alpha) at normal oxygen tension, a theory referred to as pseudo-hypoxic drive. Other molecular processes, such as oxidative stress, apoptosis or chromatin remodeling have been also proposed to play a causative role. Nevertheless, the actual contribution of each of these mechanisms has not been definitively established. Moreover, the biological factors that determine the tissue-specificity of these tumors have not been identified. In this work, we made use of the inducible SDHD-ESR mouse, a conditional mutant in the SdhD gene, which encodes the small subunit of MCII, and that acts as a tumor suppressor gene in humans. Project description:The aim of this study was to identify possible downstream target genes and investigate the underlying mechanisms of action of Bcl2l10 in ovarian cancer cells. We performed RNA sequencing (RNA-Seq) and obtained a list of differentially expressed genes (DEGs) in Bcl2l10-suppressed SKOV3 and A2780 cells. RNA-Seq data showed that DEGs after Bcl2l10 knockdown are involved in transcriptional regulation and energy metabolism in ovarian cancer cells. The RNA-Seq data were validated by quantitative real-time PCR (qRT-PCR) and western blot analysis, and the levels of metabolites after Bcl2l10 knockdown were measured by ELISA. KEGG enrichment analysis showed that the commonly downregulated genes in SKOV3 and A2780 cells after Bcl2l10 knockdown were significantly enriched in metabolic pathways. The analysis of the DEGs identified from the RNA-Seq and validated by qRT-PCR revealed that succinate dehydrogenase complex subunit D (SDHD) and isocitrate dehydrogenase 1 (IDH1), which are key enzymes of the TCA cycle that regulate oncometabolite production, are potential downstream targets of Bcl2l10. We further found that Bcl2l10 knockdown induced the accumulation of succinate and isocitrate through the downregulation of SDHD and IDH1.

Strikingly, the growth-promoting function of Cdkn1c is highly dosage sensitive but not subject to genomic imprinting. Collectively, our results suggest that the Cdkn1c locus regulates cortical development through distinct cell-autonomous and non-cell-autonomous mechanisms. More generally, our study highlights the importance to probe the relative contributions of cell intrinsic gene function and tissue-wide mechanisms to the overall phenotype.

We were not able to clarify the identity of PGL2 on 11q13. The lack of differential gene-expression of chromosome 11 genes might indicate that chromosome 11 loss, as demonstrated in SDHD-linked paragangliomas, is an important feature in the formation of a paraganglioma regardless of the genetic background.
