Project description:Powdery mildew (PM) is one of the most important crop diseases, causing severe economic losses to cucumber production worldwide. However, there are few reports about the proteomic response to PM infection in resistant cucumber. To understand the molecular mechanisms, the iTRAQ-based quantitative proteomics approach was employed to map the proteomes of resistant powdery mildew of the D8 (PM susceptible) and SSL508-28 (PM resistant segment substitution line) under PM inoculated (plants harvested 48 hours after inoculation) and PM non-inoculated (control) conditions.

Project description:Grape powdery mildew (PM), caused by the biotrophic ascomycete Erysiphe necator, is a devastating fungal disease that affects most Vitis vinifera cultivars. We have previously identified a panel of V. vinifera accessions from Central Asia with partial resistance to PM that possess a Ren1-like local haplotype. In this study we show that in addition to the typical Ren1-associated late post-penetration resistance, these accessions display a range of different levels of disease development suggesting that alternative alleles or additional genes contribute to determining the outcome of the interaction with the pathogen. To identify potential Ren1-dependent transcriptional responses and functions associated with the different levels of resistance, we sequenced and analyzed the transcriptomes of these Central Asian accessions at two time-points of PM infection. Transcriptomes were compared to identify constitutive differences and PM-inducible responses that may underlie their disease resistant phenotype. Responses to E. necator in all resistant accessions were characterized by an early up-regulation of 13 genes, most encoding putative defense functions, and a late down-regulation of 32 genes, enriched in transcriptional regulators and protein kinases. Potential Ren1-dependent responses included a hotspot of co-regulated genes on chromosome 18. We also identified 81 genes whose expression levels and dynamics correlated with the phenotypic differences between the most resistant accessions ?Karadzhandahal?, DVIT3351.27, and O34-16 and the other genotypes. This study provides a first exploration of the functions associated with varying levels of partial resistance to PM in V. vinifera accessions that can be exploited as sources of genetic resistance in grape breeding programs. Eight varieties (7 resistant to powdery mildew, 2 of which are V. vinifera spp. sylvestris, 5 of which are V. vinifera spp. sativa) are infected with powdery mildew (E. necator) at two timepoints (1 dpi and 5 dpi) with three replicates for each timepoint for each condition (uninfected vs. infected), for a total of 96 samples

Project description:The edr1 mutant of Arabidopsis thaliana displays enhanced resistance to the powdery mildew Golovinomyces cichoracearum, resulting in cell death and an absence of visible disease symptoms. To better characterize and understand the defense response of edr1, a time course of early signaling responses was performed after inoculation with powdery mildew and compared to the responses of wild-type Col-0. These time points represent early stages in the infection process, before any signs of susceptibility or resistance are visible.

Project description:Powdery mildew, caused by the fungus Blumeria graminis (DC) Speer, is one of the most important foliar diseases of cereals worldwide. It is an obligate biotrophic parasite, colonising leaf epidermal cells to obtain nutrients from the plant cells without killing them. Syringolin A (sylA), a circular peptide secreted by the phytopathogenic bacterium Pseudomonas syringae pv. syringae, triggers a hypersensitive cell death reaction (HR) at infection sites when sprayed onto powdery mildew infected wheat which essentially eradicates the fungus. The rational was to identify genes whose expression was specifically regulated during HR, i.e. genes that might be involved in the switch of compatibility to incompatibility.<br>Powdery mildew-infected or uninfected plants were treated with syringolin two days after infection and plant material for RNA extraction was collected at 0.5, 1, 2, 4, 8, 12 hours after treatment (hat), resulting in an early (2 and 4 hat) and late pool (8 and 12 hat). Plant material that was uninfected prior to syringolin treatment was collected 8 and 12 hat (late pool of uninfected plant material), and 1 hat, respectively.

Project description:To identify the nonhost resistance genes, a time-series based global expression profiling was performed in wheat using Agilent gene expression 44K microarray array. Seven-day-old wheat plants of cv. Renan was inoculated with spore density of 50-80 conidia mm-2 of adapted (Bgt) or non-adapted (Bgh) powdery mildew pathogen, and the abaxial epidermis of inoculated primary leaves or from non-inoculated control leaves was peeled at 6, 12, 24 and 74 h after inoculation. Total RNA was extracted by using the RNeasy plant mini kit with on-Column DNase digestion (Qiagen, Hilden, Germany), and hybridized to a 44K Agilent oligonucleotide custom array of wheat.

Project description:Powdery mildew caused by Erysiphe cruciferarum, is an epidemic of oil rapeseed (Brassica napus) growing worldwide, but resistant germplasm is rare in this species. We obtained the hybrid seeds of distant hybridization between powdery-mildew-immune Brassica carinata cultivar ‘White flower’ and susceptible B. napus cultivar ‘Zhongshuang11’. Five lines in the BC1F3 generation (F3 after backcross to 'Zhongshuang11') were identified to be resistant or moderately resistant. In order to identify the important biological responses to powdery mildew, the foliar transcriptomes of the resistant and susceptible plants in these progenies after powdery mildew inoculation were compared by using Illumina RNA-seq. We identified 10,454 differential expression genes (DEGs) and 1050 genes out of them are related to disease resistance. There were 271 DEGs in Group Resistance expressed at least two fold higher than in group S, while 779 DGEs expressed two fold lower. The genes highly expressed in Group Resistance are those encoding the proteins: (1) related to wax, chloroplast and cell wall metabolism, such as KCS6, CSP41B, RWA, callose synthetase 3, pectinase 9, fructosidase 2, 9s-lipoxygenase LOX2, etc.; (2) kinases including RKL, ERECTA, BAK1, BAM2, LysM receptor like kinase, and lipid transfer protein kinase ERl1 and ERl2; (3) broad spectrum powdery mildew resistance proteins RPW8, calmodulin MLO2, PMR5, MLP328, EDR2, RPS4 and RPS6, etc. In group susceptible, pectinesterase, cytochrome CYP81f2, LOX1, cysteine rich receptor protein kinases and serine / threonine protein kinases such as MEKK, RLK6, CRK45, APK1, BRl3, WAK1, WAK10, etc., and TIR-NB-LRR receptor like proteins R1M1, DSC1, DSC2 and pathogenesis-related protein PR-1 etc. were the most activated genes. The results provide the preliminarily knowledge about molecular mechanism in rapeseed defense response to powdery mildew.

Project description:Obligate biotrophs such as the virulent powdery mildew Golovinomyces orontii alter plant host cellular architecture, metabolism, and defense in order to acquire nutrients while suppressing cell death and senescence. G. orontii exclusively infects epidermal cells of Arabidopsis with clearly defined stages of infection. Host factors mediating the powdery mildew (PM) interaction are often expressed in the mesophyll cells underlying the infected epidermal cells. Therefore, in order to identify Arabidopsis processes and regulators mediating this PM interaction, we used UV laser microdissection to isolate cells at the PM infection site for global expression profiling. As part of this process, we optimized and/or developed novel tissue preparation, RNA extraction and amplification, and quality control protocols resulting in highly correlated biological replicate data. We focused on the growth and reproduction stage of the PM infection (5 days post infection) when the number of reproductive structures, conidiophores, can be quantified. Site-specific profiling increased our sensitivity dramatically, allowing us to identify specific processes, process components, and their putative regulators hidden in previous whole leaf global expression analyses. For example, the known cell cycle regulator MYB3R4 exhibits altered expression at the site of infection, as do a subset of cell-cycle-associated genes. Furthermore, null myb3r4 mutants exhibit enhanced resistance to PM with reduced conidiophores per colony, suggesting cell cycle control plays an important role in the PM interaction. Experiment Overall Design: Arabidopsis whole leaves from wild type Columbia-0 and enhanced disease susceptibility mutant eds16-1, a null isochorismate synthase 1 (At1g74710) mutant were harvested at 5 days after Golovinomyces orontii infection, microwave-fixed, paraffin-embedded and sectioned. Groups of epidermal and mesophyll cells (~20 cells/group) surrounding the G. orontii infected epidermal cell were cut using a Leica AS laser microdissection (LMD) system. In parallel, groups of epidermal and mesophyll cells were collected from uninfected leaves at 5 days from wild type Arabidopsis. LMD-isolated cells, whole leaf, whole leaf amplified and tissue scrape samples were used for RNA extraction and hybridization to Affymetrix Arabidopsis ATH1 microarrays. Gene expression profiles were obtained for wild type from all samples and for ics1 mutant from LMD infected samples. The experiment includes 2 biological replicates.
