Mating and sperm-free seminal fluid changes the gene expression in the utero-vaginal junction of White Leghorn hens

ABSTRACT: Spermatozoa being foreign to the female are to be promptly eliminated after deposition by the local immune defense of female. However, avoiding the local immune defense sperm can be stored in the utero-vaginal junction for weeks and keep their fertilization ability. Whether, UVJ changes their gene expression after mating or SF-infusion to tolerate thjose forign cells are not understood. Therefore this study was done to observe the gene expression in the utero-vaginal junction after mating or SF-infusion.

Project description:Sperms being foreign to the female are to be promptly eliminated by the female local immune defense. However, avoiding the local immune defense sperm can be stored for lenthy period in the oviductal sperm reservoir. It is currently unknown whether oviductal sperm reservoirs changes their gene expression to tolerate the spermatozoa after mating or sperm free seminal plasma infusion. Therefore this was tested using Swedish Landrace pigs in this study using cDNA microarray. We used 12 sows seperated into three groups- either oestrus sows were inseminated with 50 ml BTS (control, n=4) or mated with boars (treatment 1, n=4) or inseminated with sperm-free seminal plasma (treatment 2, n=4). The utero-tubal junction was retrieved within 24 h of treatment by operation.

Keel bone fractures are more prevalent in White Leghorn hens than in Red Jungle fowl hens-A pilot study.

Project description:Fractures and deviations to the keel bone are common in commercial laying hens, with reported variations in occurrence across strains and breeds. The aetiology is not fully understood, however, modern genetics and selection for efficient egg production has been claimed to be important factors for the keel bone fractures. To explore this further, we investigated keel bones from two different breeds, representing different degrees of selection for egg production: Red jungle fowl (n = 82), and White Leghorn (n = 32), where the latter is a selected laying breed which is the origin for many modern laying hen hybrids. Keel bones from a total of 116 birds, 53 hens and 63 roosters, were examined by necropsy at 80 weeks of age. All birds were raised in modified aviaries in the same holding facility. Overall, 24.5% of the hens had one or more fractures to the keel, with a difference in the prevalence between hens from the two breeds (p<0.01): 10% (95% CI: 3.7-24%) in the Red Jungle fowl hens and 69% (95% CI: 37-90%) in the White Leghorn hens. No roosters, regardless of breed, had keel bone fractures. Mild to moderate keel bone deviations were present in 54% (95% CI: 25-80%) of the hens and 4.7% (95% CI: 0.5-30%) of the roosters, all White Leghorns.

Transcriptomic diversification of granulosa cells during follicular development between White Leghorn and Silky Fowl hens.

Project description:Egg production rate in chicken is related to the continuity of follicle development. In this study, we found that the numbers of white prehierarchical, dominant, and yellow preovulatory follicles in the high-yielding layer breed, White Leghorn (WL), were significantly higher than those in the low egg-yielding variety, Silky Fowl (SF). The proliferation and differentiation of granulosa cells (GCs) play an important role in follicle maturation. Histological observation revealed a large number of melanocytes in the outer granulosa layer of follicles in SF but not in WL. Finally, RNA-sequencing was used to analyze the gene expression profiles and pathways of the GC layer in the follicles in both WL and SF hens. Transcriptome analysis of prehierarchical GCs (phGCs) and preovulatory GCs (poGCs) between WL and SF showed that steroid hormone-, oxytocin synthesis-, tight junction-, and endocytosis-related genes were expressed at higher levels in WL phGCs than in SF phGCs, whereas the insulin signaling pathway- and vascular smooth muscle contraction-related genes were upregulated in SF phGCs. Fatty acid synthesis, calcium signaling, and Wnt signaling pathway-related genes were expressed at higher levels in WL poGCs than in SF poGCs; however, adrenergic signaling, cGMP-PKG, and melanogenesis-related genes were upregulated in SF poGCs. These results indicate that genes that promote GC proliferation and secretion of various sex hormones are more active in WL than in SF hens. The upregulated signaling pathways in SF help in providing energy to GCs and for angiogenesis and melanogenesis. In vitro experiments confirmed that both the proliferation of poGCs and synthesis of reproductive hormones were higher in WL than in SF hens.

Cuticle deposition duration in the uterus is correlated with eggshell cuticle quality in White Leghorn laying hens.

Project description:The cuticle formed in the uterus is the outermost layer as the first defense line of eggshell against microbial invasions in most avian species, and analyzing its genetic regulation and influencing factors are of great importance to egg biosecurity in poultry production worldwide. The current study compared the uterine transcriptome and proteome of laying hens producing eggs with good and poor cuticle quality (GC and PC, the top and tail of the cuticle quality distribution), and identified several genes involved with eggshell cuticle quality (ESCQ). Overall, transcriptomic analysis identified 53 differentially expressed genes (DEGs) between PC versus GC group hens, among which 25 were up-regulated and 28 were down-regulated. No differences were found in the uterine proteome. Several DEGs, including PTGDS, PLCG2, ADM and PRLR related to uterine functions and reproductive hormones, were validated by qPCR analysis. Egg quality measurements between GC and PC hens showed GC hens had longer laying interval between two consecutive ovipositions (25.64 ± 1.23 vs 24.94 ± 1.12 h) and thicker eggshell thickness (352.01 ± 23.04 vs 316.20 ± 30.58 μm) (P < 0.05). Apart from eggshell traits, other egg quality traits didn't differ. The result demonstrated eggshell and cuticle deposition duration in the uterus is one of the major factors affecting ESCQ in laying hens. PTGDS, PLCG2, ADM and PRLR genes were discovered and might play crucial roles in cuticle deposition by regulating the uterine muscular activities and secretion function. The findings in the present study provide new insights into the genetic regulation of cuticle deposition in laying hens and establish a foundation for further investigations.

The Effect of Cooled Perches on Immunological Parameters of Caged White Leghorn Hens during the Hot Summer Months.

Project description:The objective of this study was to determine if thermally cooled perches improve hen immunity during hot summer. White Leghorn pullets at 16 week of age were randomly assigned to 18 cages of 3 banks at 9 hens per cage. Each bank was assigned to 1 of the 3 treatments up to 32 week of age: 1) thermally cooled perches, 2) perches with ambient air, and 3) cages without perches. Hens were exposed to natural ambient temperatures from June through September 2013 in Indiana with a 4 h acute heat episode at 27.6 week of age. The packed cell volume, heterophil to lymphocyte (H/L) ratio, plasma concentrations of total IgG, and cytokines of interleukin-1β and interleukin-6, plus lipopolysaccharide-induced tumor necrosis factor-α factor were measured at both 27.6 and 32 week of age. The mRNA expressions of these cytokines, toll-like receptor-4, and inducible nitric oxide synthase were also examined in the spleen of 32 week-old hens. Except for H/L ratio, thermally cooled perches did not significantly improve currently measured immunological indicators. These results indicated that the ambient temperature of 2013 summer in Indiana (24°C, 17.1 to 33.1°C) was not high enough and the 4 h heat episode at 33.3°C (32 to 34.6°C) was insufficient in length to evoke severe heat stress in hens. However, cooled perch hens had a lower H/L ratio than both air perch hens and control hens at 27.6 week of age and it was still lower compared to control hens (P < 0.05, respectively) at 32 week of age. The lowered H/L ratio of cooled perch hens may suggest that they were able to cope with acute heat stress more effectively than control hens. Further studies are needed to evaluate the effectiveness of thermally cooled perches on hen health under higher ambient temperatures.

Effects of Maternal Care During Rearing in White Leghorn and Brown Nick Layer Hens on Cognition, Sociality and Fear.

Project description:Both genetic background and maternal care can have a strong influence on cognitive and emotional development. To investigate these effects and their possible interaction, White Leghorn (LH) and Brown Nick (BN) chicks, two hybrid lines of layer hen commonly used commercially, were housed either with or without a mother hen in their first five weeks of life. From three weeks of age, the chicks were tested in a series of experiments to deduce the effects of breed and maternal care on their fear response, foraging and social motivation, and cognitive abilities. The LH were found to explore more and showed more attempts to reinstate social contact than BN. The BN were less active in all tests and less motivated than LH by social contact or by foraging opportunity. No hybrid differences were found in cognitive performance in the holeboard task. In general, the presence of a mother hen had unexpectedly little effect on behavior in both LH and BN chicks. It is hypothesized that hens from commercially used genetic backgrounds may have been inadvertently selected to be less responsive to maternal care than ancestral or non-commercial breeds. The consistent and strong behavioral differences between genetic strains highlights the importance of breed-specific welfare management processes.
