FVB mouse strain lung cells

ABSTRACT: We sacrificed 6 (3 males, 3 females) FVB mice 8 weeks old and harvested their lungs. The dissected lungs lungs were assayed to RNA extraction (via trizol) in order to identify their transcriptomic signature.

Gene Expression analysis of the HIVAN1 Susceptibility Locus in the FVB-HIVAN1CAST congenic strain

Project description:Expression profiling comparing healthy kidneys of wild-type FVB mice and wild-type full congenic FVB-HIVAN1CAST mice HIV-1 transgenic mice on the FVB/NJ background (TgFVB) represent a validated model of HIV-associated nephropathy (HIVAN). A major susceptibility locus, HIVAN1, was previously mapped to chromosome 3A1-A3 in a cross between TgFVB and CAST/EiJ (CAST) strains, and introgression of a 51.9 Mb segment encompassing HIVAN1 from CAST into TgFVB results in accelerated development of nephropathy. We performed genome-wide expression profiling of whole kidneys from wild-type (without the HIV-1 transgene) full congenic FVB-HIVAN1CAST and FVB mouse strains, with the goal of identifying genes with differential renal expression in the HIVAN1 locus that may be associated with the development of nephropathy upon exposure to HIV-1. We only profiled healthy wild-type kidneys because the profound histopathological lesions of HIV-1 transgenic mice introduce many secondary gene expression changes that can confound interpretation of transcriptomic data.

Effects of agouti overexpression in the brain and skin transcriptome of zebrafish

Project description:Twenty transgenic fish overexpressing agouti-signaling protein (ASIP, Ceinos et al., Pigment Cell and Melanoma Research 28:196-209, 2015) and wild type (WT) zebrafish were placed in 6-liter tanks and acclimated during 30 days prior to sampling to minimize physiological differences. Animals were sacrificed and the brain+pituitary and the skin dissected under the stereo microscope, placed in trizol and stored at -80C until RNA extraction. Six biological replicates (3 females and 3 males) of each tissue (skin and brain) of each group (ASIP vs WT) were analyzed in a total of 6 slides containing 4 microarrays in the 4 × 44 k format. Global gene expression analyses were performed to search for both sex-dependent and transgene-dependent effects as well as the interaction between both factors (sex x phenotype) on the brain+pituitary and skin transcriptome.

Transcriptomic comparison of mouse lung adenocarcinoma cell lines derived upon treatment with urethane

Project description:We repeatedly injected FVB and C57BL/6 intraperitonealy with urethane. Mice developed lung tumours. We sacrificed the mice, harvested their lungs and cultured the tumour cells. After over 60 passages we established 3 novel mouse lung adenocarcinoma cell lines. The aim of this study os to compare their transcriptomic profile.
