Expression data of HepG2 cells treated with AP000770.1-1 siRNAs

ABSTRACT: In our study, we found the expression of the novel long coding RNA (AP000770.1-1) was positively associated with cholesterol levels in human samples and HepG2 cells. The Affymetrix GeneChip Human Gene 1.0 ST Array were used to analyze the expression profile of mRNAs in l AP000770.1-1 knockdown HepG2 cells and control by siRNA methods.

Project description:We here compared gene expression profiles in HepG2 cells upon stimulation with 1 ng/ml TGF-beta1 for 20 min, 1 hour, 2 hours, 4 hours, and 24 hours with untreated control cells. Experiments were done in three independent replicates. The goal of this study was to determine genes regulated by TGF-beta1.HepG2 cells were obtained from DSMZ (Braunschweig, Germany) and cell identity confirmed by STR profiling using the AmpFlSTR Identfiler Direct PCR Amplification kit (Life Technologies, Darmstadt, Germany). Gene expression profiles were compared at indicated time points after stimulation with TGF-beta (1 ng/ml) using the Human Gene 1.0 ST arrays (Affymetrix). In total 18 hybridizations are included in this series. 18 samples were hybridized to Affymetrix Human Gene 1.0 ST Array

Transcript Level Expression Profiling: anticancer pathways of Smilax Glabra rhizome extract (SW) [HepG2 cells]

Project description:To find the possible signaling pathways which contribute to the anticancer effect of SW-treated HepG2 cells Global gene expression profiling is an ideal technique to obtain useful clues for exploration of the anticancer mechanism of SW. Through comparing microarray results between solvent- and SW-treated cells, differentially expressed genes were obtained (>1.5 fold). The microarray results were validated using real-time RT-PCR. We used the KEGG database, STRING database and GO database for further ananlysis, and therefore got the possible signaling pathways underlying the anticancer effect of SW. We analyzed total RNA samples of solvent- and SW-treated HepG2 cells using the Affymetrix Human Gene 1.0 ST platform. No techinical replicates were performed.
