Project description:3,3’,5.5’-Tetrabromobisphenol A (TBBPA) is a widely used brominated flame-retardant utilized in the production of electronic devices and plastic paints. The objective of this study is to use zebrafish as a model and determine the effects of TBBPA exposure on early embryogenesis. We initiated TBBPA exposures (0, 10, 20 and 40μM) at 0.75 h post fertilization (hpf) and monitored early developmental events such as cleavage, blastula and epiboly that encompass maternal-to-zygotic transition (MZT) and zygotic genome activation (ZGA). Our data revealed that TBBPA exposures induced onset of developmental delays by 3 hpf (blastula). By 5.5 hpf (epiboly), TBBPA-exposed (10-20 μM) embryos showed concentration-dependent developmental lag by up to 3 stages or 100% mortality at 40 μM. Interestingly, while continued 0.75- 48 hpf TBBPA exposures (10 μM) led to severely deformed embryos, replacing exposure solution with chemical-free media at 6 hpf mitigated this effect, with 100% normal embryos at 48 hpf. To examine the genetic basis of TBBPA-induced delays, we conducted mRNA-sequencing on embryos exposed to 0 or 40 μM TBBPA from 0.75 hpf to 2, 3.5 or 4.5 hpf. Read count data showed that while TBBPA exposures had no overall impacts on maternal or maternal-zygotic genes, collective read counts for zygotically activated genes were lower in TBBPA treatment at 4.5 hpf compared to time-matched controls, suggesting that TBBPA delays ZGA. Gene ontology assessments for both time- and stage-matched differentially expressed genes revealed TBBPA-induced inhibition of chromatin assembly- a process regulated by histone modifications. Since acetylation is the primary histone modification system operant during early ZGA, we hypothesized that TBBPA inhibits histone acetylation, resulting in lack of open chromatin within promoters of zygotic genes and delaying ZGA. Therefore, we co-exposed embryos to 20 μM TBBPA and 100 μM N-(4-Chloro-3-(trifluoromethyl)phenyl)-2-ethoxybenzamide (CTB) -a histone acetyltransferase activator that promotes histone acetylation- and showed that TBBPA-CTB co-exposures from 0.75- 3 hpf significantly reversed TBBPA-only developmental delays, suggesting that TBBPA-induced phenotypes are indeed driven by repression of histone acetylation. Collectively, our work demonstrates that TBBPA disrupts ZGA and early developmental morphology, potentially by inhibiting histone acetylation. Future studies will focus on mechanisms of TBBPA-induced chromatin modifications.

Project description:We report a flame retardant epoxy nanocomposite reinforced with 9,10-dihydro-9-oxa-10-phosphaphenantrene-10-oxide (DOPO)-tethered SiO2 (DOPO-t-SiO2) hybrid nanoparticles (NPs). The DOPO-t-SiO2 NPs were successfully synthesized through surface treatment of SiO2 NPs with (3-glycidyloxypropyl)trimethoxysilane (GPTMS), followed by a click reaction between GPTMS on SiO2 and DOPO. The epoxy nanocomposites with DOPO-t-SiO2 NPs as multifunctional additive exhibited not only high flexural strength and fracture toughness but also excellent flame retardant properties and thermal stability, compared to those of pristine epoxy and epoxy nanocomposites with a single additive of SiO2 or DOPO, respectively. Our approach allows a facile, yet effective strategy to synthesize a functional hybrid additive for developing flame retardant nanocomposites.

Project description:We herein report a new vitrimer system integrated with UV curability, recyclability, and flame retardancy. Energy-efficiency, sustainability, and safety have been required features for next-generation polymer materials. Various attempts have been made to endow thermoset polymers with rapid prototyping capacity, recyclability, and flame retardancy. Thermoset vitrimers based on covalent adaptable networks (CANs) are recyclable and remoldable but are generally not UV curable or flame retardant. Here, we present a conceptually novel option to achieve fast exchange reactions in CANs via catalyst-free mixed transesterification of a UV curable phosphate diester-based acrylate cross-linker. In this system, the phosphate diesters serve as reversible covalent bonds, hydrogen bonding ligands, and flame-retardant structures, while acrylate groups serve as UV curable units as well as transesterification collaborators. After the facile UV curing, an intrinsic flame-retardant and mechanically strong dynamic network was achieved due to abundant hydrogen bonds between P-OH and C═O structures. Additionally, this highly cross-linked network exhibited an attractive recyclability even at temperatures lower than Tg. This phosphate diester-based mixed transesterification concept represents an efficient approach for developing multifunctional vitrimers and can also be generalized into other thermally cured polymer systems.

Project description:There have been many attempts to compile comprehensive lists of flame retardants. However, this goal has proven challenging due to the heterogeneity of compounds that can be used as flame retardants coupled with changes in formulation chemistry over time. Flame retardants have been the focus of many recent existing hazard, exposure, and risk assessments. These assessments have been class-based or for individual chemical substances. Here, diverse sets of publicly available data sources from governmental organizations and the open literature were compiled to develop an inventory of chemicals used as flame retardants and organohalogen flame retardants. The chemical substances from these data sources were mapped to appropriate chemical identifiers via manual curation and deduplicated. Despite different data sources containing a large number of overlapping chemical substances, compiling information from multiple data sources was found to increase the breadth of potential flame retardant chemistries. The flame retardant and organohalogen flame retardant inventories were developed as a resource for scientists interested in better understanding properties of flame retardant and organohalogen flame retardant classes. Project description:In this study, to develop an organic/inorganic synergistic flame retardant and to reduce the dosage and cost of flame retardants, organic/inorganic synergistic flame retardants, hexakis(4-boronic acid-phenoxy)-cyclophosphazene (CP-6B), and magnesium hydroxide (MH) were chosen. The flame retardant properties of CP-6B/MH in epoxy resin (EP) were discussed. EP/CP-6B/MH had better flame retardancy and heat resistance compared with EP/CP-6B and EP/MH. A limiting oxygen index of EP/3.0%CP-6B/0.5%MH of 31.9% was achieved, and vertical burning V-0 rating was achieved. Compared with EP, the cone calorimeter dates of EP/CP-6B/MH decreased. CP-6B/MH inhibited combustion and did little to damage mechanical properties. Besides, the flame retardant mechanism was studied by scanning electron microscopy, energy-dispersive X-ray spectroscopy, X-ray diffraction, Fourier transform infrared spectroscopy, and pyrolysis-gas chromatography-mass spectrometry. CP-6B/MH exerted good synergistic effects.

Project description:Calcium alginate (Ca-Alg) fibers are renewable fibers obtained from the ocean with essential flame retardancy, which have recently been applied as components of flame-retardant paper. However, the application of Ca-Alg fibers is limited because of their tendency to smolder. Therefore, composites papers were fabricated by blending using flame-retardant polyamide-66 (FR-PA), with a 5 wt% content of phosphorous flame retardant, which will form molten carbon during combustion. When the FR-PA content is 30% of the composite paper, FR-PA forms a compact carbon layer on the surface of the Ca-Alg fibers during combustion, which isolates the mass/heat transfer and effectively suppresses the smoldering of Ca-Alg. This consists of a condensed flame retardant mechanism. Furthermore, the combustion and thermal degradation behavior of paper were analyzed by cone calorimetry (CONE), TG and TG-IR. Ca-Alg in the composite paper decomposed and released CO2 before ignition, which delayed the ignition time. Simultaneously, the FR-PA contained in the composite paper effectively inhibited the combustion of volatile combustibles in the gas phase. Overall, FR-PA and Ca-Alg improve the thermal stability of the composite paper in different temperature regions under air atmosphere. Ca-Alg reduces the formation of aromatic products and NH3 in the composite paper under N2 atmosphere. Ca-Alg-based paper with excellent flame retardancy was successfully prepared.

Project description:In this study, a novel flame retardant (PMrG) was developed by self-assembling melamine and phytic acid (PA) onto rGO, and then applying it to the improvement of the flame resistance of PLA. PMrG simultaneously decreases the peak heat release rate (pHRR) and the total heat release (THR) of the composite during combustion, and enhances the LOI value and the time to ignition (TTI), thus significantly improving the flame retardancy of the composite. The flame retardant mechanism of the PMrG is also investigated. On one hand, the dehydration of PA and the decomposition of melamine in PMrG generate non-flammable volatiles, such as H2O and NH3, which dilute the oxygen concentration around the combustion front of the composite. On the other hand, the rGO, melamine, and PA components in PMrG create a synergistic effect in promoting the formation of a compact char layer during the combustion, which plays a barrier role and effectively suppresses the release of heat and smoke. In addition, the PMrGs in PLA exert a positive effect on the crystallization of the PLA matrix, thus playing the role of nucleation agent.

Project description:Tris (2-butoxyethyl) phosphate (TBOEP) is a compound produced at high volume that is used as both a flame retardant and a plasticizer. It is persistent and bioaccumulative, yet little is known of its toxicological modes of action. Such insight may aid risk assessment in a weight-of-evidence approach supplementing current testing strategies. We used an RNA sequencing approach as an unbiased and sensitive tool to explore potential negative health effects of sub-cytotoxic concentrations of TBOEP on the transcriptome of the human liver hepatocellular carcinoma cell line, HepG2, with the lowest concentration used potentially holding relevance to human physiological levels. Over-representation and gene set enrichment analysis corresponded well and revealed that TBOEP treatments resulted in an upregulation of genes involved in protein and energy metabolism, along with DNA replication. Such increases in cell and macromolecule metabolism could explain the increase in mitochondrial activity at lower TBOEP concentrations. In addition, TBOEP affected a wide variety of biological processes, the most notable one being the general stress response, wound healing. Finally, TBOEP showed effects on steroid hormone biosynthesis and activation, regulation, and potentiation of immune responses, in agreement with other studies. As such, this study is the first study investigating genome-wide changes in gene transcription in response to TBOEP in human cells.

Project description:BackgroundHuman exposures to organophosphate flame retardants result from their use as additives in numerous consumer products. These agents are replacements for brominated flame retardants but have not yet faced similar scrutiny for developmental neurotoxicity. We examined a representative organophosphate flame retardant, triphenyl phosphate (TPP) and its potential effects on behavioral development and dopaminergic function.MethodsFemale Sprague-Dawley rats were given low doses of TPP (16 or 32 mg kg-1 day-1 ) via subcutaneous osmotic minipumps, begun preconception and continued into the early postnatal period. Offspring were administered a battery of behavioral tests from adolescence into adulthood, and littermates were used to evaluate dopaminergic synaptic function.ResultsOffspring with TPP exposures showed increased latency to begin eating in the novelty-suppressed feeding test, impaired object recognition memory, impaired choice accuracy in the visual signal detection test, and sex-selective effects on locomotor activity in adolescence (males) but not adulthood. Male, but not female, offspring showed marked increases in dopamine utilization in the striatum, evidenced by an increase in the ratio of the primary dopamine metabolite (3,4-dihydroxyphenylacetic acid) relative to dopamine levels.ConclusionsThese results indicate that TPP has adverse effects that are similar in some respects to those of organophosphate pesticides, which were restricted because of their developmental neurotoxicity.

Project description:A novel intumescent flame retardant (IFR) agent designated as Dohor-6000A has been used to prepare halogen-free flame retardant polypropylene (PP) fibers via melting spinning. Before being blended with PP resin, a surface modification of Dohor-6000A was carried out to improve its compatibility with the PP matrix. The rheological behavior of flame retardant Dohor-6000A/PP resin, the structure, morphology, mechanical properties, flammability of the Dohor-6000A/PP fibers were studied in detail, as well as the action mode of flame retardant. X-ray diffraction (XRD) showed that the addition of Dohor-6000A did not damage the crystal as well as the orientation structure of PP matrix, which was helpful to the maintenance of mechanical properties. The presence of the IFR significantly improved the flame retardant performance and thermal stability of PP fibers. When the content of Dohor-6000A reached 25%, the fibers displayed a limiting oxygen index (LOI) value of 29.1% and good melt-drop resistance. Moreover, the peak heat release rate (PHRR) and total heat release (THR) from microscale combustion colorimetry (MCC) tests were decreased by 26.0% and 16.0% in comparison with the same conditions for pure PP fibers. In the condensed phase, the IFR promoted a carbonization process and promoted the formation of a glassy or stable foam protective layer on the surface of the polymer matrix. In addition, the IFR decomposed endothermically to release of non-combustible gases such as NH3 and CO2 which dilutes the combustible gases in the combustion zone.
